Engineering of an E. coli outer membrane protein FhuA with increased channel diameter
<p>Abstract</p> <p>Background</p> <p>Channel proteins like FhuA can be an alternative to artificial chemically synthesized nanopores. To reach such goals, channel proteins must be flexible enough to be modified in their geometry, <it>i.e</it>. length and diameter. As continuation of a previous study in which we addressed the lengthening of the channel, here we report the increasing of the channel diameter by genetic engineering.</p> <p>Results</p> <p>The FhuA Δ1-159 diameter increase has been obtained by doubling the amino acid sequence of the first two N-terminal β-strands, resulting in variant FhuA Δ1-159 Exp. The total number of β-strands increased from 22 to 24 and the channel surface area is expected to increase by ~16%. The secondary structure analysis by circular dichroism (CD) spectroscopy shows a high β-sheet content, suggesting the correct folding of FhuA Δ1-159 Exp. To further prove the FhuA Δ1-159 Exp channel functionality, kinetic measurement using the HRP-TMB assay (HRP = Horse Radish Peroxidase, TMB = 3,3',5,5'-tetramethylbenzidine) were conducted. The results indicated a 17% faster diffusion kinetic for FhuA Δ1-159 Exp as compared to FhuA Δ1-159, well correlated to the expected channel surface area increase of ~16%.</p> <p>Conclusion</p> <p>In this study using a simple "semi rational" approach the FhuA Δ1-159 diameter was enlarged. By combining the actual results with the previous ones on the FhuA Δ1-159 lengthening a new set of synthetic nanochannels with desired lengths and diameters can be produced, broadening the FhuA Δ1-159 applications. As large scale protein production is possible our approach can give a contribution to nanochannel industrial applications.</p

В результате работы выполнены исследования по влиянию физико-химических свойств нефти, начальной обводненности, интенсивности перемешивания при приготовлении водонефтяных эмульсий и влиянию деэмульгаторов на динамику и эффективность процесса отстаивания эмульсии при различных температурах.A number of experimental studies have been performed in the work on preparing and defending artificial water-oil emulsions using four types of demulsifiers. As a result of the work, studies were carried out on the effect of the physico-chemical properties of oil, the initial water cut, the intensity of mixing in the preparation of water-oil emulsions, and the effect of demulsifiers on the dynamics and efficiency of the emulsion settling process at various temperatures
Non-interfering effects of active post-encoding tasks on episodic memory consolidation in humans
So far, studies that investigated interference effects of post-learning processes on episodic memory consolidation in humans have used tasks involving only complex and meaningful information. Such tasks require reallocation of general or encoding-specific resources away from consolidation-relevant activities. The possibility that interference can be elicited using a task that heavily taxes our limited brain resources, but has low semantic and hippocampal related long-term memory processing demands, has never been tested. We address this question by investigating whether consolidation could persist in parallel with an active, encoding-irrelevant, minimally semantic task, regardless of its high resource demands for cognitive processing. We distinguish the impact of such a task on consolidation based on whether it engages resources that are: (1) general/executive, or (2) specific/overlapping with the encoding modality. Our experiments compared subsequent memory performance across two post-encoding consolidation periods: quiet wakeful rest and a cognitively demanding n-Back task. Across six different experiments (total N = 176), we carefully manipulated the design of the n-Back task to target general or specific resources engaged in the ongoing consolidation process. In contrast to previous studies that employed interference tasks involving conceptual stimuli and complex processing demands, we did not find any differences between n-Back and rest conditions on memory performance at delayed test, using both recall and recognition tests. Our results indicate that: (1) quiet, wakeful rest is not a necessary prerequisite for episodic memory consolidation; and (2) post-encoding cognitive engagement does not interfere with memory consolidation when task-performance has minimal semantic and hippocampally-based episodic memory processing demands. We discuss our findings with reference to resource and reactivation-led interference theorie
Advanced Gas Turbine Cooling for the Carbon-Neutral Era
In the coming carbon-neutral era, industrial gas turbines (GT) will continue to play an important role as energy conversion equipment with high thermal efficiency and as stabilizers of the electric power grid. Because of the transition to a clean fuel, such as hydrogen or ammonia, the main modifications will lie with the combustor. It can be expected that small and medium-sized gas turbines will burn fewer inferior fuels, and the scope of cogeneration activities they are used for will be expanded. Industrial gas turbine cycles including CCGT appropriate for the carbon-neutral era are surveyed from the viewpoint of thermodynamics. The use of clean fuels and carbon capture and storage (CCS) will inevitably increase the unit cost of power generation. Therefore, the first objective is to present thermodynamic cycles that fulfil these requirements, as well as their verification tests. One conclusion is that it is necessary to realize the oxy-fuel cycle as a method to utilize carbon-heavy fuels and biomass and not generate NOx from hydrogen combustion at high temperatures. The second objective of the authors is to show the required morphology of the cooling structures in airfoils, which enable industrial gas turbines with a higher efficiency. In order to achieve this, a survey of the historical development of the existing cooling methods is presented first. CastCool® and wafer and diffusion bonding blades are discussed as turbine cooling technologies applicable to future GTs. Based on these, new designs already under development are shown. Most of the impetus comes from the development of aviation airfoils, which can be more readily applied to industrial gas turbines because the operation will become more similar. Double-wall cooling (DWC) blades can be considered for these future industrial gas turbines. It will be possible in the near future to fabricate the DWC structures desired by turbine cooling designers using additive manufacturing (AM). Another conclusion is that additively manufactured DWC is the best cooling technique for these future gas turbines. However, at present, research in this field and the data generated are scattered, and it is not yet possible for heat transfer designers to fabricate cooling structures with the desired accuracy
