    CB2 Receptor Deficiency Increases Amyloid Pathology and Alters Tau Processing in a Transgenic Mouse Model of Alzheimer\u27s Disease

    The endocannabinoid CB2 receptor system has been implicated in the neuropathology of Alzheimer\u27s disease (AD). In order to investigate the impact of the CB2 receptor system on AD pathology, a colony of mice with a deleted CB2 receptor gene, CNR2, was established on a transgenic human mutant APP background for pathological comparison with CB2 receptor-sufficient transgenic mice. J20 APP (PDGFB-APPSwInd) mice were bred over two generations with CNR2(-/-) (Cnr2(tm1Dgen)/J) mice to produce a colony of J20 CNR2(+/+) and J20 CNR2(-/-)mice. Seventeen J20 CNR2(+/+) mice (12 females, 5 males) and 16 J20 CNR2(-/-) mice (11 females, 5 males) were killed at 12 months, and their brains were interrogated for AD-related pathology with both biochemistry and immunocytochemistry (ICC). In addition to amyloid-dependent endpoints such as soluble A beta production and plaque deposition quantified with 6E10 staining, the effect of CB2 receptor deletion on total soluble mouse tau production was assayed by using a recently developed high-sensitivity assay. Results revealed that soluble A beta 42 and plaque deposition were significantly increased in J20 CNR2(-/-) mice relative to CNR2(1/1) mice. Microgliosis, quantified with ionized calcium-binding adapter molecule 1 (Iba-1) staining, did not differ between groups, whereas plaque associated microglia was more abundant in J20 CNR2(-/-) mice. Total tau was significantly suppressed in J20 CNR2(-/-) mice relative to J20 CNR2(+/+) mice. The results confirm the constitutive role of the CB2 receptor system both in reducing amyloid plaque pathology in AD and also support tehpotential of cannabinoid therapies targeting CB2 to reduce A beta; however, the results suggest that interventions may have a divergent effect on tau pathology

    Recent trends in digital text forensics and its evaluation

    The final publication is available at Springer via http://dx.doi.org/10.1007/978-3-642-40802-1_28This paper outlines the concepts and achievements of our evaluation lab on digital text forensics, PAN 13, which called for original research and development on plagiarism detection, author identification, and author profiling. We present a standardized evaluation framework for each of the three tasks and discuss the evaluation results of the altogether 58 submitted contributions. For the first time, instead of accepting the output of software runs, we collected the softwares themselves and run them on a computer cluster at our site. As evaluation and experimentation platform we use TIRA, which is being developed at the Webis Group in Weimar. TIRA can handle large-scale software submissions by means of virtualization, sandboxed execution, tailored unit testing, and staged submission.     The benefits of interprofessional education 10 years on.

    Interprofessional education (IPE) was first conceived in 1973 by a World Health Organization (WHO) expert group in Geneva. WHO member states were then charged with implementing medical education IPE pilot projects and from then to today there has a been a rapid proliferation in the number of publications on the subject. IPE has generated research into its use, conferences specific to IPE, organisations dedicated to it and policy championing it. The authors question whether there has been any major shift in the silos in which different professions might be working. The authors published an article on the benefits of IPE ( Illingworth and Chelvanayagam, 2007 ). Ten years have now passed and many changes have been implemented and experienced in health and social care and therefore a review of the literature is required. Also, it is 7 years since the publication of WHO's report outlining the role of IPE in the preparation of health professionals ( WHO, 2010 ) and, increasingly, UK Government policy champions collaborative and integrated working. The conclusions from the 2007 article acknowledged the development of IPE; however, it highlighted the need for empirical evidence to demonstrate the effectiveness of IPE in service user and carer outcomes. This article will explore whether IPE has achieved the benefits discussed in the previous article and what developments have occurred since it was published

    Persistence, extinction and spatio-temporal synchronization of SIRS cellular automata models

    Spatially explicit models have been widely used in today's mathematical ecology and epidemiology to study persistence and extinction of populations as well as their spatial patterns. Here we extend the earlier work--static dispersal between neighbouring individuals to mobility of individuals as well as multi-patches environment. As is commonly found, the basic reproductive ratio is maximized for the evolutionary stable strategy (ESS) on diseases' persistence in mean-field theory. This has important implications, as it implies that for a wide range of parameters that infection rate will tend maximum. This is opposite with present results obtained in spatial explicit models that infection rate is limited by upper bound. We observe the emergence of trade-offs of extinction and persistence on the parameters of the infection period and infection rate and show the extinction time having a linear relationship with respect to system size. We further find that the higher mobility can pronouncedly promote the persistence of spread of epidemics, i.e., the phase transition occurs from extinction domain to persistence domain, and the spirals' wavelength increases as the mobility increasing and ultimately, it will saturate at a certain value. Furthermore, for multi-patches case, we find that the lower coupling strength leads to anti-phase oscillation of infected fraction, while higher coupling strength corresponds to in-phase oscillation.Comment: 12page

    Psychotic Alzheimer\u27s disease is associated with gender-specific tau phosphorylation abnormalities

    Converging evidence suggests that psychotic Alzheimer\u27s disease (AD + P) is associated with an acceleration of frontal degeneration, with tau pathology playing a primary role. Previous histopathologic and biomarker studies have specifically implicated tau pathology in this condition. To precisely quantify tau abnormalities in the frontal cortex in AD + P, we used a sensitive biochemical assay of total tau and 4 epitopes of phospho-tau relevant in AD pathology in a postmortem sample of AD + P and AD - P. Samples of superior frontal gyrus from 26 AD subjects without psychosis and 45 AD + P subjects with psychosis were analyzed. Results of enzyme-linked immunosorbent assay demonstrate that AD + P females, but not males, had significantly higher levels of phosphorylated tau in the frontal cortex. In males, but not females, AD + P was associated with the presence of alpha-synuclein pathology. These results support a gender dissociation of pathology in AD + P. The design of future studies aimed at the elucidation of cognitive and/or functional outcomes; regional brain metabolic deficits; or genetic correlates of AD + P should take gender into consideration.     Interprofessional Education: An evaluation of a joint learning workshop for podiatry and pharmacy students

    "Interprofessional Education occurs when two or more professionals learn with, from and about each other to improve collaboration and the quality of care" (CAIPE 2002). Interprofessional education forms part of the Standards for the Initial Education and Training of Pharmacists. Working with and understanding the role of another profession has been shown to positively impact on the quality of care of the patient. Following positive pharmacy student feedback from visits to podiatry clinics an interprofessional learning workshop with case - based scenarios was developed. These were based on patients with high risk medical conditions that would impact on the work of both professions. Data from the feedback forms was evaluated and analysed to determine whether the workshop increased knowledge of the British National Formulary (BNF), the prescribing process and gave an insight in to the role of other healthcare professionals. We discuss how the student’s learning has been enhanced by the contribution of another professional group. The workshop was positively received. Students were observed working together discussing the patients’ conditions and issues relating to their care. This initially revolved around the students’ area of knowledge; however, as the session progressed it became apparent that the students were learning with, from and about each other for the benefit of patient care

    A Decade of Shared Tasks in Digital Text Forensics at PAN

    [EN] Digital text forensics aims at examining the originality and credibility of information in electronic documents and, in this regard, to extract and analyze information about the authors of these documents. The research field has been substantially developed during the last decade. PAN is a series of shared tasks that started in 2009 and significantly contributed to attract the attention of the research community in well-defined digital text forensics tasks. Several benchmark datasets have been developed to assess the state-of-the-art performance in a wide range of tasks. In this paper, we present the evolution of both the examined tasks and the developed datasets during the last decade.     Celestial Mechanics, Conformal Structures, and Gravitational Waves

    The equations of motion for NN non-relativistic particles attracting according to Newton's law are shown to correspond to the equations for null geodesics in a (3N+2)(3N+2)-dimensional Lorentzian, Ricci-flat, spacetime with a covariantly constant null vector. Such a spacetime admits a Bargmann structure and corresponds physically to a generalized pp-wave. Bargmann electromagnetism in five dimensions comprises the two Galilean electro-magnetic theories (Le Bellac and L\'evy-Leblond). At the quantum level, the NN-body Schr\"odinger equation retains the form of a massless wave equation. We exploit the conformal symmetries of such spacetimes to discuss some properties of the Newtonian NN-body problem: homographic solutions, the virial theorem, Kepler's third law, the Lagrange-Laplace-Runge-Lenz vector arising from three conformal Killing 2-tensors, and motions under inverse square law forces with a gravitational constant G(t)G(t) varying inversely as time (Dirac). The latter problem is reduced to one with time independent forces for a rescaled position vector and a new time variable; this transformation (Vinti and Lynden-Bell) arises from a conformal transformation preserving the Ricci-flatness (Brinkmann). A Ricci-flat metric representing NN non-relativistic gravitational dyons is also pointed out. Our results for general time-dependent G(t)G(t) are applicable to the motion of point particles in an expanding universe.     Fast flowing populations are not well mixed

    In evolutionary dynamics, well-mixed populations are almost always associated with all-to-all interactions; mathematical models are based on complete graphs. In most cases, these models do not predict fixation probabilities in groups of individuals mixed by flows. We propose an analytical description in the fast-flow limit. This approach is valid for processes with global and local selection, and accurately predicts the suppression of selection as competition becomes more local. It provides a modelling tool for biological or social systems with individuals in motion.Comment: 19 pages, 8 figure
