    This research aimed to respond to a locally and nationally identified need (anonymous local authority, 2014; Department of Health, 2015) to improve wellbeing and reduce negative affect (i.e. anxiety) in young people (YP) by applying self-determination theory (SDT) (Deci & Ryan, 2000) in an empowerment process in two secondary/upper schools in one local authority. Four groups (n=13) of students from year 10, 11 and 13 who self-reported to be anxious were invited to participate in focus groups to discuss the causes of their anxieties and suggest school based mechanisms of support. Each group decided how they wanted to feedback their ideas to senior management who then explained what they would change to address students’ suggestions. Participants’ group notes from the focus groups were used to examine reported causes of anxiety and suggestions to improve wellbeing. Three quantitative questionnaires were used to measure changes in anxiety, wellbeing and need fulfilment, according to SDT, while a semi-structured questionnaire was used to measure any descriptive changes noticed by the students during the research. Thematic analyses of the qualitative data found that participants attributed causes of anxiety to factors that thwarted SDT need fulfilment while the support strategies they suggested were those that nurtured SDT need fulfilment. Due to the small sample, statistical significance claims cannot be made. However, the data appear to suggest overall improvements in anxiety and one aspect of wellbeing (i.e. positive affect). An overall improvement in need fulfilment was found more strongly in the qualitative data and older participants (year 13) made greater gains in the dependent variables compared to their younger counterparts (year 10&11). This research has implications for schools and Educational Psychologists and offers suggestions for future research in this area

    Weighing Neutrinos with Galaxy Cluster Surveys

    Large future galaxy cluster surveys, combined with cosmic microwave background observations, can achieve a high sensitivity to the masses of cosmologically important neutrinos. We show that a weak lensing selected sample of ~100,000 clusters could tighten the current upper bound on the sum of masses of neutrino species by an order of magnitude, to a level of 0.03 eV. Since this statistical sensitivity is below the best existing lower limit on the mass of at least one neutrino species, a future detection is likely, provided that systematic errors can be controlled to a similar level.Comment: 4 pages, 1 figure, version accepted for publication in PR

    Association between body mass index and health-related quality of life, and the impact of self-reported long-term conditions - cross-sectional study from the south Yorkshire cohort dataset

    Background We sought to quantify the relationship between body mass index (BMI) and health-related quality (HRQoL) of life, as measured by the EQ-5D, whilst controlling for potential confounders. In addition, we hypothesised that certain long-term conditions (LTCs), for which being overweight or obese is a known risk factor, may mediate the association between BMI and HRQoL. Hence the aim of our study was to explore the association between BMI and HRQoL, first controlling for confounders and then exploring the potential impact of LTCs. Methods We used baseline data from the South Yorkshire Cohort, a cross-sectional observational study which uses a cohort multiple randomised controlled trial design. For each EQ-5D health dimension we used logistic regression to model the probability of responding as having a problem for each of the five health dimensions. All continuous variables were modelled using fractional polynomials. We examined the impact on the coefficients for BMI of removing LTCs from our model. We considered the self-reported LTCs: diabetes, heart disease, stroke, cancer, osteoarthritis, breathing problems and high blood pressure. Results The dataset used in our analysis had data for 19,460 individuals, who had a mean EQ-5D score of 0.81 and a mean BMI of 26.3 kg/m2. For each dimension, BMI and all of the LTCs were significant predictors. For overweight or obese individuals (BMI ≥ 25 kg/m2), each unit increase in BMI was associated with approximately a 3% increase in the odds of reporting a problem for the anxiety/depression dimension, a 8% increase for the mobility dimension, and approximately 6% for the remaining dimension s. Diabetes, heart disease, osteoarthritis and high blood pressure were identified as being potentially mediating variables for all of the dimensions. Conclusions Compared to those of a normal weight (18.5 < BMI < 25 kg/m2), overweight and obese individuals had a reduced HRQoL, with each unit increase in BMI associated with approximately a 6% increase in the odds of reporting a problem on any of the EQ-5D health dimensions. There was evidence to suggest that diabetes, heart disease, osteoarthritis and high blood pressure may mediate the association between being overweight and HRQoL

    Synergistic action of dual IGF1/R and MEK inhibition sensitizes childhood acute lymphoblastic leukemia (ALL) cells to cytotoxic agents and involves downregulation of STAT6 and PDAP1.

    Heterogeneous upregulation of multiple prosurvival pathways underlies resistance to damage-induced apoptosis in acute lymphoblastic leukemia (ALL) cells despite normal p53 responses. Here, we show that the dual combination of insulin-like growth factor 1 (IGF1)/IGF1 receptor (IGF1/R) and mitogen-activated protein kinase/extracellular signal-regulated kinase (ERK) kinase (MEK) inhibition using AG1024 + U0126 can sensitize apoptosis-resistant ALL cells to ionizing radiation-induced DNA damage irrespective of effect of single pathway inhibition in vitro. This AG1024 + U0126 combination also significantly potentiates the ability of the core chemotherapy compounds vincristine, dexamethasone, and daunorubicin to kill ALL cells in vitro. Evidence of the synergistic action of AG1024 + U0126 in samples with variable basal levels of phosphorylated IGF1/Rβ and ERK1/2 suggested additional targets of this drug combination. Consistent with this, gene expression profiling identified 32 "synergy genes" differentially targeted by IGF1/R + MEK inhibition and, among these, Signal transducer and activator of transcription 6 (STAT6) and platelet-derived growth factor-associated protein 1 (PDAP1) were the most differentially downregulated cluster. Pearson correlation analyses revealed that STAT6 and PDAP1 display significant expression codependency and a common expression pattern linked with other key "synergy" genes, supporting their predicted role in an STAT6-ERK-nuclear factor kappa beta (NF-κB) network. Knockdown studies revealed that loss of STAT6, but not PDAP1, impinges on the cell cycle, causing reduced numbers of viable cells. In combination with daunorubicin, STAT6 loss has an additive effect on cell killing, whereas PDAP1 loss is synergistic, indicating an important role of PDAP1 in the cellular response to this anthracycline. Inhibition of STAT6 or PDAP1 may therefore represent a potential novel therapeutic strategy for resistant ALL by enhancing sensitivity to chemotherapy
