Was macht die "Frische" von Brot aus?
Die Frische von Brot lässt sich nicht mit einheitlichen Kriterien beurteilen. Vielmehr hängt die Wahl der jeweiligen Kritereien von den Anforderungen ab, die an das Produkt gestellt werden. Sensorische Eigenschaften spielen dabei eine bedeutende Rolle. Proteomic Profiling of Mesenchymal Stem Cell Responses to Mechanical Strain and TGF-β1
Mesenchymal stem cells (MSCs) are a potential source of smooth muscle cells (SMCs) for constructing tissue-engineered vascular grafts. However, the details of how specific combinations of vascular microenvironmental factors regulate MSCs are not well understood. Previous studies have suggested that both mechanical stimulation with uniaxial cyclic strain and chemical stimulation with transforming growth factor-β1 (TGF-β1) can induce smooth muscle markers in MSCs. In this study, we investigated the combined effects of uniaxial cyclic strain and TGF-β1 stimulation on MSCs. By using a proteomic analysis, we found differential regulation of several proteins and genes, such as the up-regulation of TGF-β1-induced protein ig-h3 (BGH3) protein levels by TGF-β1 and up-regulation of calponin 3 protein level by cyclic strain. At the gene expression level, BGH3 was induced by TGF-β1, but calponin 3 was not significantly regulated by mechanical strain or TGF-β1, which was in contrast to the synergistic up-regulation of calponin 1 gene expression by cyclic strain and TGF-β1. Further experiments with cycloheximide treatment suggested that the up-regulation of calponin 3 by cyclic strain was at post-transcriptional level. The results in this study suggest that both mechanical stimulation and TGF-β1 signaling play unique and important roles in the regulation of MSCs at both transcriptional and post-transcriptional levels, and that a precise combination of microenvironmental cues may promote MSC differentiation

A reference human induced pluripotent stem cell line for large-scale collaborative studies.
Human induced pluripotent stem cell (iPSC) lines are a powerful tool for studying development and disease, but the considerable phenotypic variation between lines makes it challenging to replicate key findings and integrate data across research groups. To address this issue, we sub-cloned candidate human iPSC lines and deeply characterized their genetic properties using whole genome sequencing, their genomic stability upon CRISPR-Cas9-based gene editing, and their phenotypic properties including differentiation to commonly used cell types. These studies identified KOLF2.1J as an all-around well-performing iPSC line. We then shared KOLF2.1J with groups around the world who tested its performance in head-to-head comparisons with their own preferred iPSC lines across a diverse range of differentiation protocols and functional assays. On the strength of these findings, we have made KOLF2.1J and its gene-edited derivative clones readily accessible to promote the standardization required for large-scale collaborative science in the stem cell field
Identität und Motivation im Handball
Handball ist eine sehr bekannte und beliebte Sportart in ganz Europa. Die Zahl der Personen, welche von der Leidenschaft des Handballspiels gepackt werden, steigt stetig.
Aber dabei stellt sich die Frage, warum Menschen mit dieser Sportart beginnen und warum sie dabei bleiben? Welche GrĂźnde sind dafĂźr ausschlaggebend, ob man diese Sportart profi- oder amateurmäĂig betreibt? Welche Unterschiede lassen sich dabei erkennen? Welchen Stellenwert nimmt der Handball im Leben der betroffenen Personen ein?
Mit diesen und weiteren Themen setzt sich diese Diplomarbeit auseinander. Fßr die Ergebnissicherung wurde anhand von einschlägiger Literatur ein Fragebogen erstellt, welcher sich schon mehrfach bewährt hatte. Dieser Fragebogen, welcher zur Erhebung der einzelnen Motive und Identitäten im Handball dient, wurde von insgesamt 126 Probandinnen und Probanden im Alter von 14 und 36 Jahren ausgefßllt.
Die Ergebnisse des Fragebogens machen uns deutlich, dass vor allem soziale Faktoren wie Freunde, Bekannte oder Schule und Universität fßr den Erstkontakt mit der Sportart ausschlaggebend sind. Die Motive, warum die Spielerinnen und Spieler mit dem Handballspielen begonnen haben, beziehungsweise warum sie dabei geblieben sind, sind hauptsächlich von sozialer und erfolgsorientierter Natur. Den Personen ist es wichtig im und mit dem Team erfolgreich zu sein und dabei zusätzlich sowohl sportlich als auch menschlich etwas zu leisten.Handball is a well-known and popular sport in Europe. The number of people who are seized by the passion of the handball game is steadily rising.
But that raises the question of why people start this sport and why they stick to it? What are the reasons for playing this sport as aprofessional or an amateur? What differences can be identified? How important is handball in the lives of the affected persons?
This diploma thesis should deal with these and other topics in more detail. For the assurance of results, a questionnaire was drawn up on the basis of relevant literature, which had proven itself several times. This questionnaire, which is used for the survey of individual motives and identities in handball, was completed by a total of 126 persons between the ages of 14 and 36 years.
The results of the questionnaire make it clear that especially social factors like friends, acquaintances or school and university are crucial for the initial contact with the sport. The reasons why the players started playing handball or why they stayed with it are mainly of a social and success-oriented nature. It is important for the persons to be successful in and with the team and additionally to do something sporty as well as human
