    Observations of lightning processes using VHF radio interferometry

    A single station, multiple baseline radio interferometer was used to locate the direction of VHF radiation from lightning discharges with microsec time resolution. Radiation source directions and electric field waveforms were analyzed for various types of breakdown events. These include initial breakdown and K type events of in-cloud activity, and the leaders of initial and subsequent strokes to ground and activity during and following return strokes. Radiation during the initial breakdown of a flash and in the early stages of initial leaders to ground is found to be similar. In both instances, the activity consists of localized bursts of radiation that are intense and slow moving. Motion within a given burst is unresolved by the interferometer. Radiation from in-cloud K type events is essentially the same as that from dart leaders; in both cases it is produced at the leading edge of a fast moving streamer that propagates along a well defined, often extensive path. K type events are sometimes terminated by fast field changes that are similar to the return stroke initiated by dart leaders; such K type events are the in-cloud analog of the dart leader return stroke process

This study explores how Agile practices are integrated into an online IT course. We focus on incorporating eight Agile practices (team charters, daily stand-ups, Kanban boards, story cards, MoSCoW, timeboxing, showcases, and retrospectives). Students were surveyed at the end of the course and the results indicate Agile enhanced students’ learning and increased the quality of their final project

    Site of infusion of a commercially available direct-fed microbial on performance and digestibility in lactating Holstein cows

    Objective: The objective of this research was to evaluate site of infusion of a commercially available direct-fed microbial (DFM) containing 109 cfu/g of Lactobacillus acidophilus and 109 cfu/g Propionibacterium freudenreichii on DMI, rumen kinetics, ruminal VFA, digestibility, milk production, milk components, and blood metabolites in lactating dairy cows. Materials and Methods: Four Holstein cows equipped with ruminal cannulas were used in a Latin square design experiment with 4 periods. Each 37-d period consisted of 14 d of no treatment to prevent crossover contamination, 14 d of adaptation to treatments, 8 d of sampling, and 1 d for ruminal evacuations. Within each period, cows were assigned to 1 of 4 treatments: (1) cows were fed a TMR formulated to meet or exceed nutrient requirements plus 5 g of lactose twice daily without the addition of DFM (control); (2) cows were fed the TMR with a daily dose of DFM top dressed on the feed twice daily (TD); (3) cows were fed the TMR with ruminal infusion of the DFM administered twice daily (RuI); or (4) cows were fed the TMR plus abomasal infusion of the DFM twice daily (AbI). During the sampling period within each period, DMI and milk production were measured daily with set days for blood and rumen fluid collection. Data were analyzed using the MIXED procedure of SAS with animal within period as a random effect. Dry matter intake was not different among treatments. Results and Discussion: No differences were detected in rumen kinetics, pH, individual VFA or VFA ratios, ammonia, or digestibility. There were no differences in kilograms of milk production (P \u3e 0.87); SCC (P \u3e 0.54); or percentage of butter fat (P \u3e 0.21), milk protein (P \u3e 0.83), lactose (P \u3e 0.91), SNF (P \u3e 0.88), and MUN (P\u3e 0.49). No difference existed in most of the milk fatty acids except for 8:0, which had a greater concentration (P \u3e 0.01) in AbI versus control, TD, and RuI. Implications and Applications: Route of administering DFM overall had no effects on DMI, rumen kinetics, ruminal VFA, digestibility, milk production, or milk components in the present experiment

    In vitro starch disappearance procedure modifications

    Four in vitro experiments evaluated the effects of ruminal fluid inoculum:artificial saliva ratios, grinder type, grind size, and diet of ruminal fluid donor on in vitro starch disappearance. Experiment 1 examined rates of starch disappearance and coefficients of determination obtained by linear regression of starch disappearance using five grain sorghum (Sorghum bicolur (L.) Moench) lines, a corn (Zeu muys L.) control, and a wheat (Triticum aestivum L.) control. Grains were incubated for 4, 8, 12, 16, and 20 h with inoculum varying in proportion of ruminal fluid and artificial saliva ( l:l, 1:2, 1:3, and 1:4). In vitro rates of starch disappearance and coefficients of determination were similar for the 1:2, 1:3, and 1:4 inoculum dilutions and were higher (quadratic, P \u3c 0.01) than the 1:1 dilution. As the proportion of artificial saliva in the inoculum increased, in vitro pH increased (linear, P \u3c 0.01). In Experiment 2, six grain sorghum lines and a corn control were ground through a l-mm screen in a Udy (cyclone type) or Wiley mill. Starch disappearance for samples ground using a Udy mill were higher (P 0.10) by grain type fed; however, absolute rates of digestion varied among inoculate sources. Grinder, grind size, and ruminal fluid inoculum:artificial saliva ratios affected rate of starch disappearance in samples digested in vitro. Diet of ruminal fluid donor affected the rate of starch digestion, but not the relative ranking of the grains. If rates are to be compared across in vitro runs for different grains, these processing and dietary factors must be kept constant

    Effect of stocker management program on beef cattle skeletal muscle growth characteristics, satellite cell activity, and paracrine signaling impact on preadipocyte differentiation

    The objective of this study was to determine the effect of different stocker management programs on skeletal muscle development and growth characteristics, satellite cell (SC) activity in growing-finishing beef cattle as well as the effects of SC-conditioned media on preadipocyte gene expression and differentiation. Fall-weaned Angus steers (n = 76; 258 ± 28 kg) were randomly assigned to 1 of 4 stocker production systems: 1) grazing dormant native range (NR) supplemented with a 40% CP cottonseed meal-based supplement (1.02 kg ∙ steer–1 ∙ d–1) followed by long-season summer grazing (CON, 0.46 kg/d); 2) grazing dormant NR supplemented with a ground corn and soybean meal-based supplement fed at 1% of BW followed by short-season summer grazing (CORN, 0.61 kg/d); 3) grazing winter wheat pasture (WP) at high stocking density (3.21 steers/ha) to achieve a moderate rate of gain (LGWP, 0.83 kg/d); and 4) grazing winter WP at low stocking density (0.99 steers/ha) to achieve a high rate of gain (HGWP, 1.29 kg/d). At the end of the stocker (intermediate harvest, IH) and finishing (final harvest, FH) phases, 4 steers / treatment were harvested and longissimus muscles (LM) sampled for cryohistological immunofluorescence analysis and SC culture assays. At IH, WP steers had greater LM fiber cross-sectional area than NR steers; however, at FH, the opposite was observed (p \u3c 0.0001). At IH, CORN steers had the lowest Myf-5+:Pax7+ SC density (p = 0.020), while LGWP steers had the most Pax7+ SC (p = 0.043). At FH, CON steers had the highest LM capillary density (p = 0.003) and their cultured SC differentiated more readily than all other treatments (p = 0.017). At FH, Pax7 mRNA was more abundant in 14 d-old SC cultures from HGWP cattle (p = 0.03). Preadipocytes exposed to culture media from proliferating SC cultures from WP cattle isolated at FH had more PPARγ (p = 0.037) and less FABP4 (p = 0.030) mRNA expression compared with NR cattle. These data suggest that different stocker management strategies can impact skeletal muscle growth, SC function, and potentially impact marbling development in growing-finishing beef cattle

    Water Intake in Growing Beef Cattle

    Water is an essential part of livestock and human diets and is often thought of as an inexpensive, readily available renewable natural resource. However, the amount of competition between humans, wildlife, feed production, and livestock for high-quality water is increasing, not only from the effects of drought but from the pressure of a growing global population (Nardone et al., 2010). With limited resources available for production agriculture, there is a need to identify and select for efficient animals that can produce more product with fewer inputs. Although some work has been done in dairy cattle, very little data is available on individual animal water intake in modern beef cattle (Brew et al., 2011). The majority of the water intake data available in growing beef cattle is derived from dividing the total amount of water drunk in a pen divided by the number of animals in that pen (Sexson et al., 2010; Mader and Davis, 2004). Data derived from groups are not generally useful for the purposes of genetic evaluation, which aims to quantify individual animal variation in a trait for selection. However, in order to practice selection on a large scale, parameters for collecting phenotypic data must be established. Journal of Animal Science, 94(1), 297-305. doi:10.2527/jas2015-9601The inclusion of wet distillers grains plus solubles (WDGS) in feedlot diets has become a common practice in many regions of the United States due to the expanded production of byproducts and fluctuating corn prices related to ethanol production and other factors. In addition, societal concerns over the continued use of antimicrobials in agriculture production combined with an enhanced interest in disease and pathogen prevention in the food supply have led to an increased interest in use of direct-fed microbials (DFM) in growing and finishing cattle. Direct-fed microbials have been shown to improve ADG and feed efficiency, alter ruminal fermentation, and decrease fecal shedding of potential harmful pathogens in feedlot cattle in some experiments. The objective of this experiment was to evaluate the effects of WDGS inclusion with or without a DFM containing Lactobacillus acidophilus (1 x 10(6) cfu . heifer(-1) . d(-1)) combined with Propionibacterium freudenreichii (1 x 10(9) cfu , heifer(-1) . d(-1)) on the performance, carcass characteristics, and Escherichia coli O157:H7 shedding in feedlot heifers. In early August, 288 crossbred heifers (initial BW = 295 +/- 28 kg) were assigned to 1 of 4 treatments (12 pens per treatment; 6 heifers per pen) in a randomized complete block design with a 2 x 2 factorial arrangement of treatments. Body weights and fecal grab samples were obtained at approximately 28-d intervals throughout the experiment. Across the feeding period, heifers fed 30% WDGS tended (P = 0.09) to have greater ADG and had greater carcass-adjusted ADG (P = 0.05) compared with heifers fed dry-rolled corn (DRC). Dry matter intake was not affected (P = 0.65) by diet, although carcass-adjusted G:F tended (P = 0.10) to be improved for heifers fed WDGS. Heifers fed 30% WDGS tended (P 0.10) on performance or carcass merit in the present experiment. The incidence of E. coli O157:H7 throughout the experiment was low, with only 18 positive samples across all sampling periods. Neither WDGS inclusion nor the inclusion of L. acidophilus combined with P. freudenreichii in the diet had any effect (P > 0.10) on E. coli O157:H7 shedding in this experiment.     Effects of dietary energy and starch concentrations for newly received feedlot calves: I. Growth performance and health

    Crossbred calves (n = 572; initial BW = 186 +/= 27 kg) purchased from northern Texas, Arkansas, and southeast Oklahoma auction markets were delivered to the Willard Sparks Beef Research Center, Stillwater, OK, and used to study the effects of dietary energy and starch concentrations on performance and health of newly received feedlot calves during a 42-d receiving period. On arrival, calves were assigned randomly to one of two dietary energy levels (0.85 or 1.07 Mcal NEg/kg DM) and one of two dietary starch levels (34 or 48% of ME from starch) in a 2 X 2 factorial arrangement of treatments. Cattle were weighed and serum samples were collected on d 0, 7, 14, 28, and 42. Individual animal records of morbidity were kept for all cases of respiratory and other disease. Nasal swabs were collected from each morbid animal and cultured for upper-respiratory pathogens. There were no energy x starch level interactions for performance or health response variables. Daily gain (1.14 kg/d) and gain efficiency (ADG:DMI = 0.179) were not affected by increasing dietary energy or starch concentrations. Calves fed low-energy diets consumed (P < 0.05) more DM. No difference (P = 0.54) was detected in morbidity for calves fed high-energy (62.4% calves treated) compared with low-energy (65.8% calves treated) diets; however, calves fed the high-starch diets had numerically (P = 0.11) greater morbidity than calves fed low-starch diets (68.8 vs. 59.4% calves treated, respectively). There were no energy or starch effects on Mannheimia haemolytica or Pasteurella multocida antibody titers; however, day effects (P < 0.02) occurred. On d 7, 14, and 28, calves had antibody titers for P. multocida that were greater (P < 0.05) than titers on d 0. In addition, calves had greater antibody titers to M. haemolytica on d 7 and 14 than on d 0. Nasal swabs revealed that calves fed the high-energy diets tended (P = 0.06) to have a lower percentage of morbid calves with P. multocida during the first antimicrobial treatment and a lower percentage of Haemophilus somnus isolates during the first (P = 0.01) and second (P = 0.06) antimicrobial treatments than calves fed the low-energy diets. Although animal performance was not influenced, the present data suggest that feeding the high-energy diet decreased the percentage of P. multocida and H. somnus pathogens in calves that received one or more antimicrobial treatments.Peer reviewedAnimal ScienceVeterinary PathobiologyVeterinary Medicine Outreac

    Effects of fat and/or methionine hydroxy analog added to a molasses-urea-based supplement on ruminal and postruminal digestion and duodenal flow of nutrients in beef steers consuming low-quality lovegrass hay

    Five crossbred beef steers (initial BW = 338.6 ± 7.8 kg) fitted with ruminal and duodenal cannulas were used in a 5 × 5 Latin square design experiment to evaluate the effects of methionine hydroxy analog (MHA) and/or yellow grease (fat) added to a molassesurea- based supplement on intake and characteristics of digestion. Steers were fed low-quality hay (long-stem lovegrass Eragrostis curvula: 3.3% CP, 76.8% NDF; DM basis) ad libitum and supplemented with 0.91 kg/d (as fed) of 1 of 4 supplements in a 2 × 2 + 1 factorial arrangement of treatments. Supplemental treatments were 1) control (no supplement, NC); 2) molassesurea liquid supplement (U); 3) U containing (as-fed basis) 1.65% MHA (UM); 4) U containing (as-fed basis) 12% fat (UF); and 5) U containing (as-fed basis) 1.65% MHA and 12% fat (UMF). Total and forage OM intake (kg/d and as % of BW) increased (P \u3c 0.01) with molasses-urea, decreased (P ≤ 0.04) with MHA, and were not affected (P = 0.61) with fat supplementation. Total tract NDF digestibility increased (P = 0.01) with molasses-urea supplementation, and was less (P = 0.01) for fat than for nonfat supplementation. Total and microbial N flowing to the duodenum increased (P = 0.01) with molasses-urea supplementation. Although, total N flowing to duodenum was not affected (P = 0.27), microbial N decreased (P = 0.01), and nonammonia nonmicrobial N (NANMN) increased (P = 0.01) with fat supplementation. Extent of in situ OM and NDF digestibility at 96 h increased (P = 0.01) with molasses-urea supplementation, but were not affected (P ≥ 0.14) by either MHA or fat supplementation. Duodenal flow of total AA, essential AA, and nonessential AA increased (P ≤ 0.02) with molasses-urea supplementation. Total and nonessential serum AA concentration decreased (P \u3c 0.01) with molasses-urea supplementation. Total ruminal VFA concentration increased (P = 0.01) with molasses-urea supplementation, and was not affected (P ≥ 0.14) by MHA or fat supplementation. Fat can be used in molasses-urea liquid supplements for cattle consuming low-quality forage to increase energy intake without negatively affecting forage intake or characteristics of digestion. However, adding MHA did not further improve the response to urea supplementation of cattle consuming low-quality forage. Conversely, the inclusion of MHA on urea supplement decreased forage intake

    Vitamin E supplementation of newly arrived feedlot calves

    Seven hundred fifteen crossbred (primarily British) calves purchased in southern Oklahoma and northern Texas auction barns were received at the Willard Sparks Beef Research Center, Stillwater, OK, and used to study effects of duration (days) of vitamin E feeding during a 42-d receiving period on animal performance, health, and serum cholesterol and vitamin E concentrations. Upon arrival, calves were blocked by load (seven loads), sorted by BW (light, n = 4 pens per load; and heavy, n = 4 pens per load), and assigned randomly to one of four dietary treatments (n = 2 pens per load; 14 pens per treatment). Experimental diets were formulated to provide 2,000 IU calf^-1 d^-1 of supplemental vitamin E (DL-alpha-tocopherol acetate) for 0 (CON), 7 (E7), 14 (E14), or 28 (E28) d. Vitamin E was delivered in a pelleted supplement that was added to the basal diet in decreasing concentrations as DMI increased (2.0 kg of DMI = 6%; 4.0 kg of DMI = 4%; and 6.0 kg of DMI = 2%). Serum samples were collected on d 0, 14, 28, and 42 for determination of cholesterol, alpha-tocopherol (d 0, 28, and 42), and antibody (IgG) concentrations. Duration of vitamin E supplementation did not affect ADG (0.98 kg/d; P = 0.56) or G:F (0.189; P = 0.87). Serum cholesterol concentrations decreased (day effect; P < 0.001) for all treatments from d 0 (average = 127 mg/100 mL) to 14 (average = 62 mg/100 mL). Serum alpha-tocopherol decreased (day effect; P < 0.001) from d 0 (5.2 micro-g/mL) to 28 (1.8 micro-g/mL); however, on d 28, a greater (P < 0.001) serum alpha-tocopherol concentration was observed for E28 (3.4 micro-g/mL) calves than for CON (1.1 micro-g/mL), E7 (1.2 micro-g/mL), or E14 (1.5 micro-g/mL) calves. Respiratory disease was diagnosed in 64.6% of calves in this study. Medical costs were less (P = 0.08) for calves fed vitamin E for 28 d (4.88/calf)thanforcalvesfedthecontroldiet(4.88/calf) than for calves fed the control diet (6.29/calf). Carcass characteristics were not affected (P = 0.19 to 0.88) by dietary treatments. Supplemental vitamin E formulated for 2,000 IU calf^-1 d^-1 had little influence on performance and overall health status of calves under our experimental conditions; however, the increased serum concentrations of alpha-tocopherol when vitamin E was fed for 28 d suggests that any potential effects of vitamin E on health status might be time-dependent.Peer reviewedAnimal ScienceVeterinary PathobiologyVeterinary Medicine OutreachStatistic
