    The Orphan Nuclear Receptor ERRĪ³ Regulates Hepatic CB1 Receptor-Mediated Fibroblast Growth Factor 21 Gene Expression

    BACKGROUND: Fibroblast growth factor 21 (FGF21), a stress inducible hepatokine, is synthesized in the liver and plays important roles in glucose and lipid metabolism. However, the mechanism of hepatic cannabinoid type 1 (CB1) receptor-mediated induction of FGF21 gene expression is largely unknown. RESULTS: Activation of the hepatic CB1 receptor by arachidonyl-2'-chloroethylamide (ACEA), a CB1 receptor selective agonist, significantly increased FGF21 gene expression. Overexpression of estrogen-related receptor (ERR) Ī³ increased FGF21 gene expression and secretion both in hepatocytes and mice, whereas knockdown of ERRĪ³ decreased ACEA-mediated FGF21 gene expression and secretion. Moreover, ERRĪ³, but not ERRĪ± and ERRĪ², induced FGF21 gene promoter activity. In addition, deletion and mutation analysis of the FGF21 promoter identified a putative ERRĪ³-binding motif (AGGTGC, a near-consensus response element). A chromatin immunoprecipitation assay revealed direct binding of ERRĪ³ to the FGF21 gene promoter. Finally, GSK5182, an ERRĪ³ inverse agonist, significantly inhibited hepatic CB1 receptor-mediated FGF21 gene expression and secretion. CONCLUSION: Based on our data, we conclude that ERRĪ³ plays a key role in hepatic CB1 receptor-mediated induction of FGF21 gene expression and secretion

    We examine the roles of massless Dirac spinon and spin singlet pair excitations on the phase transition in dāˆ’waved-wave superconductors. Although the massless spinon excitations in the presence of the spin singlet pair excitations do not alter the nature of the phase transition at T=0T = 0, that is, the XY universality class, they are seen to induce an additional attractive interaction potential between vortices, further stabilizing vortex-antivortex pairs at low temperature for lightly doped high TcT_c samples.Comment: 5 pages, 1 figur

    Impacts of Comorbidities on the Association between Arterial Stiffness and Obstructive Sleep Apnea in the Elderly

    BACKGROUND: Although the impact of obstructive sleep apnea (OSA) on cardiovascular risk is reasonably well established in middle-aged patients, debate persists whether OSA also increases this risk in the elderly. Arterial stiffness has been used as an early independent predictor of cardiovascular risk. STUDY OBJECTIVES: We sought to determine whether OSA had significant effects on the arterial stiffness in the elderly population and evaluate the impacts of comorbidities on the association between arterial stiffness and OSA. METHODS: We performed a cross-sectional study in a university hospital. Elderly participants (ā‰„60 yr) were invited to participate in our study between November 2010 and January 2013. OSA was diagnosed using gold standard polysomnography and arterial stiffness was assessed by brachial-ankle pulse wave velocity (baPWV) and cardio-ankle vascular index (CAVI) as well as by central systolic and diastolic blood pressure (cSBP and cDBP). High-sensitivity C-reactive protein (hs-CRP) level was also measured. RESULTS: We found no significant association between the severity of OSA and arterial stiffness-related parameters such as cSBP, cDBP, baPWV, CAVI, and hs-CRP. However, in patients without comorbid medical conditions or use of medications (n=101), we showed a modest association between OSA and arterial stiffness-related parameters and hs-CRP. CONCLUSION: We conclude that OSA is associated with increased arterial stiffness in otherwise healthy elderly population, although the association was obviated by comorbidities and medications perhaps due to ceiling effects

    Clinical and Radiological Evaluation After Chemical Synovectomy With Rifampicin in Hemophilic Arthropathy: Korean Experience With a 2-Week Interval Protocol

    Objective To assess the clinical outcome of chemical synovectomy with rifampicin in hemophilic arthropathy by using the World Federation of Hemophilia (WFH) scoring system and plain radiograph. Methods We performed rifampicin synovectomy (RS) on 30 joints of 28 hemophilic patients diagnosed as hemophilic arthropathy stage Iā€“III (based on Fernandez-Palazzi clinical classification). Clinical status (bleeding frequency, pain, joint physical status) and radiological staging were evaluated as parts of the WFH scoring system before and 1 year after RS. The patients were divided into two groups by the Arnold-Hilgartner scale of the initial X-ray as stage 3 or less for the low-stage group (n=17) and over 3 for the high-stage group (n=13). Results Total WFH joint physical scores were reduced after injection, and the number of bleeding episodes and pain showed especially significant improvement. For other subscores of the WFH joint physical score, only swelling, range of motion, and crepitus showed statistically significant improvement. According to the severity of the radiologic finding, the WFH joint physical score of both the low-stage and high-stage groups showed significant improvement. In the radiological aspect, the low-stage group, without joint space narrowing at the initial plain radiograph, showed no further aggravation after injection. However, in the high-stage group, radiology found aggravation regardless of the procedure. Conclusion It is suggested that chemical synovectomy with rifampicin may prevent hemarthrosis and improve clinical symptoms. Especially in the early stage of arthropathy without joint-space narrowing, it seems to have an additional benefit that delays radiological aggravation and preserves joint status

    An Antireflective Nanostructure Array Fabricated by Nanosilver Colloidal Lithography on a Silicon Substrate

    An alternative method is presented for fabricating an antireflective nanostructure array using nanosilver colloidal lithography. Spin coating was used to produce the multilayered silver nanoparticles, which grew by self-assembly and were transformed into randomly distributed nanosilver islands through the thermodynamic action of dewetting and Oswald ripening. The average size and coverage rate of the islands increased with concentration in the range of 50ā€“90 nm and 40ā€“65%, respectively. The nanosilver islands were critically affected by concentration and spin speed. The effects of these two parameters were investigated, after etching and wet removal of nanosilver residues. The reflection nearly disappeared in the ultraviolet wavelength range and was 17% of the reflection of a bare silicon wafer in the visible range

    SARS-CoV-2 vaccine effectiveness and clinical outcomes in hemodialysis patients: the NHIS-COVID-19 cohort study in South Korea

    BackgroundPatients undergoing hemodialysis (HD) have a high risk of novel coronavirus disease 2019 (COVID-19) and poor clinical outcomes. This study aimed to investigate severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) vaccine effectiveness against infection and deaths in the South Korean population undergoing HD.MethodsWe conducted a retrospective cohort study to compare the incidence of COVID-19 and post-diagnosis mortality between patients who were either never vaccinated or fully or partially vaccinated. The Korean nationwide COVID-19 registry and the Korean National Health Insurance Service databases were used. Adult patients without a history of COVID-19 were included between October 8, 2020, and December 31, 2021. The study outcomes were COVID-19 diagnosis, severe clinical COVID-19-related events, and post-diagnosis death.ResultsEighty-five thousand eighteen patients undergoing HD were included, of whom 69,601 were fully vaccinated, 2,213 were partially vaccinated and 13,204 were unvaccinated. Compared with the unvaccinated group, the risk of being diagnosed with COVID-19 in patients who were fully vaccinated decreased during the study period (adjusted odds ratio [aOR]ā€‰=ā€‰0.147; 95% confidence interval [CI]ā€‰=ā€‰0.135ā€“0.159). There were 1,140 (1.3%) patients diagnosed with COVID-19. After diagnosis, fully vaccinated patients were significantly less likely to die than unvaccinated patients (aORā€‰=ā€‰0.940; 95% CIā€‰=ā€‰0.901ā€“0.980) and to experience severe clinical events (aORā€‰=ā€‰0.952; 95% CIā€‰=ā€‰0.916ā€“0.988).ConclusionFull vaccination against COVID-19 was associated with a reduced risk of both infection and severe clinical outcomes in the South Korean population undergoing HD. These findings support the use of vaccination against SARS-CoV-2 among patients undergoing HD
