    A comparison study for two fuzzy-based systems: improving reliability and security of JXTA-overlay P2P platform

    This is a copy of the author's final draft version of an article published in the journal Soft computing.The reliability of peers is very important for safe communication in peer-to-peer (P2P) systems. The reliability of a peer can be evaluated based on the reputation and interactions with other peers to provide different services. However, for deciding the peer reliability there are needed many parameters, which make the problem NP-hard. In this paper, we present two fuzzy-based systems (called FBRS1 and FBRS2) to improve the reliability of JXTA-overlay P2P platform. In FBRS1, we considered three input parameters: number of interactions (NI), security (S), packet loss (PL) to decide the peer reliability (PR). In FBRS2, we considered four input parameters: NI, S, PL and local score to decide the PR. We compare the proposed systems by computer simulations. Comparing the complexity of FBRS1 and FBRS2, the FBRS2 is more complex than FBRS1. For simulations, we used ns-3. We compare the performance for two architectures considering transmission rate and OLSR protocol. The simulation results show that for transmission rate 600 and 1200 [kbps], the throughput of Hybrid WMN is higher than I/B WMN. For transmission rate 600 and 1200 [kbps], the delay and jitter of Hybrid WMN is lower than I/B WMN.     Novel analgesics targeting brain-derived neurotrophic factor for neuropathic pain

     Brain-derived neurotrophic factor (BDNF) is necessary for the development, growth, and maintenance of nerve cells. BDNF is expressed in the dorsal root ganglion (DRG); binds to the Tropomyosin receptor kinasa B (TrkB) receptor, which has a tyrosine kinase domain, in the spinal cord; and plays an important role as a pain modulator. BDNF expression is increased in various types of pain, including acute pain, neuropathic pain, and cancer pain. Activation of the BDNF–TrkB pathway transmits pain information. In order to inhibit the BDNF–TrkB pathway, by sequestering BDNF, we constructed a cDNA expression plasmid encoding the extracellular region of rat TrkB fused to enhanced green fluorescent protein (EGFP). When the expression plasmid vector was administered to rat models of neuropathic pain, induced by spinal nerve ligation, statistically significant relief of pain was observed in terms of a 50% paw-withdrawal threshold using the von Frey test. The expression of TrkB-EGFP mRNA was detected in L5 lumbar vertebral nerves by quantitative reverse transcriptase polymerase chain reaction. To verify the pain-suppressive effect of the expression vector, truncated TrkB protein, without EGFP, was purified, and administered to pain model rats. A statistically significant suppressive effect of the truncated TrkB protein on neuropathic pain was observed 2 days after administration. The pain-suppressive effect of the truncated TrkB protein was more effective than that of the TrkB-Fc chimera protein and lasted longer than that of the TrkB antagonist ANA-12. Our results suggested that the truncated TrkB cDNA expression vector and truncated TrkB protein could be used as molecular targeted drugs in patients with neuropathic pain

    Improving reliability of JXTA-overlay P2P platform: a comparison study for two fuzzy-based systems

Thus, it is important for each peer to have some number of neighbor peers. Moreover, it is more significant to discuss if each peer has reliable neighbor peers. In reality, each peer might be faulty or might send obsolete, even incorrect information to the other peers. We have implemented a P2P platform called JXTA-Overlay, which defines a set of protocols that standardize how different devices may communicate and collaborate among them. It abstracts a new layer on the top of JXTA through a set of primitive operations and services that are commonly used in JXTA-based applications and provides a set of primitives that can be used by other applications, which will be built on top of the overlay, with complete independence. JXTA-Overlay provides a set of basic functionalities, primitives, intended to be as complete as possible to satisfy the needs of most JXTA-based applications. In this paper, we present two fuzzy-based systems (called FRS1 and FRS2) to improve the reliability of JXTA-Overlay P2P platform. We make a comparison study between the fuzzy-based reliability systems. Comparing the complexity of FRS1 and FRS2, the FRS2 is more complex than FRS1.     Effects of ytterbium laser surface treatment on the bonding of two resin cements to zirconia.

    Monolithic zirconia crowns bonded to zirconia abutments have become more commonly used in the construction of cement-retained implant superstructures. The present study aimed to examine the effects of laser surface treatments on the bond strength of two resin cements to zirconia. Three types of surfaces were examined: untreated, alumina blasted, and ytterbium laser treated; and two types of resin cements: 4-META/MMA-TBB resin cement and composite resin cement. Half of the specimens were subjected to a thermocycling process. Subsequently, a shear bond test was carried out. In addition, surface roughness was measured for each surface type. The results showed that laser treatment increased zirconia surface roughness and that laser treatment significantly increased shear bond strength after the thermocycling of both cement types compared to no treatment. Our experimental results suggested that ytterbium laser surface treatment of zirconia increased the bond strength of resin cements.福岡歯科大学2021年

    Theranostic Agent Combining Fullerene Nanocrystals and Gold Nanoparticles for Photoacoustic Imaging and Photothermal Therapy

    Developing photoactivatable theranostic platforms with integrated functionalities of biocompatibility, targeting, imaging contrast, and therapy is a promising approach for cancer diagnosis and therapy. Here, we report a theranostic agent based on a hybrid nanoparticle comprising fullerene nanocrystals and gold nanoparticles (FGNPs) for photoacoustic imaging and photothermal therapy. Compared to gold nanoparticles and fullerene crystals, FGNPs exhibited stronger photoacoustic signals and photothermal heating characteristics by irradiating light with an optimal wavelength. Our studies demonstrated that FGNPs could kill cancer cells due to their photothermal heating characteristics in vitro. Moreover, FGNPs that are accumulated in tumor tissue via the enhanced permeation and retention effect can visualize tumor tissue due to their photoacoustic signal in tumor xenograft model mice.     Histone deacetylase inhibitor (SAHA) and repression of EZH2 synergistically inhibit proliferation of gallbladder carcinoma

    医薬保健研究域医学系Polycomb group protein EZH2, frequently overexpressed in malignant tumors, is the catalytic subunit of polycomb repressive complex 2 (PRC2). PRC2 interacts with HDACs in transcriptional silencing and relates to tumor suppressor loss. We examined the expression of HDAC isoforms (HDAC 1 and 2) and EZH2, and evaluated the possible use of HDAC inhibitor suberoylanilide hydroxamic acid (SAHA) and EZH2 repressor for gallbladder carcinoma. We used 48 surgically resected gallbladders and cultures of human gallbladder epithelial cells (HGECs), gallbladder carcinoma (TGBC2TKB), and cholangiocarcinoma (HuCCT-1 and TFK-1) cell lines for examination. Immunohistochemically, EZH2 was overexpressed in gallbladder carcinoma, especially poorly differentiated carcinoma, but not in normal epithelium. In contrast, HDAC1/2 were expressed in both carcinoma and normal epithelium in vivo. This pattern was verified in cultured cells; EZH2 was highly expressed only in TGBC2TKB, whereas HDAC1/2 were expressed in HGECs and TGBC2TKB. Interestingly, SAHA treatment caused significant cell number decline in three carcinoma cells, and this effect was synergized with EZH2 siRNA treatment; however, HGECs were resistant to SAHA. In TGBC2TKB cells, the expression of EZH2 and HDAC1/2 were decreased by SAHA treatment, and p16INK4a, E-cadherin, and p21were simultaneously activated; however, no such findings were obtained in HGECs, suggesting that the effect of SAHA depends on the EZH2-mediated tumor suppressor loss. In conclusion, this study suggests a possible mechanism by which carcinoma cells but not normal cells are sensitive to SAHA and indicates the efficacy of this new anticancer agent in combination with EZH2 repression in gallbladder carcinoma. © 2009 Japanese Cancer Association

    Berberine improved experimental chronic colitis by regulating interferon-gamma- and IL-17A-producing lamina propria CD4(+) T cells through AMPK activation

    The herbal medicine berberine (BBR) has been recently shown to be an AMP-activated protein kinase (AMPK) productive activator with various properties that induce anti-inflammatory responses. We investigated the effects of BBR on the mechanisms of mucosal CD4+T cell activation in vitro and on the inflammatory responses in T cell transfer mouse models of inflammatory bowel disease (IBD). We examined the favorable effects of BBR in vitro, using lamina propria (LP) CD4+ T cells in T cell transfer IBD models in which SCID mice had been injected with CD4+CD45RBhigh T cells. BBR suppressed the frequency of IFN-γ- and Il-17A-producing LP CD4+ T cells. This effect was found to be regulated by AMPK activation possibly induced by oxidative phosphorylation inhibition. We then examined the effects of BBR on the same IBD models in vivo. BBR-fed mice showed AMPK activation in the LPCD4+ T cells and an improvement of colitis. Our study newly showed that the BBR-induced AMPK activation of mucosal CD4+ T cells resulted in an improvement of IBD and underscored the importance of AMPK activity in colonic inflammation

    Overexpression of TFAM or Twinkle Increases mtDNA Copy Number and Facilitates Cardioprotection Associated with Limited Mitochondrial Oxidative Stress

    Background Mitochondrial DNA (mtDNA) copy number decreases in animal and human heart failure (HF), yet its role in cardiomyocytes remains to be elucidated. Thus, we investigated the cardioprotective function of increased mtDNA copy number resulting from the overexpression of human transcription factor A of mitochondria (TFAM) or Twinkle helicase in volume overload (VO)-induced HF. Methods and Results Two strains of transgenic (TG) mice, one overexpressing TFAM and the other overexpressing Twinkle helicase, exhibit an approximately 2-fold equivalent increase in mtDNA copy number in heart. These TG mice display similar attenuations in eccentric hypertrophy and improved cardiac function compared to wild-type (WT) mice without any deterioration of mitochondrial enzymatic activities in response to VO, which was accompanied by a reduction in matrix-metalloproteinase (MMP) activity and reactive oxygen species after 8 weeks of VO. Moreover, acute VO-induced MMP-2 and MMP-9 upregulation was also suppressed at 24 h in both TG mice. In isolated rat cardiomyocytes, mitochondrial reactive oxygen species (mitoROS) upregulated MMP-2 and MMP-9 expression, and human TFAM (hTFAM) overexpression suppressed mitoROS and their upregulation. Additionally, mitoROS were equally suppressed in H9c2 rat cardiomyoblasts that overexpress hTFAM or rat Twinkle, both of which exhibit increased mtDNA copy number. Furthermore, mitoROS and mitochondrial protein oxidation from both TG mice were suppressed compared to WT mice. Conclusions The overexpression of TFAM or Twinkle results in increased mtDNA copy number and facilitates cardioprotection associated with limited mitochondrial oxidative stress. Our findings suggest that increasing mtDNA copy number could be a useful therapeutic strategy to target mitoROS in HF.Peer reviewe
