    Arsitektur di nusantara saat ini lebih mengedepankan ‘tren\u27 modern dan kurang mempertimbangkan seni ketukangan dengan aspek lokal dalam berkarya sehingga identitas arsitektur nusantara semakin terkikis dan budaya/kearifan lokal bukan lagi menjadi pertimbangan utama para penggiat arsitektur dalam merancang. Perancangan objek ini berusaha untuk meningkatkan kemampuan para tukang agar terus berinovasi dalam mencipta serta mengenalkan potensi ketukangan lokal kepada masyarakat umum. Dengan mengeksplorasi potensi alam dan ketukangan menggunakan pendekatan arsitektur ekologis kedalam objek rancang Galeri Seni Ketukangan diharapkan dapat melestarikan identitas kelokalan arsitektur nusantara serta para tukang di masa depan dapat terus berinovasi dalam berkarya dan dapat menarik antusias masyarakat terutama para penggiat arsitektur untuk kembali menerapkan aspek budaya lokal nusantara dalam mencipta karya. Objek rancang mewadahi kegiatan eksplorasi ketukangan untuk melestarikan identitas serta potensi material arsitektur di nusantara. Metoda desain yang akan digunakan adalah metoda desain William M. Pena & Steven A. Pharsall dengan pendekatan ekologis guna meminimalisir dampak objek rancang terhadap lahan dan lingkungan

    A programmable triangular neighborhood function for a Kohonen self-organizing map implemented on chip

    An efficient transistor level implementation of a flexible, programmable Triangular Function (TF) that can be used as a Triangular Neighborhood Function (TNF) in ultra-low power, self-organizing maps (SOMs) realized as Application-Specific Integrated Circuit (ASIC) is presented. The proposed TNF block is a component of a larger neighborhood mechanism, whose role is to determine the distance between the winning neuron and all neighboring neurons. Detailed simulations carried out for the software model of such network show that the TNF forms a good approximation of the Gaussian Neighborhood Function (GNF), while being implemented in a much easier way in hardware. The overall mechanism is very fast. In the CMOS 0.18 mu m technology, distances to all neighboring neurons are determined in parallel, within the time not exceeding 11 ns, for an example neighborhood range, R, of 15. The TNF blocks in particular neurons require another 6 ns to calculate the output values directly used in the adaptation process. This is also performed in parallel in all neurons. As a result, after determining the winning neuron, the entire map is ready for the adaptation after the time not exceeding 17 ns, even for large numbers of neurons. This feature allows for the realization of ultra low power SOMs, which are hundred times faster than similar SOMs realized on PC. The signal resolution at the output of the TNF block has a dominant impact on the overall energy consumption as well as the silicon area. Detailed system level simulations of the SOM show that even for low resolutions of 3 to 6 bits, the learning abilities of the SUM are not affected. The circuit performance has been verified by means of transistor level Hspice simulations carried out for different transistor models and different values of supply voltage and the environment temperature - a typical procedure completed in case of commercial chips that makes the obtained results reliable.     The functional cooperation of 5−HT1A5-HT_{1A} and mGlu4R in HEK-293 cell line

    Background The serotonin 5-HT1A receptor (5-HT1AR) and metabotropic glutamate receptor 4 (mGlu4) have been implicated as sites of antipsychotic drug action. 5-HT1AR belongs to the A class of G protein-coupled receptors (GPCRs); mGlu4 is a representative of class C GPCRs. Both receptors preferentially couple with Gi protein to inhibit cAMP formation. The present work aimed to examine the possibility of mGlu4 and 5-HT1A receptor cross-talk, the phenomenon that could serve as a molecular basis of the interaction of these receptor ligands observed in behavioral studies. Methods First, in vitro studies were performed to examine the pharmacological modulation of interaction of the mGlu4 and 5-HT1A receptors in the T-REx 293 cell line using SNAP- or HALO-tag and cAMP accumulation assay. Next, the colocalization of these two receptors was examined in some regions of the mouse brain by applying RNAScope dual fluorescence in situ hybridization, immunohistochemical labeling, and proximity ligation assay (PLA). Results The ex vivo and in vitro results obtained in the present work suggest the existence of interactions between mGlu4 and 5-HT1A receptors. The changes were observed in cAMP accumulation assay and were dependent on expression and activation of mGlu4R in T-REx 293cell line. Moreover, the existence of spots with proximity expression of both receptors were showed by PLA, immunofluorescence labeling and RNAscope methods. Conclusion The existence of interactions between mGlu4 and 5-HT1A receptors may represent another signaling pathway involved in the development and treatment psychiatric disorders such as schizophrenia or depression

    Mesolimbic dopamine D_{2} receptor plasticity contributes to stress resilience in rats subjected to chronic mild stress

    RATIONALE: Few studies have investigated neurobiological and biochemical differences between stress-resilient and stress-vulnerable experimental animals. OBJECTIVES: We investigated alterations in mesolimbic dopamine D(2) receptor density and mRNA expression level in stressed rats at two time points, i.e. after 2 and 5 weeks of chronic mild stress (CMS). METHODS: We used the chronic mild stress paradigm because it is a well-established animal model of depression. Two groups of stressed rats were distinguished during CMS experiments: (1) stress reactive (70 %), which displayed a decrease in the drinking of a palatable sucrose solution during the stress regimen, and (2) stress resilient (30 %), which exhibited an unaltered drinking profile when compared with the unchallenged control group. [(3)H]Domperidone was used as a ligand to label dopamine D(2) receptors, and a mixture of three specific oligonucleotides was used to evaluate dopamine D(2) receptor mRNA changes in various regions of the rat brain. RESULTS: CMS strongly affected the mesolimbic dopamine circuit in stress-resilient group after 2 weeks and stress-reactive group of rats after 5 weeks which exhibited a decrease in the level of dopamine D(2) receptor protein without alterations in D(2) mRNA expression. Stress-resilient animals, but not stress-reactive animals, effectively adapted to the extended stress and coped with it. The increase in D(2) mRNA expression returned the dopamine D(2) receptor density to control levels in stress-resilient rats after 5 weeks of CMS, but not in stress-reactive animals. CONCLUSIONS: These results clearly demonstrate that, despite earlier blunting, the activation of dopamine receptor biosynthesis in the dopamine mesoaccumbens system in stress-resilient rats is involved in active coping with stressful experiences, and it exhibits a delay in time

    Habenula as a possible target for treatment-resistant depression phenotype in Wistar Kyoto rats

    The mechanisms of treatment-resistant depression (TRD) are not clear and are difficult to study. An animal model resembling human TRD is the Wistar Kyoto rat strain. In the present study, we focused on selecting miRNAs that differentiate rats of the WKY strain from Wistar Han (WIS) rats in two divisions of the habenula, the lateral and medial (LHb and MHb, respectively). Based on our preliminary study and literature survey, we identified 32 miRNAs that could be potentially regulated in the habenula. Six miRNAs significantly differentiated WKY rats from WIS rats within the MHb, and three significantly differentiated WKY from WIS rats within the LHb. Then, we selected relevant transcripts regulated by those miRNAs, and their expression in the habenular nuclei was investigated. For mRNAs that differentiated WKY rats from WIS rats in the MHb (Cdkn1c, Htr7, Kcnj9, and Slc12a5), their lower expression correlated with a higher level of relevant miRNAs. In the LHb, eight mRNAs significantly differentiated WKY from WIS rats (upregulated Htr4, Drd2, Kcnj5, and Sstr4 and downregulated Htr2a, Htr7, Elk4, and Slc12a5). These data indicate that several important miRNAs are expressed in the habenula, which differentiates WKY rats from WIS rats and in turn correlates with alterations in the expression of target transcripts. Of particular note are two genes whose expression is altered in WKY rats in both LHb and MHb: Slc12a5 and Htr7. Regulation of KCC2 via the 5-HT7 receptor may be a potential target for the treatment of TRD

KONS (Kompleksowa opieka nad chorymi z niewydolnoƛcią serca) is the name of a Polish programme for the coordinated care for patients with heart failure that has been proposed as a complex heart failure management solution based on European Society of Cardiology (ESC) Heart Failure recommendations. The challenge is that there are currently no available systemic solutions for educating these patients. Our project aimed to evaluate the effectiveness of a pilot model of education and telemonitoring known as ‘The Weak Heart’ for patients hospitalised due to decompensation of heart failure with reduced ejection fraction (HFrEF). Materials and methods. ‘The Weak Heart’ educational model consists of three phases: phase I - training and certification of nurses to perform the role of heart failure nurse specialists; phase II - checking the effectiveness of the model in a clinical setting; and phase III - implementation of patient education standards in cardiac centres. Data collected will be used to evaluate the impact of the programme on patients’ understanding regarding heart failure, their compliance with recommendations, and their pro-health behaviours in terms of self-care and self-control. Conclusion. ‘The Weak Heart’ programme will allow conclusions to be drawn concerning the usefulness and effectiveness of the first targeted heart failure-based patient educational model in Poland. Positive results in this pilot project could become the basis for implementing systemic changes, and introducing the model into everyday clinical practice
