    Nest survival of Tricolored Blackbirds in California\u27s Central Valley

    The Tricolored Blackbird (Agelaius tricolor), almost entirely restricted to California, USA, has recently been proposed for listing under the U.S. Endangered Species Act. Tricolored Blackbirds historically nested in wetlands, but a large proportion of the population now nests in agricultural grain fields where the crop is ready to harvest before the young have fledged. Since 1991, federal agencies have paid farmers to delay harvesting in an effort to increase nesting productivity. However, the relative nesting success of Tricolored Blackbirds breeding in agricultural fields versus wetlands is unknown. Our objectives were to estimate daily survival rate (DSR) of nests, identify habitat covariates that influence nest survival, and estimate the number of young produced per nest. During 2011–2012, we monitored 1,323 Tricolored Blackbird nests in 12 colonies using small temperature data loggers. We modeled DSR using Program RMark with combinations of the following variables: site, habitat type, nest initiation date, nest height, water depth, nest density, colony population size, year, and the proportion of nearby nests that failed. Nest survival varied greatly (range: 0.024–0.719) but was not explained by habitat type. Nest height and nest density were positively associated with DSR. DSR was lowest midway through the breeding season and declined with colony population size. Number of young produced per nest varied by site, was lowest in intermediate-sized colonies of 1,000–5,000 birds, and was highest in 2011. DSR and number of young fledged per nest were similar in agricultural fields and in wetlands. Our results suggest that Tricolored Blackbirds benefit from policies that allow them to complete their nesting cycle in agricultural fields

    Stress-Regulated Translational Attenuation Adapts Mitochondrial Protein Import through Tim17A Degradation

    SummaryStress-regulated signaling pathways protect mitochondrial proteostasis and function from pathologic insults. Despite the importance of stress-regulated signaling pathways in mitochondrial proteome maintenance, the molecular mechanisms by which these pathways maintain mitochondrial proteostasis remain largely unknown. We identify Tim17A as a stress-regulated subunit of the translocase of the inner membrane 23 (TIM23) mitochondrial protein import complex. We show that Tim17A protein levels are decreased downstream of stress-regulated translational attenuation induced by eukaryotic initiation factor 2α (eIF2α) phosphorylation through a mechanism dependent on the mitochondrial protease YME1L. Furthermore, we demonstrate that decreasing Tim17A attenuates TIM23-dependent protein import, promotes the induction of mitochondrial unfolded protein response (UPR)-associated proteostasis genes, and confers stress resistance in C. elegans and mammalian cells. Thus, our results indicate that Tim17A degradation is a stress-responsive mechanism by which cells adapt mitochondrial protein import efficiency and promote mitochondrial proteostasis in response to the numerous pathologic insults that induce stress-regulated translation attenuation

    Does fire influence the landscape-scale distribution of an invasive mesopredator?

    Predation and fire shape the structure and function of ecosystems globally. However, studies exploring interactions between these two processes are rare, especially at large spatial scales. This knowledge gap is significant not only for ecological theory, but also in an applied context, because it limits the ability of landscape managers to predict the outcomes of manipulating fire and predators. We examined the influence of fire on the occurrence of an introduced and widespread mesopredator, the red fox (Vulpes vulpes), in semi-arid Australia. We used two extensive and complimentary datasets collected at two spatial scales. At the landscape-scale, we surveyed red foxes using sand-plots within 28 study landscapes - which incorporated variation in the diversity and proportional extent of fire-age classes - located across a 104 000 km2 study area. At the site-scale, we surveyed red foxes using camera traps at 108 sites stratified along a century-long post-fire chronosequence (0-105 years) within a 6630 km2 study area. Red foxes were widespread both at the landscape and site-scale. Fire did not influence fox distribution at either spatial scale, nor did other environmental variables that we measured. Our results show that red foxes exploit a broad range of environmental conditions within semi-arid Australia. The presence of red foxes throughout much of the landscape is likely to have significant implications for native fauna, particularly in recently burnt habitats where reduced cover may increase prey species\u27 predation risk

    Refining pathological evaluation of neoadjuvant therapy for adenocarcinoma of the esophagus

    AIM: To assess tumour regression grade (TRG) and lymph node downstaging to help define patients who benefit from neoadjuvant chemotherapy.METHODS: Two hundred and eighteen consecutive patients with adenocarcinoma of the esophagus or gastro-esophageal junction treated with surgery alone or neoadjuvant chemotherapy and surgery between 2005 and 2011 at a single institution were reviewed. Triplet neoadjuvant chemotherapy consisting of platinum, fluoropyrimidine and anthracycline was considered for operable patients (World Health Organization performance status ? 2) with clinical stage T2-4 N0-1. Response to neoadjuvant chemotherapy (NAC) was assessed using TRG, as described by Mandard et al. In addition lymph node downstaging was also assessed. Lymph node downstaging was defined by cN1 at diagnosis: assessed radiologically (computed tomography, positron emission tomography, endoscopic ultrasonography), then pathologically recorded as N0 after surgery; ypN0 if NAC given prior to surgery, or pN0 if surgery alone. Patients were followed up for 5 years post surgery. Recurrence was defined radiologically, with or without pathological confirmation. An association was examined between t TRG and lymph node downstaging with disease free survival (DFS) and a comprehensive range of clinicopathological characteristics.RESULTS: Two hundred and eighteen patients underwent esophageal resection during the study interval with a mean follow up of 3 years (median follow up: 2.552, 95%CI: 2.022-3.081). There was a 1.8% (n = 4) inpatient mortality rate. One hundred and thirty-six (62.4%) patients received NAC, with 74.3% (n = 101) of patients demonstrating some signs of pathological tumour regression (TRG 1-4) and 5.9% (n = 8) having a complete pathological response. Forty four point one percent (n = 60) had downstaging of their nodal disease (cN1 to ypN0), compared to only 15.9% (n = 13) that underwent surgery alone (pre-operatively overstaged: cN1 to pN0), (P < 0.0001). Response to NAC was associated with significantly increased DFS (mean DFS; TRG 1-2: 5.1 years, 95%CI: 4.6-5.6 vs TRG 3-5: 2.8 years, 95%CI: 2.2-3.3, P < 0.0001). Nodal down-staging conferred a significant DFS advantage for those patients with a poor primary tumour response to NAC (median DFS; TRG 3-5 and nodal down-staging: 5.533 years, 95%CI: 3.558-7.531 vs TRG 3-5 and no nodal down-staging: 1.114 years, 95%CI: 0.961-1.267, P < 0.0001).CONCLUSION: Response to NAC in the primary tumour and in the lymph nodes are both independently associated with improved DFS

    Expression of miR-145 and miR-449 in U87 Glioblastoma Cells

    MicroRNAs are known to play a critical oncogenic role in glioblastoma. Several miRNAs have been shown to play roles in growth and cell cycle control in glioblastoma, and have potential as diagnostic markers or as therapeutic targets. miRNA-145 is overexpressed in glioblastoma and is thought to downregulate srGAP1 (SLIT-ROBO Rho GTPase-activating protein1), which promotes an invasive phenotype. This is in contrast to miRNA-145’s proposed tumor-supressive role in many other cancers (1). miRNA-449 is thought to be a tumor suppressor that interrupts the cell cycle and induces apoptosis via suppression of E2F1, CCND1, and GPR158. Therefore it is highly downregulated in many tumor cells (2,3). Both miRNAs-145 and -449 are the topic of continued inquiry in understanding their expression and their molecular targets in glioma cells. Here we attempt to establish the baseline expression of miRNA-145 and miRNA-449 in the U87 cell line using RT-PCR. Weekly passaging of cells was carried out, followed by RNA isolation and RT-PCR. Baseline average concentrations were established for miRNA-145 and miRNA-449 using a set of serial dilutions and a standard curve. Further experimentation will be required to establish a baseline concentration for these miRNAs in healthy glial cells

    Effects of time since fire on birds : how informative are generalized fire response curves for conservation management?

    Fire is both a widespread natural disturbance that affects the distribution of species and a tool that can be used to manage habitats for species. Knowledge of temporal changes in the occurrence of species after fire is essential for conservation management in fire-prone environments. Two key issues are: whether postfire responses of species are idiosyncratic or if multiple species show a limited number of similar responses; and whether such responses to time since fire can predict the occurrence of species across broad spatial scales. We examined the response of bird species to time since fire in semiarid shrubland in southeastern Australia using data from surveys at 499 sites representing a 100-year chronosequence. We used nonlinear regression to model the probability of occurrence of 30 species with time since fire in two vegetation types, and compared species\u27 responses with generalized response shapes from the literature. The occurrence of 16 species was significantly influenced by time since fire: they displayed six main responses consistent with generalized response shapes. Of these 16 species, 15 occurred more frequently in mid- or later-successional vegetation (&gt;20 years since fire), and only one species occurred more often in early succession (&lt;5 years since fire). The models had reasonable predictive ability for eight species, some predictive ability for seven species, and were little better than random for one species. Bird species displayed a limited range of responses to time since fire; thus a small set of fire ages should allow the provision of habitat for most species. Postfire successional changes extend for decades and management of the age class distribution of vegetation will need to reflect this timescale. Response curves revealed important seral stages for species and highlighted the importance of mid- to late-successional vegetation (&gt;20 years). Although time since fire clearly influences the distribution of numerous bird species, predictive models of the spatial distribution of species in fire-prone landscapes need to incorporate other factors in addition to time since fire.<br /

    Enhanced Aromatic Sequons Increase Oligosaccharyltransferase Glycosylation Efficiency and Glycan Homogeneity

    SummaryN-Glycosylation plays an important role in protein folding and function. Previous studies demonstrate that a phenylalanine residue introduced at the n-2 position relative to an Asn-Xxx-Thr/Ser N-glycosylation sequon increases the glycan occupancy of the sequon in insect cells. Here, we show that any aromatic residue at n-2 increases glycan occupancy in human cells and that this effect is dependent upon oligosaccharyltransferase substrate preferences rather than differences in other cellular processing events such as degradation or trafficking. Moreover, aromatic residues at n-2 alter glycan processing in the Golgi, producing proteins with less complex N-glycan structures. These results demonstrate that manipulating the sequence space surrounding N-glycosylation sequons is useful both for controlling glycosylation efficiency, thus enhancing glycan occupancy, and for influencing the N-glycan structures produced
