Regeneration Rates of Dendrobium Bobby Messina Plbs with Ascorbic Acid Using PVS2 Vitrification
Cryopreservation techniques using PVS2 vitrification was applied on PLBs of Dendrobium Bobby Messina, with survival monitored through observations of growth rate and the 2,3,5-triphenyltetrazolium chloride (TTC) analyses. The parameters optimized were PLBs size, preculture concentration, preculture duration, PVS2 incubation temperature and duration. The optimized parameters obtained were 3-4mm of PLBs precultured in 0.2M sucrose for 1 day, treated with a mixture of 2M glycerol and 0.4M sucrose supplemented with half strength liquid MS media at 25°C for 20 minutes and subsequently dehydrated with plant vitrification solution 2 (PVS2) at 0°C for 20 minutes prior storage in liquid nitrogen. Following rapid warming in a water bath at 40°C for 90 seconds, PLBs were washed with a half strength liquid MS media supplemented with 1.2M sucrose. Subsequently, PLBs were cultured on half strength semi-solid MS media supplemented with 2% (w/v) sucrose in the absence of growth regulator. The optimized vitrification method was successful in preserving this orchid as it produced growth recovery in cryopreserved PLBs up to 40%. Ascorbic acid was added in the media to evaluate the regeneration process of cryopreserved PLBs. However, growth recovery rate was only 10% at 0.6mM ascorbic acid. RAPD analysis using 6 primers indicated that cryopreserved and non-cryopreserved PLBs from vitrification method were genetically faithful to the mother plant. However, 3 primers showed polymorphism and 1 primers indicated partial polymorphism between the cryopreserved and non-cryopreserved PLBs in comparative to the mother plant
Challenges of industrial wastewater treatment: utilizing Membrane bioreactors (MBRs) in conjunction with artificial intelligence (AI) technology
\ua9 2024 The Author(s). Published by Informa UK Limited, trading as Taylor & Francis Group. In the past, decisions on wastewater treatment methods have predominantly rested on expert opinions, utilizing the Delphi method. Yet, with an anticipated increase in diversification and customization, especially in the “small-batch and diverse” market over the next decade, addressing the formulation and execution of wastewater treatment for these non-traditional production processes will present substantial challenges. Relying solely on Delphi experts’ decision-making within a short and time-constrained production planning window is expected to prove inadequate. Predominantly relies on the authors’ over 15 years of industry experience in wastewater treatment, this perspective paper proposes an inventive solution that integrates Membrane Bioreactors (MBRs) with Artificial Intelligence (AI) applications. This approach signifies a more advanced method for industrial wastewater treatment compared to conventional methods, with the intention of garnering increased interest for future research endeavors
Quark deconfinement phase transition for improved quark mass density-dependent model
By using the finite temperature quantum field theory, we calculate the finite
temperature effective potential and extend the improved quark mass
density-dependent model to finite temperature. It is shown that this model can
not only describe the saturation properties of nuclear matter, but also explain
the quark deconfinement phase transition successfully. The critical temperature
is given and the effect of - meson is addressed.Comment: 18 pages, 7 figure
Alteration of tumour response to radiation by interleukin-2 gene transfer
We have previously shown that BALB/c-derived EMT6 mammary tumours transfected with interleukin (IL)-2 have decreased hypoxia compared to parental tumours, due to increased vascularization. Since hypoxia is a critical factor in the response of tumours to radiation treatment, we compared the radiation response of IL-2-transfected tumours to that of parental EMT6 tumours. Because the IL-2 tumours have an altered host cell composition, which could affect the interpretation of radiation sensitivity as measured by clonogenic cells, we employed flow cytometric analysis to determine the proportion of tumour cells vs host cells in each tumour type. Using this approach, we were able to correct the plating efficiency based on the number of actual tumour cells derived from tumours, making the comparison of the two types of tumours possible. We also excluded the possibility that cytotoxic T-cells present in EMT6/IL-2 tumours could influence the outcome of the clonogenic cell survival assay, by demonstrating that the plating efficiency of cells derived from EMT6/IL-2 tumours remained unchanged after depletion of Thy-1+cells. The in vivo radiation response results demonstrated that IL-2-transfected tumours were more sensitive to radiation than parental EMT6 tumours. The hypoxic fraction of the EMT6/IL-2 tumours growing in vivo was markedly decreased relative to parental EMT6 tumours thus the increased sensitivity results from the increased vascularity we have previously observed in these tumours. These results indicate the potential therapeutic benefit of combining radiation and immunotherapy in the treatment of tumours. © 2000 Cancer Research Campaig
Geometry of the extreme Kerr black holes
Geometrical properties of the extreme Kerr black holes in the topological
sectors of nonextreme and extreme configurations are studied. We find that the
Euler characteristic plays an essential role to distinguish these two kinds of
extreme black holes. The relationship between the geometrical properties and
the intrinsic thermodynamics are investigated.Comment: Latex version, 10 page
Turbulent mixing of particles under tidal bores: An experimental analysis
A tidal bore develops in an estuary when the tidal range exceeds 4.5-6 m and the estuarine bathymetry amplifies the tidal wave. The bore is an abrupt rise in water depth associated with a discontinuity in velocity and pressure fields at the front. Herein the free-surface properties and the turbulent mixing of light-weight particles were investigated during the passage of tidal bores. The free-surface properties were recorded using a non-intrusive technique, while particle tracking was performed under undular and breaking bores. A basic result was the identification of a broad spectrum of particle trajectories, linked with the existence of large-scale vortical structures. These turbulent structures were responsible for the vertical water mixing as a tidal bore propagates upstream in an estuary. The large-scale eddies were also responsible for the rapid longitudinal dispersion of particulates, such as fish eggs, with some form of preferential motion, depending upon the particle's vertical elevation
Sigma-2 receptors as a biomarker of proliferation in solid tumours
Over the past several years, our group has provided considerable evidence that the expression of sigma-2 (σ2) receptors may serve as a biomarker of tumour cell proliferation. In these in vitro studies, σ2receptors were expressed 8–10 times more in proliferative (P) tumour cells than in quiescent (Q) tumour cells, and the extent and kinetics of their expression were independent of a number of biological, physiological and environmental factors often found in solid tumours. Moreover, the expression of σ2receptors followed both the population growth kinetics when Q-cells were recruited into the P-cell compartment and the proliferative status of human breast tumour cells treated with cytostatic concentrations of tamoxifen. However, these in vitro studies may or may not be indicative of what might occur in solid tumours. In the present study, the σ2receptor P:Q ratio was determined for the cells from subcutaneous 66 (diploid) and 67 (aneuploid) tumours grown in female nude mice. The σ2receptor P:Q ratio of the 66 tumours was 10.6 compared to the σ2receptor P:Q ratio of 9.5 measured for the 66 tissue culture model. The σ2receptor P:Q ratio of the 67 tumours was 4.5 compared to the σ2receptor P:Q ratio of ≈ 8 measured for the 67 tissue culture model. The agreement between the solid tumour and tissue culture data indicates that: (1) the expression of σ2receptors may be a reliable biomarker of the proliferative status of solid tumours and (2) radioligands with both high affinity and high selectivity for σ2receptors may have the potential to non-invasively assess the proliferative status of human solid tumours using imaging techniques such as positron emission tomography or single-photon emission computerized tomography. © 2000 Cancer Research Campaig
Automorphism Modular Invariants of Current Algebras
We consider those two-dimensional rational conformal field theories (RCFTs)
whose chiral algebras, when maximally extended, are isomorphic to the current
algebra formed from some affine non-twisted Kac--Moody algebra at fixed level.
In this case the partition function is specified by an automorphism of the
fusion ring and corresponding symmetry of the Kac--Peterson modular matrices.
We classify all such partition functions when the underlying finite-dimensional
Lie algebra is simple. This gives all possible spectra for this class of RCFTs.
While accomplishing this, we also find the primary fields with second smallest
quantum dimension.Comment: 32 pages, plain Te
