    Effects of flywheel training on strength-related variables in female populations. A systematic review

    This study aimed to evaluate the effect of flywheel training on female populations, report practical recommendations for practitioners based on the currently available evidence, underline the limitations of current literature, and establish future research directions. Studies were searched through the electronic databases (PubMed, SPORTDiscus, and Web of Science) following the preferred reporting items for systematic reviews and meta-analysis statement guidelines. The methodological quality of the seven studies included in this review ranged from 10 to 19 points (good to excellent), with an average score of 14-points (good). These studies were carried out between 2004 and 2019 and comprised a total of 100 female participants. The training duration ranged from 5 weeks to 24 weeks, with volume ranging from 1 to 4 sets and 7 to 12 repetitions, and frequency ranged from 1 to 3 times a week. The contemporary literature suggests that flywheel training is a safe and time-effective strategy to enhance physical outcomes with young and elderly females. With this information, practitioners may be inclined to prescribe flywheel training as an effective countermeasure for injuries or falls and as potent stimulus for physical enhancement

    Myosin-V Opposes Microtubule-Based Cargo Transport and Drives Directional Motility on Cortical Actin

    SummaryIntracellular transport is driven by motor proteins that either use microtubules or actin filaments as their tracks [1], but the interplay between these transport pathways is poorly understood [2–4]. Whereas many microtubule-based motors are known to drive long-range transport, several actin-based motors have been proposed to function predominantly in cargo tethering [4–6]. How these opposing activities are integrated on cargoes that contain both types of motors is unknown. Here we use inducible intracellular transport assays to show that acute recruitment of myosin-V to kinesin-propelled cargo reduces their motility near the cell periphery and enhances their localization at the actin-rich cell cortex. Myosin-V arrests rapid microtubule-based transport without the need for regulated auto- or other inhibition of kinesin motors. In addition, myosin-V, despite being an ineffective long-range transporter, can drive slow, medium-range (1–5 μm), point-to-point transport in cortical cell regions. Altogether, these data support a model in which myosin-V establishes local cortical delivery of kinesin-bound cargos through a combination of tethering and active transport

De directie Platteland van het ministerie van LNV heeft bij het publiceren van het meerjarenprogramma van Agenda Vitaal Platteland geen digitale viewer gepubliceerd omdat men bang was voor verkeerde interpretatie van de digitale kaarten. In dit project is gekeken naar methoden en cartografische oplossingen om voortaan zonder angst voor misinterpretaties digitale nationale beleidskaarten te kunnen verspreiden. De oplossing is gezocht in het opstellen van een handreiking, zodat kaarten ook daadwerkelijk weergeven wat er bedoeld is door de maker

    The transcription factor GATA6 is essential for branching morphogenesis and epithelial cell differentiation during fetal pulmonary development

    Recent loss-of-function studies in mice show that the transcription factor GATA6 is important for visceral endoderm differentiation. It is also expressed in early bronchial epithelium and the observation that this tissue does not receive any contribution from Gata6 double mutant embryonic stem (ES) cells in chimeric mice suggests that GATA6 may play a crucial role in lung development. The aim of this study was to determine the role of GATA6 in fetal pulmonary development. We show that Gata6 mRNA is expressed predominantly in the developing pulmonary endoderm and epithelium, but at E15.5 also in the pulmonary mesenchyme. Blocking or depleting GATA6 function results in diminished branching morphogenesis both in vitro and in vivo. TTF1 expression is unaltered in chimeric lungs whereas SPC and CC10 expression are attenuated in abnormally branched areas of chimeric lungs. Chimeras generated in a ROSA26 background show that endodermal cells in these abnormally branched areas are derived from Gata6 mutant ES cells, implicating that the defect is intrinsic to the endoderm. Taken together, these data demonstrate that GATA6 is not essential for endoderm specification, but is required for normal branching morphogenesis and late epithelial cell differentiation

    Changes in membrane sphingolipid composition modulate dynamics and adhesion of integrin nanoclusters

    Sphingolipids are essential constituents of the plasma membrane (PM) and play an important role in signal transduction by modulating clustering and dynamics of membrane receptors. Changes in lipid composition are therefore likely to influence receptor organisation and function, but how this precisely occurs is difficult to address given the intricacy of the PM lipid-network. Here, we combined biochemical assays and single molecule dynamic approaches to demonstrate that the local lipid environment regulates adhesion of integrin receptors by impacting on their lateral mobility. Induction of sphingomyelinase (SMase) activity reduced sphingomyelin (SM) levels by conversion to ceramide (Cer), resulting in impaired integrin adhesion and reduced integrin mobility. Dual-colour imaging of cortical actin in combination with single molecule tracking of integrins showed that this reduced mobility results from increased coupling to the actin cytoskeleton brought about by Cer formation. As such, our data emphasizes a critical role for the PM local lipid composition in regulating the lateral mobility of integrins and their ability to dynamically increase receptor density for efficient ligand binding in the process of cell adhesion
