    Managing treatment-experienced pediatric and adolescent HIV patients: role of darunavir

    Darunavir is currently the most recently approved HIV-1 protease inhibitor. It is approved for twice-daily dosing with ritonavir in treatment-experienced patients as young as 6 years of age and is available in numerous pill strengths. Emergence of darunavir-specific mutations is generally slow; therefore it can retain activity against viral strains that are resistant to other protease inhibitors, including tipranavir. Darunavir pharmacokinetics, clinical efficacy, resistance mutations and pharmacodynamics, and adverse effects are reviewed here. Substantial data support its use as a potent, well-tolerated option for salvage therapy in highly treatment-experienced children and adolescents

    On the incidence rate of first overtone Blazhko stars in the Large Magellanic Cloud

    By using the full span of multicolor data on a representative sample of first overtone RR Lyrae stars in the Large Magellanic Cloud (LMC) we revisit the problem of the incidence rate of the amplitude/phase-modulated (Blazhko) stars. Multicolor data, obtained by the MAssive Compact Halo Objects (MACHO) project, are utilized through a periodogram averaging method. This enables us to increase the number of detected multiperiodic variables by 18% relative to the number obtained by the analysis of the best single color data. We also test the maximum modulation period detectable in the present dataset. We find that variables showing amplitude/phase modulations with periods close to the total time span can still be clearly separated from the class of stars showing period changes. This larger limit on the modulation period, the more efficient data analysis and the longer time span lead to a substantial increase in the incidence rate of the Blazhko stars in comparison with earlier results. We find altogether 99 first overtone Blazhko stars in the full sample of 1332 stars, implying an incidence rate of 7.5%. Although this rate is nearly twice of the one derived earlier, it is still significantly lower than that of the fundamental mode stars in the LMC.     University Students' Democratic Values and Attitudes toward Democracy in Hungary

    In December 2012 Hungarian students rallied to protest against cutbacks in scholarships and to the introduction of tuition fees. Two years later, in October 2014, thousands of (mostly young) people gathered at anti-government demonstrations against a proposal to include the taxation of Internet usage in the Taxation Law. The article brings results on changes in patterns of democratic citizenship among Hungarian students and demonstrates that despite their issue related ‘rational rebellion’ democratic transition resulted in a low-level of political participation and commitment to democratic values among them. We describe democratic attitudes of Hungarian college and university students by using both qualitative and quantitative data asking if they have developed a commitment to democracy as a system of rule

    GRATITUDE, RELIGIOUSNESS AND WELL-BEING

    Background: Previous researches have identified the positive effects of gratitude on happiness and well-being. It has been found that spirituality can enhance gratitude and well-being. Our study aimed to examine the link between gratitude and subjective wellbeing among religious and non-religious people. Furthermore we tested if a 4-week long gratitude diary has a positive effect on subjective well-being. Subjects and methods: In our online, qualitative investigation the sample consisted of 54 males and 169 females (mean age = 39.13, SD=15.90). 54.1% of respondents rel24.8igious on my own waynd 21.2% as -experimental group (leading a gratitude diary for 4 weeks) with 103 individuals and a control group with 120 individuals with no differences in age, gender and religiousness. We compared the questionna (filled out both before and after the intervention) of the experimental group (n=103) with the results of the control group (n=120) similarly filled out on two different occasions. We applied Gratitude Resentment and Appreciation Test and Subjective Well-being Scale. Results: Our results showed that religious people showed elevated level of gratitude (F(2, 219)=23.66, p<0.001) but same wellbeing (F(2, 219)=1.97, p=0.142) compared to non-religious groups. In the experiment group the gratitude and the subjective wellbeing both increased (p<0.01), and there was no significant changes in control group. 2x2 ANOVA showed significant interaction effect (Subjective well-being: (F(1, 221)=13.32, p<0.001); Gratitude: F(1, 221)=12.43, p<0.001). Conclusion: Religiousness is linked to higher gratitude and an increase in gratitude can result in an increase in subjective wellbeing. The importance of gratitude diary both among religious and non-religious people will be discussed

    Occlusion of retinal capillaries caused by glial cell proliferation in chronic ocular inflammation

    The inner blood-retinal barrier is a gliovascular unit in which glial cells surround capillary endothelial cells and regulate retinal capillaries by paracrine interactions. During chronic ocular inflammation, microvascular complications can give rise to vascular proliferative lesions, which compromise visual acuity. This pathologic remodelling caused by proliferating MĂŒller cells determines occlusion of retinal capillaries. The aim of the present study was to identify qualitative and quantitative alterations in the retinal capillaries in patients with post-traumatic chronic ocular inflammation or post-thrombotic vascular glaucoma. Moreover, we investigated the potential role of vascular endothelial growth factor (VEGF) and pro-inflammatory cytokines in retinal inflammation. Our electron microscopy findings demonstrated that during chronic ocular inflammation, thickening of the basement membrane, loss of pericytes and endothelial cells and proliferation of MĂŒller cells occur with irreversible occlusion of retinal capillaries. Angiogenesis takes place as part of a regenerative reaction that results in fibrosis. We believe that VEGF and pro-inflammatory cytokines may be potential therapeutic targets in the treatment of this disease although further studies are required to confirm these findings

    The stellar masses of 25000 galaxies at 0.2<z<1.0 estimated by the COMBO-17 survey

    We present an analysis of stellar mass estimates for a sample of 25000 galaxies from the COMBO-17 survey over the interval 0.2<z<1.0. We have developed, implemented, and tested a new method of estimating stellar mass-to-light ratios, which relies on redshift and spectral energy distribution (SED) classification from 5 broadband and 12 medium band filters. We find that the majority (>60%) of massive galaxies with M_* > 10^{11} solar masses at all z<1 are non-star-forming; blue star-forming galaxies dominate at lower masses. We have used these mass estimates to explore the evolution of the stellar mass function since z=1. We find that the total stellar mass density of the universe has roughly doubled since z~1. Our measurements are consistent with other measurements of the growth of stellar mass with cosmic time and with estimates of the time evolution of the cosmic star formation rate. Intriguingly, the integrated stellar mass of blue galaxies with young stars has not significantly changed since z~1, even though these galaxies host the majority of the star formation: instead, the growth of the total stellar mass density is dominated by the growth of the total mass in the largely passive galaxies on the red sequence.Comment: Astronomy and Astrophysics in press. 15 pages, 12 figure

    Investigation of the Antiremodeling Effects of Losartan, Mirabegron and Their Combination on the Development of Doxorubicin-Induced Chronic Cardiotoxicity in a Rat Model

    Despite the effectiveness of doxorubicin (DOXO) as a chemotherapeutic agent, dose-dependent development of chronic cardiotoxicity limits its application. The angiotensin-II receptor blocker losartan is commonly used to treat cardiac remodeling of various etiologies. The beta-3 adrenergic receptor agonist mirabegron was reported to improve chronic heart failure. Here we investigated the effects of losartan, mirabegron and their combination on the development of DOXO-induced chronic cardiotoxicity. Male Wistar rats were divided into five groups: (i) control; (ii) DOXO-only; (iii) losartan-treated DOXO; (iv) mirabegron-treated DOXO; (v) losartan plus mirabegron-treated DOXO groups. The treatments started 5 weeks after DOXO administration. At week 8, echocardiography was performed. At week 9, left ventricles were prepared for histology, qRT-PCR, and Western blot measurements. Losartan improved diastolic but not systolic dysfunction and ameliorated SERCA2a repression in our DOXO-induced cardiotoxicity model. The DOXO-induced overexpression of Il1 and Il6 was markedly decreased by losartan and mirabegron. Mirabegron and the combination treatment improved systolic and diastolic dysfunction and significantly decreased overexpression of Smad2 and Smad3 in our DOXO-induced cardiotoxicity model. Only mirabegron reduced DOXO-induced cardiac fibrosis significantly. Mirabegron and its combination with losartan seem to be promising therapeutic tools against DOXO-induced chronic cardiotoxicity.Peer reviewe

    Role of direct bioautographic method for detection of antistaphylococcal activity of essential oils.

    The aim of the present study was the chemical characterization of some traditionally used and therapeutically relevant essential oils (thyme, eucalyptus, cinnamon bark, clove, and tea tree) and the optimized microbiological investigation of the effect of these oils on clinical isolates of methicillin-resistant Staphylococcus aureus (MRSA) and methicillin-susceptible S. aureus (MSSA). The chemical composition of the oils was analyzed by TLC, and controlled by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). The antibacterial effect was investigated using a TLC-bioautographic method. Antibacterial activity of thyme, clove and cinnamon oils, as well as their main components (thymol, carvacrol, eugenol, and cinnamic aldehyde) was observed against all the bacterial strains used in this study. The essential oils of eucalyptus and tea tree showed weak activity in the bioautographic system. On the whole, the antibacterial activity of the essential oils could be related to their most abundant components, but the effect of the minor components should also be taken into consideration. Direct bioautography is more cost-effective and better in comparison with traditional microbiological laboratory methods (e.g. disc-diffusion, agar-plate technique)

    Fetal renal artery flow and renal echogenicity in the chronically hypoxic state.

    peer reviewedThe object of this study was to investigate the fetal renal arterial blood flow in normal and hyperechogenic kidneys during the third trimester of gestation. The pregnancies screened were all chronically hypoxic. Depending on the etiology of the intrauterine chronic hypoxia, the cases were divided into two study groups. Group I comprised 120 pregnant women with pregnancy-associated hypertension and/or proteinuria. Group II consisted of 87 pregnancies with intrauterine growth retardation. Both study groups included pregnant women from the third trimester. Hyperechogenic renal medullae were detected in 15 out of 120 cases with pregnancy-associated hypertension and/or proteinuria, and in 22 fetuses of the 87 pregnancies involving intrauterine growth retardation. Fetal renal hyperechogenicity appears to be an indicator of fetal arterial circulatory depression, correlated with pathological changes in the resistance index for the fetal renal arteries. The fetal renal arterial blood flow resistance index was significantly lower in hyperechogenic cases. This may also be an in utero indication of subsequent intrauterine and neonatal complications, such as cesarean section because of fetal distress (43%), treatment in a neonatal intensive care unit (51%) or increased perinatal mortality (5.4%, as compared with 0.8-1.0% in the normal population). Detailed ultrasound and Doppler examinations of renal parenchyma and arteries appear to be useful methods in the prenatal diagnosis of reduced renal perfusion and of intrauterine hypoxia to detect possible pathological fetal conditions in utero
