A központi idegrendszer és a myocardium szabályozásában szerepet játszó adenozin-receptorok és PET izotóppal jelölt ligandjaik kötődés kinetikájának vizsgálata PET technikával = PET investigation of the binding kinetics of PET isotope-labelled ligands and adenosine receptors involved in the regulation of myocardial and CNS function
Az adenozin-receptorok alapvető szerepet játszanak a központi idegrendszer és a szívizom működésének szabályozásában. Célunk az in vivo PET vizsgálatokra alkalmas adenozin-receptor ligandok előállítása és biológiai tesztelése volt. Xantin típusú molekulák és a [11C]metiljodid reakciójával előállítottunk több A2A típusú specifikus antagonistát az adenozin-receptor expresszió és kötődés in vivo tanulmányozása céljából. Az A1, A2 és A3 adenozin-receptor expresszió in vivo tanulmányozásához pedig az 18F izotóppal jelölt 5?-N-etil-karboxamidadenonozint (NECA) állítottuk elő. A farmakológiai vizsgálatok bizonyították a jelölt ligandok specifikusságát. A ligandok szervezeten belüli eloszlását, specifikus kötődésüket a receptorokhoz, valamint az akkumulációs kinetikájukat in vitro, ex vivo és in vivo (dinamikus PET mérések) vizsgálatokban bizonyítottuk. Eredményeink alapján a szintetizált PET izotóppal jelölt ligandok alkalmasak az in vivo PET mérésekre. A kidolgozott radiokémiai és biológiai módszerek megteremtették a lehetőséget a PET izotóppal jelölt receptor regulációs kísérletek végzésére a Debreceni Egyetem PET Centrumában. | The adenosine receptors play crucial role in the regulation of the function of the myocardium and the central nervous system. The aim of this work was the development and biological test of adenosine receptor ligands which are suitable for in vivo PET examinations. By the chemical reaction of different xantine type molecules and [11C] methyl iodide we produced several A2A adenosine receptor specific antagonists for in vivo testing the receptor ligand interactions. To study the A1, A2 and A3 adenosine receptor expression 18F isotope labeled 5?-N-etil-carboxamidadenosine (NECA) was produced. The specificity of the produced ligands was proven by different pharmacological tests. The biological distribution of the ligands as well as their binding kinetics to the receptors was tested in vivo by dynamic PET scans and ex vivo or in vitro experiments. On the basis of our experiments we can conclude, that the developed ligands are highly specific and suitable for in vivo PET examinations. The protocols worked out provide a firm base for having been receptor/ligand research at the PET Center of the University of Debrecen
