TCR-transgenic T cells and YB-1-based oncolytic virotherapy improve survival in a preclinical Ewing sarcoma xenograft mouse model

BackgroundEwing sarcoma (EwS) is an aggressive and highly metastatic bone and soft tissue tumor in pediatric patients and young adults. Cure rates are low when patients present with metastatic or relapsed disease. Therefore, innovative therapy approaches are urgently needed. Cellular- and oncolytic virus-based immunotherapies are on the rise for solid cancers.MethodsHere, we assess the combination of EwS tumor-associated antigen CHM1319-specific TCR-transgenic CD8+ T cells and the YB-1-driven (i.e. E1A13S-deleted) oncolytic adenovirus XVir-N-31 in vitro and in a xenograft mouse model for antitumor activity and immunostimulatory properties.ResultsIn vitro both approaches specifically kill EwS cell lines in a synergistic manner over controls. This effect was confirmed in vivo, with increased survival using the combination therapy. Further in vitro analyses of immunogenic cell death and antigen presentation confirmed immunostimulatory properties of virus-infected EwS tumor cells. As dendritic cell maturation was also increased by XVir-N-31, we observed superior proliferation of CHM1319-specific TCR-transgenic CD8+ T cells only in virus-tested conditions, emphasizing the superior immune-activating potential of XVir-N-31.ConclusionOur data prove synergistic antitumor effects in vitro and superior tumor control in a preclinical xenograft setting. Combination strategies of EwS-redirected T cells and YB-1-driven virotherapy are a highly promising immunotherapeutic approach for EwS and warrant further evaluation in a clinical setting

„…Quella storia di sangue e corpi nudi…“

In seinem Frühwerk ruft Italo Calvino die Frage nach der Leistungsfähigkeit des literarischen Diskurses in Bezug auf die narrative Konstruktion von Erlebnis und Erfahrung auf. In der Dissertation wird aufgezeigt, dass die frühe Literatur Calvinos und die Bewegung des literarischen Neorealismus sich aus einem historischen Selbstverständnis herleiten, das wiederum selbst konstrukthaft ist. Calvino koppelt in seinem Frühwerk neorealistische Referenz an mediale Perspektivität und reflektiert das problematische Verhältnis von Erleben, Erfahren und Darstellen innerhalb der Texte metafiktional. Gleichzeitig bringt Calvino in seinem Frühwerk auch eine Zeitkritik vor, wenn er den Verlust des Vertrauens in die Leistungsfähigkeit politischer Ideologien evident macht. Beides – der umfassende Selbsterkundungsgedanke, aber auch Zeitkritik – findet sich in Calvinos späterer Narrativik wieder, womit sich eine größere Nähe des Frühwerks von Calvino zu seinem Spätwerk ergibt als bisher angenommen

Chamäleon Hochschuldidaktik

Die Reflexion von Kompetenzen, Rollen und Haltungen ist von großer Bedeutung für die eigene Professionalisierung und Weiterentwicklung von Hochschuldidaktiker/inne/n – und leistet damit einen Beitrag zu "guter Lehre", so die Grundannahme dieses Beitrags. Kontext der folgenden Darstellungen ist der Workshop "Chamäleon Hochschuldidaktik – Kompetenzen, Rollen und Haltungen von Hochschuldidaktiker/inne/n spielen eine Rolle", den wir, die Interne Fortbildung und Beratung (IFB) der Ruhr-Universität Bochum, im Rahmen der Dortmunder Spring School 2014 angeboten haben. Die Teilnehmenden haben sich anhand einer exemplarischen Arbeitssituation mit ihren Kompetenzen, Rollen und Haltungen auseinandergesetzt. Die Ergebnisse spiegeln die Rollenvielfalt sowie ein breites Kompetenzspektrum im Rahmen von hochschuldidaktischen Tätigkeiten wider. Es wird deutlich, dass die Haltung insbesondere im Rahmen des Tätigkeitsfelds "Beratung" relevant ist.Considering competences, roles and attitudes is important for optimizing and improving the work of higher educational institutions. The following explanations arose from discussions within the work-shop "Chameleon Hochschuldidaktik – attitudes, roles and competencies play a role" conducted at the Dortmunder Spring School in 2014 by the strategic unit for professional development, Ruhr-Universität Bochum. The participants looked at the day-to-day tasks undertaken by a model example and discussed its competencies, roles and attitudes. The results from the workshop show that there is a great variety of roles and a broad range of competencies in the field of higher education didactics. "Attitude" in particular is of increasing importance in the field of consulting, and so this article focuses in particular on that area of activity

Two Hundred Years After Once Upon a Time: The Legacy of the Brothers Grimm and Their Tales in Germany

Because there have been excellent studies about the influence and heritage of the Grimms’ tales up to 1990—I am thinking here of Donald Haase’s edited book The Reception of Grimms’ Fairy Tales—in this essay I focus on the legacy of the Grimms’ tales in the German sociocultural context of the last twenty years. I discuss some of the more recent popular manifestations of their stories in German literature and film and then move on to an analysis of scholarly studies that, in contrast to popular culture, have grounded the legacy of the Grimms in substantial ways. In the conclusion I argue that English-speaking scholars who study the Grimms’ fairy tales should be paying more attention to the inter- relationship between philology and narratology in the Grimms’ collection of folktales
