    Subsystems for future access networks

    Current evolution and tendencies of Telecom Networks in general and more specifically optical Metro and Access Networks and their convergence are reported. Based on this evolution, a set of research lines are foreseen regarding subsystems and devices as: high speed optical sources, modulators and receivers, for the next generation of Passive Optical Networks.     Characterization of rare germline variants in familial multiple myeloma

    Multiple myeloma (MM) is a malignancy of plasma cells, characterized by the presence of monoclonal immunoglobulin, known as M protein1. MM is preceded by monoclonal gammopathy of undetermined significance (MGUS) which is also a precursor of immunoglobulin light chain (AL) amyloidosis. Previous studies have reported a 2- to 4-fold increased risk of MGUS or MM in first-degree relatives of MM or MGUS patients, suggesting the existence of inherited susceptibility. For many years, high-risk germline predisposing genes have been lacking for MM. However, recent sequencing efforts have proposed a few novel candidates, most notably loss-offunction (LoF) variants in the tumor suppressor gene DIS3 and in the histone demethylase gene KDM1A, and others as recently reviewed in detail in Pertesi et al. In addition to the suspected rare, high-penetrance variants, genome-wide association studies have identified over 20 common, low-penetrance variants associated with the risk of MM; these were estimated to account for about 15% of the familial MM risk

    Inhibition of AP-1 signaling by JDP2 overexpression protects cardiomyocytes against hypertrophy and apoptosis induction

    AimsExpression and activity of the transcription factor AP-1 are enhanced during cardiac remodelling and heart failure progression. In order to test if AP-1 inhibition may limit processes contributing to cardiac remodelling, ventricular cardiomyocytes of mice with cardiac overexpression of the AP-1 inhibitor JDP2 were analysed under stimulation of hypertrophy, apoptosis, or contractile function.Methods and resultsThree models of JDP2 overexpressing mice were analysed: JDP2 was overexpressed either life-long, for 7 weeks, or 1 week. Then cardiomyocytes were isolated and stimulated with ÎČ-adrenoceptor agonist isoprenaline (ISO, 50 nM). This enhanced cross-sectional area and the rate of protein synthesis in WT but not in JDP2 overexpressing cardiomyocytes. To induce apoptosis, cardiomyocytes were stimulated with 3 ng/mL TGFÎČ1. Again, JDP2 overexpression prevented apoptosis induction compared with WT cells. Determination of contractile function under electrical stimulation at 2 Hz revealed enhancement of cell shortening, and contraction and relaxation velocities under increasing ISO concentrations (0.3-30 nM) in WT cells. This inotropic effect was abrogated in JDP2 overexpression cells. Responsiveness to increased extracellular calcium concentrations was also impaired in JDP2 overexpressing cardiomyocytes. Simultaneously, a reduction of SERCA expression was found in JDP2 mice.ConclusionA central role of AP-1 in the induction of hypertrophy and apoptosis in cardiomyocytes is demonstrated. Besides these protective effects of AP-1 inhibition on factors of cardiac remodelling, AP-1-inhibition impairs contractile function. Therefore, AP-1 acts as a double-edged sword that mediates mal-adaptive cardiac remodelling, but is required for maintaining a proper contractile function of cardiomyocytes. © 2013 Published on behalf of the European Society of Cardiology.     Protected Ethernet Rings for Optical Access Networks

    In this paper we propose a centralized link layer architecture for providing low latency fault recovery for optical access rings. This architecture exploits the naturally uneven breakdown of network management responsibilities between the components of an access ring. Important administrative operations like ring status checking, fault detection and recovery are aggregated at the HUB component located in the Central Office of the access network. Consequently, when compared with a standardized Rapid Spanning Tree Protocol implementation, the design of the proposed architecture benefits from a simplified link layer design of the Optical Network Unit, in addition to a significantly reduced fault recovery delay in the ring. We also present an Ethernet-based protocol that realizes our centralized protection model. The design principle of this protocol, responsible for the message passing required to react to topology changes in the network, is simple enough to allow quick reaction times, and to support QoS-aware prioritization of network traffic. The performance of the proposed architecture is evaluated using analytical and simulative means, and the performance aspects of the ring protocol relevant to network protection are compared with those provided by the Rapid Spanning Tree Protocol

    Abstract — We propose a rate-adaptive optical wireless transmission system based on orthogonal frequency division multiplexing for indoor communications. The investigations rely on realistic parameters of the key system components and focus on throughput maximization. We will show that a dynamically adaptive system can greatly enhance performance when compared to static operation, and how a loading algorithm, which optimally performs in powerlimited systems, needs to be adjusted if the specific terms of the optical wireless channel are to be rigorously obeyed. Our investigations include scenarios in which the non-negativity constraint on the optical source driving signal is strictly met and in which a certain amount of symmetric clipping is tolerated. In the latter case, the system can be regarded as power-limited and conventional loading algorithms are hence the most suitable. We will show that the transmission rate can be significantly improved even further by accepting a minor increase in the error rate as a result of controlled clipping, and we will compare our results with the upper system capacity limit.     QoS-aware Traffic Protection for Access Rings

The design principle of this protocol, responsible for the message passing required to react to topology changes resulting from faults, is to be simple enough to allow quick reaction times. The architecture prioritizes the restoration of network services according to their QoS importance, to ensure that the quality guarantees promised to them are honored at all times. The fault recovery delay of the proposed architecture is evaluated by simulation. In conjunction, the delay and jitter characteristics experienced by different traffic flows during periods of normal operation and faults are measured and compared
