    The Rise and Fall of Digital Music Distribution Services: a Cross-Case Comparison of MP3.com, Napster and Kazaa

    The aim of this research is to assess the role of digital music distribution services in the downturn and restructuring of the music industry. The research was bound by the systems of innovation conceptual framework and explores the dynamics between the technical, social, legal and economic dimensions with the purpose of understanding how the traditional supply chain has changed. The cases of MP3.com, Napster and Kazaa are studied for their historical significance and the sheer breadth of issues they bring to the fore. Data was collected using documentation review and interviews. The findings of the research show that the creation of digital music distribution services initially resulted in disintermediation. However, it was not long before record companies reintermediated into the supply chain with pay-per-download (PPD) and subscription payment schemes. Today, the law seems to be catching up with entities that illegally distribute music but it has not ceased the development of newer online establishments that are technically not in breach of legislation

    The Master Gardener Volunteer Program led by Royal Botanic Gardens and Domain Trust. The catalyst for the inception of the Master Gardener Volunteer Program (MGVP) in Sydney arose when Philip Pettitt (the then Community Greening Coordinator) travelled to the United States to attend the Botanic Gardens Conservation International Conference in 2015 and was inspired by the Brooklyn Urban Gardener Course operating in New York and Brooklyn. Upon his return, the Royal Botanic Gardens and Domain Trust (RBGDT) piloted three short courses in 2016 funded by the Office of Environment and Heritage. Further funding was received from the Department of Social Services Resilient Communities Fund in 2018 to expand the MGVP for a period of three years. This study focuses on the delivery of the MGVP at 17 sites in New South Wales, Australia. These include: Liverpool, Curran, Central Coast, Wollongong, Riverwood, Mt Annan, Auburn, Sydney, Eden, Newcastle, Miller, Telopea, Willmott, Bega, Warrigal, Yallah, and Nowra. The overarching objectives of this research were to investigate the Master Gardener Volunteers’ self-reported changes to wellbeing, social cohesion, skill development, and participation as they engage over time in the MGVP; and identify the reported strengths of the program delivery and areas for improvement

    Serum Profiles of C-Reactive Protein, Interleukin-8, and Tumor Necrosis Factor-α in Patients with Acute Pancreatitis

    Background-Aims. Early prediction of the severity of acute pancreatitis would lead to prompt intensive treatment resulting in improvement of the outcome. The present study investigated the use of C-reactive protein (CRP), interleukin IL-8 and tumor necrosis factor-α (TNF-α) as prognosticators of the severity of the disease. Methods. Twenty-six patients with acute pancreatitis were studied. Patients with APACHE II score of 9 or more formed the severe group, while the mild group consisted of patients with APACHE II score of less than 9. Serum samples for measurement of CRP, IL-8 and TNF-α were collected on the day of admission and additionally on the 2nd, 3rd and 7th days. Results. Significantly higher levels of IL-8 were found in patients with severe acute pancreatitis compared to those with mild disease especially at the 2nd and 3rd days (P = .001 and P = .014, resp.). No significant difference for CRP and TNF-α was observed between the two groups. The optimal cut-offs for IL-8 in order to discriminate severe from mild disease at the 2nd and 3rd days were 25.4 pg/mL and 14.5 pg/mL, respectively. Conclusions. IL-8 in early phase of acute pancreatitis is superior marker compared to CRP and TNF-α for distinguishing patients with severe disease

    Biosynthesis of γ-aminobutyric acid by lactic acid bacteria in surplus bread and its use in bread making

    Aims: The aim of this study was to investigate the effectiveness of bread as substrate for γ-aminobutyric acid (GABA) biosynthesis, establishing a valorization strategy for surplus bread, repurposing it within the food chain. Methods and results: Surplus bread was fermented by lactic acid bacteria (LAB) to produce GABA. Pediococcus pentosaceus F01, Levilactobacillus brevis MRS4, Lactiplantibacillus plantarum H64 and C48 were selected among 33 LAB strains for the ability to synthesize GABA. Four fermentation experiments were set up using surplus bread as such, added of amylolytic and proteolytic enzymes, modifying the pH or mixed with wheat bran. Enzyme-treated slurries led to the release of glucose (up to 20 mg g-1 ) and free amino acid, whereas the addition of wheat bran (30% of bread weight) yielded the highest GABA content (circa 800 mg kg-1 of dry weight) and was the most suitable substrate for LAB growth. The selected slurry was ultimately used as an ingredient in bread making causing an increase in free amino acids. Conclusions: Besides the high GABA concentration (148 mg kg-1 dough), the experimental bread developed in this study was characterized by good nutritional properties, highlighting the efficacy of tailored bioprocessing technologies as means to mitigate food wastage. Significance and impact of study: Our results represent a proof of concept of effective strategies to repurpose food industry side streams

    The Ten-Year Road: Joys and Challenges on the Road to Tenure

    This paper explores the pre-tenure experiences of five assistant professors employed in the faculty of education of a research-intensive university. Acting as co-researchers, the authors researched their experiences through a critical narrative approach. The analysis, informed by critically-oriented writing that extends Wenger\u27s Communities of Practice, takes as axiomatic the notion that globalized processes of economic restructuring are mediating work in the academy and examines its local manifestations. Discussions explore issues of power, equity, shifting identities, and the need for improved navigational resources. The authors found that the process of critically and collaboratively researching their pre-tenure experiences offered insight into sites of personal and professional agency and also served as the impetus to form the social semiotic spaces that encouraged a sense of community.     Challenges for Developing Complex System Governance

    This paper examines the challenges and practice implications for Complex System Governance (CSG). CSG is presented as an emerging field focused on the design, execution, and evolution of the higher order (metasystem) functions necessary to provide control, communication, coordination, and integration of a complex system. This paper is focused on three primary objectives. First, we introduce the complex system problem domain that the CSG field is being designed to address. The pervasiveness of this problem domain is demonstrated by a short examination of the water utilities sector. Second, we expound the nature of CSG and an emerging reference model that defines the functions of CSG. These functions must be performed by any system that maintains viability (continued existence). The CSG reference model rests on the underlying conceptual foundations built from Systems Theory (axioms and propositions governing system integration and coordination) and Management Cybernetics (communication and control for effective system organization). Third, we explore the particular challenges that must be addressed if the potential of the emerging CSG field is to be realized. The paper concludes by suggesting the potential that the CSG field brings for enhancing practitioner capabilities to more effectively deal with complex systems and their associated problems

    Predator-induced defences in Daphnia pulex: Selection and evaluation of internal reference genes for gene expression studies with real-time PCR

    <p>Abstract</p> <p>Background</p> <p>The planktonic microcrustacean <it>Daphnia pulex </it>is among the best-studied animals in ecological, toxicological and evolutionary research. One aspect that has sustained interest in the study system is the ability of <it>D. pulex </it>to develop inducible defence structures when exposed to predators, such as the phantom midge larvae <it>Chaoborus</it>. The available draft genome sequence for <it>D. pulex </it>is accelerating research to identify genes that confer plastic phenotypes that are regularly cued by environmental stimuli. Yet for quantifying gene expression levels, no experimentally validated set of internal control genes exists for the accurate normalization of qRT-PCR data.</p> <p>Results</p> <p>In this study, we tested six candidate reference genes for normalizing transcription levels of <it>D. pulex </it>genes; alpha tubulin (aTub), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), TATA box binding protein (Tbp) syntaxin 16 (Stx16), X-box binding protein 1 (Xbp1) and CAPON, a protein associated with the neuronal nitric oxide synthase, were selected on the basis of an earlier study and from microarray studies. One additional gene, a matrix metalloproteinase (MMP), was tested to validate its transcriptional response to <it>Chaoborus</it>, which was earlier observed in a microarray study. The transcription profiles of these seven genes were assessed by qRT-PCR from RNA of juvenile <it>D. pulex </it>that showed induced defences in comparison to untreated control animals. We tested the individual suitability of genes for expression normalization using the programs geNorm, NormFinder and BestKeeper. Intriguingly, Xbp1, Tbp, CAPON and Stx16 were selected as ideal reference genes. Analyses on the relative expression level using the software REST showed that both classical housekeeping candidate genes (aTub and GAPDH) were significantly downregulated, whereas the MMP gene was shown to be significantly upregulated, as predicted. aTub is a particularly ill suited reference gene because five copies are found in the <it>D. pulex </it>genome sequence. When applying aTub for expression normalization Xbp1 and Tbp are falsely reported as significantly upregulated.</p> <p>Conclusions</p> <p>Our results suggest that the genes Xbp1, Tbp, CAPON and Stx16 are suitable reference genes for accurate normalization in qRT-PCR studies using <it>Chaoborus</it>-induced <it>D. pulex </it>specimens. Furthermore, our study underscores the importance of verifying the expression stability of putative reference genes for normalization of expression levels.</p

    Nutritional intake and food sources in an adult urban Kenyan population

    Urbanisation is hastening the transition from traditional food habits to less healthy diets, which are becoming more common among Kenyans. No up-to-date studies on usual dietary intake and the main food sources of adult Kenyans are available. The aim of the present study was to identify the main food sources of nutrients in the diet of urban adult Kenyans and explore potential associations with demographic variables including age, sex, level of education, occupation and body mass index. The study adopted a cross-sectional design. The dietary intake of 486 adult Kenyans from Nairobi was assessed using a validated, culture-sensitive, semi-quantitative food frequency questionnaire. Binary logistic regression models were used to evaluate associations between food sources and demographic variables. Macronutrient intakes as a proportion of total energy intake (TEI) were within international dietary guidelines. Cereals and grain products (34.0%), sugar, syrups, sweets and snacks (9.8%), fruits (9.7%) and meat and eggs (8.8%) were the major contributors to TEI. Cereals and grain products contributed 42.5% to carbohydrates, followed by fruits (12.4%) and sugar, syrups, sweets and snacks (10.6%). The most important sources of protein and total fat were cereals and grain products (23.3% and 19.7%, respectively) and meat and eggs (22.0% and 18.7%, respectively). Sex, age and level of education were associated with the choice of food groups. Although macronutrient intakes were within guidelines, the Kenyan diet was revealed to be high in sugars, salt and fibre, with differences in food sources according to demographic variables. These results can act as an incentive to national authorities to implement nutritional strategies aiming to raise awareness of healthier dietary patterns among Kenyans. © 2022 The Authors.     Enhanced Approaches for Identifying Amadori Products: Application to Peanut Allergens

    The dry roasting of peanuts is suggested to influence allergic sensitization as a result of the formation of advanced glycation end products (AGEs) on peanut proteins. Identifying AGEs is technically challenging. The AGEs of a peanut allergen were probed with nano-scale liquid chromatography−electrospray ionization−mass spectrometry (nanoLC−ESI−MS) and tandem mass spectrometry (MS/MS) analyses. Amadori product ions matched to expected peptides and yielded fragments that included a loss of three waters and HCHO. As a result of the paucity of b and y ions in the MS/MS spectrum, standard search algorithms do not perform well. Reactions with isotopically labeled sugars confirmed that the peptides contained Amadori products. An algorithm was developed on the basis of information content (Shannon entropy) and the loss of water and HCHO. Results with test data show that the algorithm finds the correct spectra with high precision, reducing the time needed to manually inspect data. Computational and technical improvements allowed for better identification of the chemical differences between modified and unmodified proteins
