    Investigating the mechanism of a novel glycine-dependent riboswitch and a putative non-coding regulatory RNA in Streptococcus pyogenes

    We investigated gene expression regulation by a putative glycine(Gly) riboswitch located in the 5′-UTR of a SAF protein gene in the S.pyogenes. Gly-dependency was studied using a luciferase reporter gene system. Maximal reporter gene expression happened in the presence of low Gly concentrations. RT-qPCR showed that in the presence of Gly (≥1 mM), expression of the gene was downregulated. Growth in the presence of 0.1 mM Gly led to the production of a full-length transcript. We conclude that the Gly riboswitch in S. pyogenes represses gene expression in the presence of high Gly concentrations

    Evaluating knowledge, attitude and practice of intensive care unit nurses in administering medications via enteral tubes

    Introduction : The purpose of the study was to evaluate Intensive Care Unit nurses’ knowledge, attitude, self-reported practice, and observed practice regarding medication administration via enteral feeding tube.Method:in this cross sectional study, a questionnaire and a checklist was developed for evaluating the nurses' knowledge, attitude, self-reported practice and real practice regarding medication administration via enteral feeding tube. Mean score of knowledge, and the percentage of different domains of attitude and practice was reported.Results: 53 nurses were included in this study. The mean number of medications which were prescribed for each patient was 15.8±4.5. From these medications, 5.5±2.8 drugs were administered via enteral feeding tube. There were few nurses (5%) with proper knowledge regarding correct dosage forms selection. About half of the nurses had proper knowledge regarding tube flushing and drug dilution,however according to direct researcher’s observation, 11% of nurses flushed the tubes before medication administration.Conclusion:The current study showed the disparity between the nurses' knowledge, self-reported practice, and proper performance in drug administration through enteral feeding tube. The most important problems in knowledge and attitude of nurses included crushing the dosage forms and in real practice in diluting medications and tube flushes domain.  </p

    Assessment of Radiation Dose to the Lens of the Eye and Thyroid of Patients Undergoing Head and Neck Computed Tomography at Five Hospitals in Mashhad, Iran

    Introduction: In recent years, the number of computed tomography (CT) scans, which is a high-dose technique, has increased significantly. Head and neck CT is performed frequently and thyroid, particularly in children, has always been considered a sensitive organ. In recent years, radiobiologists and health physicists have been more concerned about the safety of lenses of the eyes, as cataract is no longer considered a deterministic effect. Material and Methods: In the present study, incurred doses to the thyroid and lens of the eye of 140 patients who underwent common head and neck CT at five hospitals were measured by thermoluminescent dosimeters (TLD-100). The patients were divided into two age groups of pediatrics and adults. TLD chips were placed on the patient’s skin surface. For each patient, scan parameters, sex and age were recorded. Exposed TLDs were read by a manual TLD reader. Results: The verage absorbed dose of the thyroid, as well as the lenses of the left and right eyes were 5.89±1.74, 15.84±2.81 and 16.25±2.57, respectively, for the pediatric patients and 5.00±1.17, 17.64±1.69 and 24.41±1.89 for adults. Patient-specific organ doses were influenced by the scanned region, scan protocol and patient's age. Conclusion: In the present study, the mean eye dose was much lower than the 500 mGy threshold recommended by International Commission on Radiological Protection (ICRP) for lens of the eye damage, thus, it appears to be clinically safe. While CT scan remains a crucial tool, further dose reduction can be achieved by controlling different factors affecting patient doses

    Global systematic review and meta-analysis of health-related quality of life in Behcet’s patients

    Background: Behcet’s disease (BD) is a chronic fatal illness with a relapsing remitting nature and significant organ-threatening morbidity and mortality. The aim of this research was to examine studies which were conducted on investigation of prevalence of quality of life among patients with Behcet’s disease. Methods: A total of 13 articles were extracted from four main databases including PubMed, EMBASE, Scopus, and Web of Science from the onset of 2000 to January 2021. All studies published in English with the purpose of examining quality of life (QOL) among patients with BD or investigating its main determinants were included. Results: Totally, 1137 BD patients participated in 13 studies. Based on random effect analysis, the total score of physical health-related QOL was 46.7 (95% CI=41.26 to 52.13) and the total score of mental health-related QOL was 49.01 (95% CI=43.83 to 54.18) representing a moderate level of QOL among BD patients. Furthermore, weighted effect size analyses showed a significant correlation between QOL and variables such as patients’ age, gender, disease duration and depression (pvalue: 0.00). Conclusion: As the symptoms of BD worsen over time, patients confront with more severe body pain, mobility restrictions, and difficulties in chewing, eating, speaking and swallowing which negatively affect social interactions of patients and reduce their QOL. Furthermore, depression was proved to act as a deteriorating factor for Health-Related Quality of Life (HRQOL) among BD patients. Thus, patients need to be psychologically supported by a specialized team and be informed during the course of treatment to gain useful information about the disease, treatment approaches and coping strategies

    The analgesic effect of Curcuma Longa rhizome methanol extract in male rat

    Background: There are several reports on the therapeutic effects of curcuma longa in traditional medicine such as anti inflammatory,  antinocieptive , antioxidant    ,anti carcinogenic and hepatoprotective effects. Therefore, In this study we investigated the the analgesic effect of this Plant in experimental models of acute and chronic Pain as compare to Sodium Salicylate. Methods: Curcuma Longa extract were prepared using percolation method. Different dose of root extract (100/200/300 mg/ kg i. p ) and sodium salicylate (300 mg/kg i. P) were given. The analgesic activity was assessed in experimental models of acute pain (tail flick) and chronic pain (used formalin test). Findings: All dose of extract and sodium salicylate had analgesic effect than control group in tail flick test& second of phase formalin test (P<0.05). Conclusion: It seems that Curcuma Longa induces its inhibitory effect on acute and chronic Pain

    Regulatory RNAs play important roles in the control of bacterial gene expression. In this study, we investigated gene expression regulation by a putative glycine riboswitch located in the 5′-untranslated region of a sodium:alanine symporter family (SAF) protein gene in the group A Streptococcus pyogenes serotype M49 strain 591. Glycine-dependent gene expression mediated by riboswitch activity was studied using a luciferase reporter gene system. Maximal reporter gene expression was observed in the absence of glycine and in the presence of low glycine concentrations. Differences in glycine-dependent gene expression were not based on differential promoter activity. Expression of the SAF protein gene and the downstream putative cation efflux protein gene was investigated in wild-type bacteria by RT-qPCR transcript analyses. During growth in the presence of glycine (≥1 mM), expression of the genes were downregulated. Northern blot analyses revealed premature transcription termination in the presence of high glycine concentrations. Growth in the presence of 0.1 mM glycine led to the production of a full-length transcript. Furthermore, stability of the SAF protein gene transcript was drastically reduced in the presence of glycine. We conclude that the putative glycine riboswitch in S. pyogenes serotype M49 strain 591 represses expression of the SAF protein gene and the downstream putative cation efflux protein gene in the presence of high glycine concentrations. Sequence and secondary structure comparisons indicated that the streptococcal riboswitch belongs to the class of tandem aptamer glycine riboswitches
