NDM-525: EFFECTS OF TORNADO WIND SPEEDS ON CONCRETE ROAD BARRIERS
Wind speeds can be difficult to measure during tornadoes due to their destructive nature. They pose a significant threat to lives and infrastructure in many parts of Canada and the U.S. The Enhanced-Fujita scale focuses on estimating these wind speeds by observing damage to different types of buildings, but significantly less research has been performed on the damage of other structures. Learning more about the effects of high wind speeds on these structures will help improve the ease and accuracy of future tornado classification. A wind tunnel study was performed at the Boundary Layer Wind Tunnel Laboratory of Western University. The study focusses on estimating the wind speeds that cause overturning in a standard 32” concrete “Jersey” barrier. On April 27, 2014, an EF4 Tornado struck Mayflower, Arkansas, and among the damage, several of these concrete barriers were blown over during the storm. The goal of this study was to find the overturning wind velocity and compare it to other damage in this event. This study was performed by placing a 1:8 scale-model of these barriers in a wind tunnel at a variety of orientations and wind speeds. Through analysis, it was determined that an instantaneous wind velocity of 4.55 to 4.85 m/s would cause overturning. These values correspond to an instantaneous wind speed of 340-360 km/h at full scale. It was estimated that the 3-second gust (used for EF rating) was 300-320 km/h, which sits at the top of the 267-322 km/h classification range for an EF4 tornado
Wind Speed Estimates for Garage Door Failures in Tornadoes
Severe wind events, such as tornadoes, pose a significant threat to lives and infrastructure in many locations around the world. Residential buildings are the structures most affected by these events, since they are widespread and often not designed to withstand severe loading. For the wood-frame, low-rise houses typical of North America, once the envelope of the building has been breached, such as through the failure of a garage door, the loss of the entire roof structure becomes much more likely. One of the issues with garage doors is their flexibility; as they begin to deflect under wind load, relatively large openings allow air flow into the internal volume. As a result of these positive pressures on the garage door, positive pressures are transferred into the internal volume, subsequently reducing the net wind load on the door. The objectives of this study are to determine failure net pressures of garage doors through experimental testing, and to combine those results with internal pressure models including the effects of garage door flexibility in order to estimate the failure wind speeds of garage doors. Six garage doors of various types are tested, and the failure wind speeds acquired through the internal pressure model are compared to the Enhanced Fujita Scale. Experimental testing found the failure net pressures of the garage doors to be between 0.42 and 1.75 kPa. With the internal pressure model showing that the net load on the garage doors is typically reduced to 34–46% of the external pressure, the resulting range of expected failure wind speeds obtained was 130–265 km/h. This range is found to encompass and exceed the expected failure wind speeds in the EF-Scale of 130–185 km/h, which would only be applicable for the weaker range of garage doors
The Northern Tornadoes Project - Uncovering Canada’s True Tornado Climatology
Canada is a vast country with most of its population located along its southern border. Large areas are sparsely populated and/or heavily forested, and severe weather reports are rare when thunderstorms occur there. Thus, it has been difficult to accurately assess the true tornado climatology and risk. It is also important to establish a reliable baseline for tornado-related climate change studies.
The Northern Tornadoes Project (NTP), led by Western University, is an ambitious multidisciplinary initiative aimed at detecting and documenting every tornado that occurs across Canada. A team of meteorologists and wind engineers collects research-quality data during each damage investigation, via thorough ground surveys and high-resolution satellite, aircraft and drone imaging. Crowdsourcing through social media is also key to tracking down events. In addition, NTP conducts research to improve our ability to detect and accurately assess tornadoes that affect forests, cropland and grassland. An open data website allows sharing of resulting data sets and analyses.
Pilot investigations were carried out during the warm seasons of 2017 and 2018, with the scope expanding from the detection of any tornadoes in heavily forested regions of central Canada in 2017 to the detection of all EF1+ tornadoes in Ontario plus all significant events outside of Ontario in 2018. The 2019 season was the first full campaign, systemically collecting research-quality tornado data across the entire country. To date, the Project has found 89 tornadoes that otherwise would not have been identified, and increased the national tornado count in 2019 by 78%.

CoGeNT Interpretations
Recently, the CoGeNT experiment has reported events in excess of expected
background. We analyze dark matter scenarios which can potentially explain this
signal. Under the standard case of spin independent scattering with equal
couplings to protons and neutrons, we find significant tensions with existing
constraints. Consistency with these limits is possible if a large fraction of
the putative signal events is coming from an additional source of experimental
background. In this case, dark matter recoils cannot be said to explain the
excess, but are consistent with it. We also investigate modifications to dark
matter scattering that can evade the null experiments. In particular, we
explore generalized spin independent couplings to protons and neutrons, spin
dependent couplings, momentum dependent scattering, and inelastic interactions.
We find that some of these generalizations can explain most of the CoGeNT
events without violation of other constraints. Generalized couplings with some
momentum dependence, allows further consistency with the DAMA modulation
signal, realizing a scenario where both CoGeNT and DAMA signals are coming from
dark matter. A model with dark matter interacting and annihilating into a new

Maresin 1 promotes inflammatory resolution, neuroprotection, and functional neurological recovery after Spinal Cord Injury
Resolution of inflammation is defective after spinal cord injury (SCI), which impairs tissue integrity and remodeling and leads to functional deficits. Effective pharmacological treatments for SCI are not currently available. Maresin 1 (MaR1) is a highly conserved specialized proresolving mediator (SPM) hosting potent anti-inflammatory and proresolving properties with potent tissue regenerative actions. Here, we provide evidence that the inappropriate biosynthesis of SPM in the lesioned spinal cord hampers the resolution of inflammation and leads to deleterious consequences on neurological outcome in adult female mice. We report that, after spinal cord contusion injury in adult female mice, the biosynthesis of SPM is not induced in the lesion site up to 2 weeks after injury. Exogenous administration of MaR1, a highly conserved SPM, propagated inflammatory resolution after SCI, as revealed by accelerated clearance of neutrophils and a reduction in macrophage accumulation at the lesion site. In the search of mechanisms underlying the proresolving actions of MaR1 in SCI, we found that this SPM facilitated several hallmarks of resolution of inflammation, including reduction of proinflammatory cytokines (CXCL1, CXCL2, CCL3, CCL4, IL6, and CSF3), silencing of major inflammatory intracellular signaling cascades (STAT1, STAT3, STAT5, p38, and ERK1/2), redirection of macrophage activation toward a prorepair phenotype, and increase of the phagocytic engulfment of neutrophils by macrophages. Interestingly, MaR1 administration improved locomotor recovery significantly and mitigated secondary injury progression in a clinical relevant model of SCI. These findings suggest that proresolution, immunoresolvent therapies constitute a novel approach to improving neurological recovery after acute SCI.SIGNIFICANCE STATEMENT Inflammation is a protective response to injury or infection. To result in tissue homeostasis, inflammation has to resolve over time. Incomplete or delayed resolution leads to detrimental effects, including propagated tissue damage and impaired wound healing, as occurs after spinal cord injury (SCI). We report that inflammation after SCI is dysregulated in part due to inappropriate synthesis of proresolving lipid mediators. We demonstrate that the administration of the resolution agonist referred to as maresin 1 (MaR1) after SCI actively propagates resolution processes at the lesion site and improves neurological outcome. MaR1 is identified as an interventional candidate to attenuate dysregulated lesional inflammation and to restore functional recovery after SCI
Pan-Cancer Analysis of lncRNA Regulation Supports Their Targeting of Cancer Genes in Each Tumor Context
Long noncoding RNAs (lncRNAs) are commonly dys-regulated in tumors, but only a handful are known toplay pathophysiological roles in cancer. We inferredlncRNAs that dysregulate cancer pathways, onco-genes, and tumor suppressors (cancer genes) bymodeling their effects on the activity of transcriptionfactors, RNA-binding proteins, and microRNAs in5,185 TCGA tumors and 1,019 ENCODE assays.Our predictions included hundreds of candidateonco- and tumor-suppressor lncRNAs (cancerlncRNAs) whose somatic alterations account for thedysregulation of dozens of cancer genes and path-ways in each of 14 tumor contexts. To demonstrateproof of concept, we showed that perturbations tar-geting OIP5-AS1 (an inferred tumor suppressor) andTUG1 and WT1-AS (inferred onco-lncRNAs) dysre-gulated cancer genes and altered proliferation ofbreast and gynecologic cancer cells. Our analysis in-dicates that, although most lncRNAs are dysregu-lated in a tumor-specific manner, some, includingOIP5-AS1, TUG1, NEAT1, MEG3, and TSIX, synergis-tically dysregulate cancer pathways in multiple tumorcontexts
Pan-cancer Alterations of the MYC Oncogene and Its Proximal Network across the Cancer Genome Atlas
Although theMYConcogene has been implicated incancer, a systematic assessment of alterations ofMYC, related transcription factors, and co-regulatoryproteins, forming the proximal MYC network (PMN),across human cancers is lacking. Using computa-tional approaches, we define genomic and proteo-mic features associated with MYC and the PMNacross the 33 cancers of The Cancer Genome Atlas.Pan-cancer, 28% of all samples had at least one ofthe MYC paralogs amplified. In contrast, the MYCantagonists MGA and MNT were the most frequentlymutated or deleted members, proposing a roleas tumor suppressors.MYCalterations were mutu-ally exclusive withPIK3CA,PTEN,APC,orBRAFalterations, suggesting that MYC is a distinct onco-genic driver. Expression analysis revealed MYC-associated pathways in tumor subtypes, such asimmune response and growth factor signaling; chro-matin, translation, and DNA replication/repair wereconserved pan-cancer. Spatial Organization and Molecular Correlation of Tumor-Infiltrating Lymphocytes Using Deep Learning on Pathology Images
Beyond sample curation and basic pathologic characterization, the digitized H&E-stained images
of TCGA samples remain underutilized. To highlight this resource, we present mappings of tumorinfiltrating lymphocytes (TILs) based on H&E images from 13 TCGA tumor types. These TIL
maps are derived through computational staining using a convolutional neural network trained to
classify patches of images. Affinity propagation revealed local spatial structure in TIL patterns and
correlation with overall survival. TIL map structural patterns were grouped using standard
histopathological parameters. These patterns are enriched in particular T cell subpopulations
derived from molecular measures. TIL densities and spatial structure were differentially enriched
among tumor types, immune subtypes, and tumor molecular subtypes, implying that spatial
infiltrate state could reflect particular tumor cell aberration states. Obtaining spatial lymphocytic
patterns linked to the rich genomic characterization of TCGA samples demonstrates one use for
the TCGA image archives with insights into the tumor-immune microenvironment
Highly contiguous assemblies of 101 drosophilid genomes
Over 100 years of studies in Drosophila melanogaster and related species in the genus Drosophila have facilitated key discoveries in genetics, genomics, and evolution. While high-quality genome assemblies exist for several species in this group, they only encompass a small fraction of the genus. Recent advances in long-read sequencing allow high-quality genome assemblies for tens or even hundreds of species to be efficiently generated. Here, we utilize Oxford Nanopore sequencing to build an open community resource of genome assemblies for 101 lines of 93 drosophilid species encompassing 14 species groups and 35 sub-groups. The genomes are highly contiguous and complete, with an average contig N50 of 10.5 Mb and greater than 97% BUSCO completeness in 97/101 assemblies. We show that Nanopore-based assemblies are highly accurate in coding regions, particularly with respect to coding insertions and deletions. These assemblies, along with a detailed laboratory protocol and assembly pipelines, are released as a public resource and will serve as a starting point for addressing broad questions of genetics, ecology, and evolution at the scale of hundreds of species
