    Endogenous Retinoic Acid Activity in Principal Cells and Intercalated Cells of Mouse Collecting Duct System

    Background: Retinoic acid is the bioactive derivative of vitamin A, which plays an indispensible role in kidney development by activating retinoic acid receptors. Although the location, concentration and roles of endogenous retinoic acid in postnatal kidneys are poorly defined, there is accumulating evidence linking post-natal vitamin A deficiency to impaired renal concentrating and acidifying capacity associated with increased susceptibility to urolithiasis, renal inflammation and scarring. The aim of this study is to examine the presence and the detailed localization of endogenous retinoic acid activity in neonatal, young and adult mouse kidneys, to establish a fundamental ground for further research into potential target genes, as well as physiological and pathophysiological roles of endogenous retinoic acid in the post-natal kidneys.Methodology/Principal Findings: RARE-hsp68-lacZ transgenic mice were employed as a reporter for endogenous retinoic acid activity that was determined by X-gal assay and immunostaining of the reporter gene product, beta-galactosidase. Double immunostaining was performed for beta-galactosidase and markers of kidney tubules to localize retinoic acid activity. Distinct pattern of retinoic acid activity was observed in kidneys, which is higher in neonatal and 1- to 3-week-old mice than that in 5- and 8-week-old mice. The activity was present specifically in the principal cells and the intercalated cells of the collecting duct system in all age groups, but was absent from the glomeruli, proximal tubules, thin limbs of Henle's loop and distal tubules.Conclusions/Significance: Endogenous retinoic acid activity exists in principal cells and intercalated cells of the mouse collecting duct system after birth and persists into adulthood. This observation provides novel insights into potential roles for endogenous retinoic acid beyond nephrogenesis and warrants further studies to investigate target genes and functions of endogenous retinoic acid in the kidney after birth, particularly in the collecting duct system

    Synthesis of di-nuclear metal complexes, their anticancer and catalytic properties

    This thesis is related to synthesis of ruthenium (II) arene complexes with nitrogen donor and investigation of their biological and catalytic activities. As we known, platinum is mostly used as a metal in chemotherapy drugs. The limitations of platinum-based drugs, dose dependent side effects and development of drug resistance mechanisms, have boosted the research for finding other metal-based drugs. Among metals, ruthenium is probably the one showing the greatest promises. This thesis consists of three parts. In the first part, we mention the history of metals used in chemotherapy treatments and complexes that play a role in the discovery of ruthenium. In the second part experimental works are given. In the third part, characterization, catalytic and anticarcinogenic properties of the synthesized complexes are given. Starting from the amines (1), the oxoacetate-derived compounds (2), then the amide-derived compounds (3) and finally the amine ligands (4) by reduction reaction was synthesized. The resulting ligands were reacted with [Ru (p-cymene) Cl2]2 to obtain cationic Ru (II) arene complexes (5). The synthesis steps are summarized in Scheme I. These synthesized complexes (5) were investigated for anticarcinogenic activities in various diseased cell lines and used as catalysts in transfer hydrogenation reactions.Bu tez azot donorlü Rutenyum (II) aren komplekslerinin sentezi ve biyolojik aktiviteleri ile katalitik aktivitelerinin araştırılmasına ilişkindir. Rutenyum (II) aren kompleksleri antikanserojenik aktiviteleri bakımından oldukça umut verici ajanlardır. Bilindiği gibi kemoterapi ilaçlarında metal olarak çoğunlukla platin metali kullanılmaktadır. Platinin getirmiş olduğu sınırlamalar ve yan etkileri nedeniyle yeni metal arayışlarına girilmiştir ve en olası adayın rutenyum olduğu görülmektedir. Tez üç bölümden oluşmaktadır. Birinci bölümde kemoterapi tedavilerinde kullanılan metallerin tarihinden ve rutenyumun keşfinde rol oynayan komplekslerden söz edilmiştir. İkinci bölümde deneysel çalışmalara yer verilmiştir. Üçüncü bölümde ise, sentezlenen komplekslerin karakterizasyonu, katalitik özellikleri ve antikanserojen özellikleri yer verilmiştir. Başlangıçta aminlerden (1) yola çıkarak okso asetat türevli bileşikler (2), daha sonra amit türevli bileşikler (3) ve son olarak indirgenme reaksiyonlarıyla amin ligandları (4) sentezlendi. Elde edilen ligandlar [Ru(p-simen)Cl2]2 ile etkileştirilerek katyonik Ru(II) aren kompleksleri (5) elde edidi. Sentez basamakları Şema I’de özetlenmiştir. Sentezlenen bu komplekslerin (5) çeşitli hastalıklı hücre hatlarında antikanserojen aktiviteleri araştırıldı ve transfer hidrojenasyon reaksiyonlarında katalizör olarak kullanıldı

    Hemoglobin, hematocrit, serum iron, total serum iron binding capacity, transferrin saturation and ferritin levels of 27 pregnant women and their babies at delivery and at 8-10 weeks of age and of their mature breast milk were determined. Eighteen pregnant women with iron deficiency anemia and 9 healthy pregnant women as controls constituted the groups in this study. Excepting serum iron (p 0.02). Besides, a significant relation (r:0.535) was found between the ferritin values of the cord blood and of the infants' sera at 8-10 weeks of age

We report herein a case of metastatic tumor at the site of transthoracic needle biopsy following a curative resection in a patient with stage IB bronchogenic carcinoma. The patient was managed with aggressive chest wall resection and subsequent musculocutaneus flap transposition. however he died I I months after the initial operation. The tumor implantation risk and the related complications should be considered in patients with operable bronchogenic carcinoma undergoing a tranthoracic needle aspiration biopsy.     Multimodal imaging features of a spontaneously resolved unilateral congenital macrovessel-related macular edema in a 13-year-old boy

    Purpose: Congenital retinal macrovessels (CRM) are large aberrant blood vessels that cross the horizontal raphe. Visual acuity may be negatively affected in cases afflicted with CRM due to macular serous detachment, hemorrhage, exudation, foveolar cyst formation and retinal artery occlusion. Even the presence of an anomalous vessel itself running through the foveal avascular zone may compromise the visual acuity. In this case report, we discuss the multimodal imaging characteristics of a case of symptomatic unilateral CRM-related cystoid macular edema and serous macular detachment in a 13-year-old boy.Observations: Optical coherence tomography angiography (OCTA) imaging of the superficial retina revealed the aberrant vessel and anomalous arteriovenous communication between the aberrant vessel and the retinal artery. The foveal avascular zone also appeared partly distorted.Conclusions: The present case reports the second-youngest patient afflicted with unilateral CRM and associated macular edema. There was spontaneous resolution of macular edema within six weeks. In light of the observations in this case and the literature review, the need to refrain from rapid therapeutic intervention in the eyes of patients affected with CRM and macular edema must be emphasized
