Peripheral and central compensatory mechanisms for impaired vagus nerve function during peripheral immune activation
Background
Determining the etiology and possible treatment strategies for numerous diseases requires a comprehensive understanding of compensatory mechanisms in physiological systems. The vagus nerve acts as a key interface between the brain and the peripheral internal organs. We set out to identify mechanisms compensating for a lack of neuronal communication between the immune and the central nervous system (CNS) during infection.
Methods
We assessed biochemical and central neurotransmitter changes resulting from subdiaphragmatic vagotomy and whether they are modulated by intraperitoneal infection. We performed a series of subdiaphragmatic vagotomy or sham operations on male Wistar rats. Next, after full, 30-day recovery period, they were randomly assigned to receive an injection of Escherichia coli lipopolysaccharide or saline. Two hours later, animal were euthanized and we measured the plasma concentration of prostaglandin E2 (with HPLC-MS), interleukin-6 (ELISA), and corticosterone (RIA). We also had measured the concentration of monoaminergic neurotransmitters and their metabolites in the amygdala, brainstem, hippocampus, hypothalamus, motor cortex, periaqueductal gray, and prefrontal medial cortex using RP-HPLC-ED. A subset of the animals was evaluated in the elevated plus maze test immediately before euthanization.
Results
The lack of immunosensory signaling of the vagus nerve stimulated increased activity of discrete inflammatory marker signals, which we confirmed by quantifying biochemical changes in blood plasma. Behavioral results, although preliminary, support the observed biochemical alterations. Many of the neurotransmitter changes observed after vagotomy indicated that the vagus nerve influences the activity of many brain areas involved in control of immune response and sickness behavior. Our studies show that these changes are largely eliminated during experimental infection.
Conclusions
Our results suggest that in vagotomized animals with blocked CNS, communication may transmit via a pathway independent of the vagus nerve to permit restoration of CNS activity for peripheral inflammation control
Central mechansms of compensation a long-term impairment of immunosensoric and immunosuppressive functions of vagus nerve
Nerw błędny jest jedną z głównych dróg neuroimmunokomunikacji. Zaburzenie aktywności nerwu obserwowane jest w przebiegu wielu chorób neurodegeneracyjnych i psychicznych, w których obserwuje się również zmiany procesów zapalnych. Dane literaturowe sugerują, iż efekty wagotomii podprzeponowej (VG) są zmienne w czasie i początkowe nasilenie efektów prozapalnych ustępuje wraz z wydłużającym się okresem rekonwalescencji po zabiegu.
W badaniach przeprowadzonych miesiąc po zabiegu VG, sprawdzono; 1) aktywność neurotransmisyjną wybranych obszarów OUN, 2) czy istnieją przesłanki o rozwijaniu się alternatywnych mechanizmów kompensujących immunosensoryczne funkcje nerwu, 3) jakie mogą być neurotransmisyjne podstawy przywrócenia przeciwzapalnej aktywności osi HPA, 4) czy występują zaburzenia zachowań chorobowych, 5) czy istnieją neurotransmisyjne przesłanki do powstawania zaburzeń społecznego aspektu zachowań chorobowych.
Badania przeprowadzono na szczurach stada niekrewniaczego Wistar. Wykonano behawioralny test EPM oraz wykorzystano szereg technik biochemicznych i molekularnych takich jak HPLC, RIA, ELISA, mikromacierze RNA i RT-qPCR.
Uzyskane dane sugerują, że trzydzieści dni po zabiegu VG 1) zmienia się funkcjonowanie systemów serotoninergicznego i dopaminergicznego, a 2) zmiany te, są częściowo niwelowane na wczesnym etapie rozwoju śródotrzewnowego stanu zapalnego. Pomimo to, 3) aktywność wydzielnicza osi HPA jest podobna do tej u zwierząt kontrolnych, prawdopodobnie na skutek hamującej aktywności ciał migdałowatych. 4) Aktywność lokomotoryczna zwierząt jest obniżona, niezależnie od występowania u nich stanu zapalnego. 5) VG może prowadzić do osłabienia zainteresowania kontaktami społecznymi a ponadto, może intensyfikować odczucia bólowe poprzez zaburzenie obwodowego wydzielania endogennych substancji przeciwbólowych.
Uzyskane dane stanowią podstawę do bardziej szczegółowych badań na temat związku długotrwale obniżonej aktywności nerwu błędnego i zaburzeń psychicznych związanych
z wycofaniem społecznym, oraz chorób neurodegeneracyjnych.Praca została zrealizowana w ramach grantu NCN (2012/07/B/NZ4/00205)

Współczesne badania nad komunikacją neuroimmunologiczną pozwalają coraz lepiej zrozumieć odpowiedź odpornościową organizmu oraz zachowania człowieka i zwierząt w chorobie. Jedną z najważniejszych dróg tej komunikacji jest nerw błędny. Jego aferentne zakończenia zbierają informacje bezpośrednio, wykorzystując receptory dla cząsteczek sygnałowych układu odpornościowego oraz pośrednio dzięki komunikacji z innymi, wyspecjalizowanymi komórkami. Wskutek tych procesów ośrodkowy układ nerwowy otrzymuje informacje niezbędne do regulacji odpowiedzi odpornościowej, aktywności hormonalnej i zachowania. Celem niniejszego opracowania jest podsumowanie dotychczasowej wiedzy na temat procesów fizjologicznych pozwalających nerwowi błędnemu na pośredniczenie w komunikacji między pobudzonym układem odpornościowym a ośrodkowym układem nerwowym.Contemporary research on neuroimmune communication allowed us to expand on our understanding of the immune response as well as human and animals sickness behavior. One of the most important paths of communication is the vagus nerve. Its afferent endings gather information directly thanks to their receptors for signaling molecules of the immune system, as well as indirectly by communicating with other specialized cells. As a result, central nervous system collects information necessary for regulating the immune response, hormonal activity and behavior. The purpose of this work is to summarize our current knowledge on the physiological processes that allow the vagus nerve to mediate the communication between the immune system and the central nervous system
Class I HDAC Inhibition Leads to a Downregulation of FANCD2 and RAD51, and the Eradication of Glioblastoma Cells
HDAC inhibitors (HDACi) hold great potential as anticancer therapies due to their ability to regulate the acetylation of both histone and non-histone proteins, which is frequently disrupted in cancer and contributes to the development and advancement of the disease. Additionally, HDACi have been shown to enhance the cytotoxic effects of DNA-damaging agents such as radiation and cisplatin. In this study, we found that histone deacetylase inhibits valproic acid (VPA), synergized with PARP1 inhibitor (PARPi), talazoparib (BMN-673), and alkylating agent, and temozolomide (TMZ) to induce DNA damage and reduce glioblastoma multiforme. At the molecular level, VPA leads to a downregulation of FANCD2 and RAD51, and the eradication of glioblastoma cells. The results of this study indicate that combining HDACi with PARPi could potentially enhance the treatment of glioblastoma, the most aggressive type of cancer that originates in the brain
