    Education and Indigenous Territorial Struggles : A study on the Sapara people’s experiences with the education system in the Ecuadorian Amazon

    The focus of this thesis is on the education of indigenous peoples, especially on how education can facilitate territorial self-determination and political emancipation for them. Indigenous movements world-wide and in Ecuador have focused on creating education respectful of and relevant to indigenous cultural background and knowledge. This thesis explores further the interconnectedness of education and indigenous territorial politics, as they have been together in the forefront of the indigenous movement in Ecuador, and they link the epistemological struggle of recognising Indigenous knowledges to environmental issues prevalent in the country dependent on extractivism. As indigenous peoples often inhabit environmentally vulnerable regions, this thesis examines how for the indigenous groups of Ecuadorian Amazon the relationship between education and territory can aim to be mutually beneficial, encouraging both preservation of the diverse cultures and environment in the biocultural landscapes. The research was conducted as an ethnographical case study on the province of Pastaza, situated in the Ecuadorian Amazon with a special focus on the indigenous group known as Sapara, who are the smallest of 14 indigenous nationalities recognised under the plurinational state of Ecuador. The data of the study consisted of 27 semi-structured interviews and participant observation recorded in the field diary, accompanied by historical analysis of intercultural bilingual education in Ecuador. This thesis illustrates the place-making practices and histories of indigenous peoples, acknowledged under the term Indigenous knowledge, as they form a foundation for territorial politics. Possibility for epistemological diversity in the education system is understood through principle of interculturality, as articulated by the indigenous movement itself as a radical project of recognising lived heritage of cultural and historical differences in dialogue between various segments of society. The topic is examined through the concept of territory, which emphasises a question of governance in plurinational Ecuador, where indigenous nationalities struggle to exercise control over their respective territories. Territory is formed of competing political projects that aim to define and redefine its meaning, which also opens up a definition of territory to scrutinise what type of power actually operates in these political projects and rejects assumptions of simple top-down governance as the only possible territorial form. This theoretical framework facilitates the analysis of education as a part of territorial strategies. The main argument of this thesis is that education constructs a significant part of reinforcing political emancipation and territorial self-determination of indigenous peoples. Based on historical and ethnographical analysis, the thesis illustrates how education functions as a privileged arena of cultural struggle to achieve epistemological diversity that includes Indigenous knowledges alongside with Western science. Simultaneously, education, which is perceived pivotal for living well, acts as a societal force that can transform material foundations of life, since indigenous peoples modify their residential patterns in order to access education. As indigenous territories remain only partly autonomous, since the nation-state retains control over subsoil resources, land continues to be an arena of competing political interests. This accentuates the importance of planning education practices to facilitate living inside the whole territory, since inhabiting space asserts the claims of indigenous groups effectively, allowing them to practice a strategy of dispersal

    Analysis of H3K4me3 and H3K27me3 bivalent promotors in HER2+ breast cancer cell lines reveals variations depending on estrogen receptor status and significantly correlates with gene expression

    Background: The role of histone modifications is poorly characterized in breast cancer, especially within the major subtypes. While epigenetic modifications may enhance the adaptability of a cell to both therapy and the surrounding environment, the mechanisms by which this is accomplished remains unclear. In this study we focus on the HER2 subtype and investigate two histone trimethylations that occur on the histone 3; the trimethylation located at lysine 4 (H3K4me3) found in active promoters and the trimethylation located at lysine 27 (H3K27me3) that correlates with gene repression. A bivalency state is the result of the co-presence of these two marks at the same promoter.Methods: In this study we investigated the relationship between these histone modifications in promoter regions and their proximal gene expression in HER2+ breast cancer cell lines. In addition, we assessed these patterns with respect to the presence or absence of the estrogen receptor (ER). To do this, we utilized ChIP-seq and matching RNA-seq from publicly available data for the AU565, SKBR3, MB361 and UACC812 cell lines. In order to visualize these relationships, we used KEGG pathway enrichment analysis, and Kaplan-Meyer plots.Results: We found that the correlation between the three types of promoter trimethylation statuses (H3K4me3, H3K27me3 or both) and the expression of the proximal genes was highly significant overall, while roughly a third of all genes are regulated by this phenomenon. We also show that there are several pathways related to cancer progression and invasion that are associated with the bivalent status of the gene promoters, and that there are specific differences between ER+ and ER- HER2+ breast cancer cell lines. These specific differences that are differentially trimethylated are also shown to be differentially expressed in patient samples. One of these genes, HIF1AN, significantly correlates with patient outcome.Conclusions: This study highlights the importance of looking at epigenetic markings at a subtype specific level by characterizing the relationship between the bivalent promoters and gene expression. This provides a deeper insight into a mechanism that could lead to future targets for treatment and prognosis, along with oncogenesis and response to therapy of HER2+ breast cancer patients.</p

    Parental Education and Pre-School Children's Objectively Measured Sedentary Time : The Role of Co-Participation in Physical Activity

    Parental co-participation in physical activity (PA) may be a beneficial parenting practice for diminishing children's sedentary time (ST). Less information is available, however, on the explanatory role of co-participation in PA regarding parental educational differences in children's ST. Preschool-aged children (N = 864, mean age 4.8, 52% boys) with their parents participated in a cross-sectional DAGIS (Increased Health and Wellbeing in Pre-schools) study between years 2015 and 2016. Children (N = 821) wore an accelerometer for one week. Parents were informed of their educational background, and the frequency of visits with their child in nature, to parks or playgrounds, their own yard, and indoor sport facilities (N = 808). Testing the associations required multiple regression analyses. Parents with a low educational background reported more frequent visits with their child to their own yard, and these visits were associated with children's lower ST. More highly educated parents co-visited indoor sport facilities more frequently, although this did not have a significant association with children's ST. More frequent visits in nature were associated with a lower ST at weekdays, regardless of educational background. Future health promotion strategies should inform parents that frequent co-participation in PA, for example, in one's own yard, is beneficial for lowering children's ST.Peer reviewe

    A cross-sectional study of children's temperament, food consumption and the role of food-related parenting practices

    Although evidence exists of the association between children's temperament and weight, only few studies have examined how temperament is associated with actual food consumption among preschoolers. We examined concurrent associations between children's temperament and the consumption of different foods, and investigated whether the association between children's temperament and vegetable consumption is mediated by vegetable-related parenting practices. We utilized the data from the cross-sectional DAGIS study of 864 preschool children aged between three to six and their families, conducted between 2015 and 2016 in Finland. The parents reported their children's temperament, food consumption, and their vegetable-related parenting practices. Adjusted logistic regression analyses found positive associations between surgency and vegetable consumption as well as between effortful control and vegetable consumption. Both associations were mediated by one examined vegetable-related parenting practice: enhanced availability and autonomy support. No associations were found between children's negative affectivity and food consumption or vegetable-related parenting practices. In conclusion, children's temperament may be an important factor behind food-related parenting practices and children's diet. However, further longitudinal research and research covering different food-related parenting practices and home environment factors is necessary to better understand the complex associations between temperament and food consumption among young children.Peer reviewe

    The Contribution of Preschool Meals to the Diet of Finnish Preschoolers

    Preschool meals may influence the formation of children’s dietary habits and health. We assessed the contribution of preschool meals to the diet of Finnish children. We used food record data from the cross-sectional DAGIS survey and selected recording days which included all three meals (breakfast, lunch, afternoon snack) at preschool. We analyzed the diet of three- to four-year-olds (n = 324) and five- to six-year-olds (n = 233). Preschool meals accounted for 54% of the weekday’s energy intake in both age groups, and provided ≥60% of total fiber, polyunsaturated fatty acids, and vitamins D and E. More than 60% of fish dishes but only one third of total daily fresh fruit were consumed at preschool. The mean (SD) percentages of energy from protein and fat at preschool were 17% (3%) and 30% (7%) in the younger and 17% (3%) and 31% (6%) in the older age group, respectively. The mean proportions of energy from added sugar at preschool were below 5% in both age groups. On average, salt intake exceeded recommendations and 60% of salt came from preschool food. Tackling high salt intake should be a future goal of guidance for early childhood education and care food services

    Placental DNA methylation marks are associated with maternal depressive symptoms during early pregnancy

    Maternal depressive symptoms during pregnancy are a significant risk factor for adverse developmental and health outcomes of the offspring. The molecular mechanisms mediating the long-term effects of this exposure are not well understood. Previous studies have found association between prenatal exposure to maternal psychological distress and placental DNA methylation of candidate genes, which can influence placental barrier function and development of the fetus. Our objective in this study was to determine epigenome wide association of maternal depressive symptoms in early pregnancy with the placental DNA methylation. For this purpose we examined DNA methylomes of 92 placental samples by using reduced representation bisulfite sequencing. The placental samples were collected after deliveries of 39 girls and 59 boys, whose mothers had Edinburgh Postnatal Depression Score ranging from 0 to 19 at gestational week 14. According to our results maternal depressive symptoms are associated with DNA methylation of 2833 CpG sites, which are particularly over-represented in genic enhancers. The genes overlapping or nearest to these sites are functionally enriched for development of neurons and show expression enrichment in several regions of developing brain. The genomic regions harboring the DNA methylation marks are enriched for single nucleotide polymorphisms associated with mental disease trait class. Potential cellular signaling cascades mediating the effects include inflammatory and hormonal pathways. As a conclusion our results suggest that maternal depressive symptoms during early pregnancy are associated with DNA methylation marks in placenta in genes, which are important for the development and long-term health of the brain. Whether similar marks can be detected in exposed children remains to be elucidated in further studies.</p
