    An Oxidized Fat Containing Diet Decreases Weight Gain but Increases Adiposity in Mice Fed a Low Fat Diet

    Introduction: Fast and convenience foods are abundant, relatively inexpensive, and accommodating to the fast-paced lifestyle of many Americans. One popular method of cooking used by many fast food establishments is deep-fat frying. Soybean oil is commonly used for frying and is rich in polyunsaturated fatty acids (PUFA) such as linoleic acid (LA). When soybean oil is used for deep-fat frying, LA becomes oxidized (Ox-LA). Endogenous Ox-LA has the capacity to be a ligand to peroxisome proliferator-activated receptor gamma (PPAR¥ã), a nuclear transcription factor that regulates adipocyte maturation. It is not yet known whether or not dietary Ox-LA has the same capacity with respect to PPAR¥ã. Considering the fact that dietary oxidized lipids are abundant in the typical American diet, it is important to know if they regulate weight gain and especially adipose tissue mass. In this study, we investigate the effects of fresh and heated soybean oil on weight gain and adiposity in mice fed isocaloric low fat diets. Methods: Soybean oil was heated on a hot plate, under a hood, at 190¨¬C for three hours. Fresh soybean oil served as the source of unoxidized oil (Unox-oil) and the heated oil served as the source of oxidized oil (Ox-oil). Both the Ox-oil and Unox-oil were incorporated into a low-fat (10% of calories) mouse chow by Research Diets, Inc. (New Brunswick, NJ). Sixteen C57BL/6J mice were divided into two groups and fed low fat diets with Ox-oil (low fat oxidized, LFO) or with Unox-oil (low fat unoxidized, LFU). Another group of 8 mice were pair fed to the LFO group with the Unox-oil containing chow (PLU). Mice in the LFO and LFU groups were fed ad libitum and known amounts of fresh food was added to the cages every three days. Leftover food was weighed. Body weights were measured once a week. After 16 weeks mice were euthanized and epididymal white adipose tissue (EWAT), retroperitoneal white adipose tissue (RWAT), inguinal white adipose tissue (IWAT), and intrascapular brown adipose tissue (IBAT) samples were collected, weighed and stored at -80 degrees Celsius until further analysis. Fat pads were homogenized and cytosolic and nuclear proteins were extracted by standard methods. These extracts were subjected to Western blotting to determine the amount of PPAR¥ã in the cytosol and nuclear compartments of the fat pads. Differences in group means were analyzed by Mann Whitney U test. Comparisons were considered statistically significant at a p-value of \u3c 0.05. Results: Final mean body weights were significantly different when comparing the mice in the LFU group to the pair fed mice (PLU) (mean ¡¾ SD; 29.52 ¡¾ 1.09 grams (g) and 26.85 ¡¾ 1.44 g, respectively; p \u3c 0.05). Mice fed a low fat diet consisting of Ox-oil (LFO) had a final mean body weight of 27.88 ¡¾ 2.03 g. Mice in the LFU group gained significantly more weight on average than did mice in the LFO or PLU groups (mean ¡¾ SD; 8.86 ¡¾ 1.37g, 7.10 ¡¾ 1.47 g, and 5.71 ¡¾ 1.13 g, respectively). Although mean food intakes were not significantly different between any of the three groups, the average food intake was greatest for the LFU mice in comparison to the LFO and the PLU mice (mean ¡¾ SD; 20.65 ¡¾ 0.09 g/week, 18.40 ¡¾ 0.05 g/week, and 18.38 ¡¾ 0.19 g/week, respectively). Feeding efficiency (g of weight gain/g of food consumed) was the highest in the LFU mice compared to the PLU mice (mean ¡¾ SD; 0.031 ¡¾ 0.005 g/g and0.022 ¡¾ 0.004 g/g) and this difference was statistically significant. The LFO mice gained less weight per gram of food consumed than did the LFU mice (mean ¡¾ SD; 0.028 ¡¾ 0.006 g/g). Mean weights of all fat pads in the LFO group were significantly greater than those of the LFU and PLU mice (mean ¡¾ SD; 0.329 ¡¾ 0.109g, 0.199 ¡¾ 0.055g, and 0.219 ¡¾ 0.041 for EWAT, 0.091 ¡¾ 0.039g, 0.050 ¡¾ 0.026g, and 0.051 ¡¾ 0.017 for RWAT, 0.221 ¡¾ 0.065g, 0.135 ¡¾ 0.053g, and 0.144 ¡¾ 0.038 for IWAT, and 0.079 ¡¾ 0.012g, 0.055 ¡¾ 0.013g, and 0.062 ¡¾ 0.011 for IBAT, respectively). PPAR¥ã protein in the cytosol of EWAT fat pads was analyzed and quantified in comparison to the amount of Glyceraldehyde-3-phosphate dehydrogenase (GAPDH; loading control) present. Mean PPAR¥ã /GAPDH ratios for LFU mice was 0.226 ¡¾ 0.082, for LFO mice was 0.264 ¡¾ 0.122, and for PLU mice was 0.234 ¡¾ 0.108. Mean PPAR¥ã:GAPDH ratios were not significantly different between any of the groups. Conclusion: It appears that the consumption of oxidized oil caused a significant decrease in weight gain and food intake (although not significant) and a significant increase in fat pad mass in mice compared to those consuming a diet with unoxidized oil. The lack of difference in the amount of PPAR¥ã among the three groups of mice suggests that the changes in weight gain and fat pad mass among the oxidized oil consuming animals is not mediated through regulation of PPAR¥ã protein. To our knowledge, ours is the first study to report that mice consuming a low fat diet inclusive of dietary oxidized lipids exhibit greater adiposity than do mice consuming a low fat diet consisting of unoxidized lipids

    ‘Grounding a PIE in the sky’:Laying empirical foundations for a psychologically informed environment (PIE) to enhance well-being and practice in a homeless organisation

    While psychologically informed environments (PIEs) are gaining in prominence in efforts to improve well-being and practice in the homeless sector, their empirical foundations remain tenuous. We present a unique scoping needs analysis of staff and client well-being, staff attitudes and the social–therapeutic climate in a UK-based homeless prevention organisation (prior to PIE implementation). Our aims were: (a) to apply a robust framework to pinpoint need and target forthcoming PIE initiatives and (b) to establish a validated needs baseline that informs and measures efficacy of PIE for its future development. Four established personal and practice well-being measures were administered to 134 (predominantly ‘frontline’) staff and 50 clients. Staff completed the: Warwick-Edinburgh Mental Wellbeing Scale (WEMWBS), Professional Quality of Life Scale (measuring compassion satisfaction [CS], burnout [BO] and secondary traumatic stress [STS]), Attitudes related to Trauma-informed Care Scale (ARTIC-10; measuring practice attitudes towards trauma-informed values) and the Essen Climate Evaluation Schema (EssenCES; measuring perceptions of client cohesion, safety and practitioner relationships in housing projects). Clients completed the WEMWBS and EssenCES. Vulnerability to STS was evident in nearly two-thirds of frontline staff and it was a statistically significant predictor of BO. It was not, however, associated with lesser levels of CS. We discuss this complex dynamic in relation to highlighted strategic recommendations for the PIE framework, and the identified potential challenges in implementing trauma-informed and reflective practice in the organisation. We conclude with a critique of the value and the lessons learnt from our efforts to integrate stronger empirical substance into the PIE approach

    Application and Analysis of Project Management Techniques in Development of a Haptic Feedback Navigation System

    GPS navigation systems and applications have transformed the way drivers get to their destination. With the guidance of a computer-generated voice, users can receive turn-by-turn directions without the dangerous distraction of a map – but what about deaf drivers who cannot hear this voice? Despite the millions of deaf and hard-of-hearing drivers in the United States, this group has limited solutions for safe, convenient, and simple navigation. A team of six undergraduate students came together to provide a solution to this problem in the form of a wristband that uses haptic feedback to guide users to their destination.To develop this solution, several project management techniques were applied, with varying degrees of success. The goal was to apply these techniques to this student project as if it was a professional product development project, with appropriate timelines and cost estimates. After an initial project management plan including a work breakdown structure (WBS), responsibility matrix, feasibility study, and earned value plan was developed, the project began. Data regarding project progress was collected on the 30th and 60th day of progress and used to calculate project performance metrics

    Resilience as a Moderator Between Adverse Childhood Events and Health in Motherhood

    The purpose of this study was to examine the effects of adverse childhood events (ACEs) and motherhood on a weighted linear composite of health status and health harming behavior, while moderating for a measure of resilience. Felitti’s theory of ACEs and Garmezy’s theory of resilience are the theoretical framework for this study. The research questions that are addressed in this study are how ACEs and resilience affect health-related outcomes in women and how motherhood influence health-related outcomes as a function of ACEs when moderated by resilience. A quantitative research design with a convenience sample was used to anonymously survey 205 women (104 mothers, 101 nonmother). The survey included the 10-item ACEs questionnaire, the 10-item Connor-Davidson Resilience Scale short version, and two 15-item health and health behavior questionnaires. Once data were collected a three-way multivariate analysis of variance (MANOVA) was conducted using SPSS that showed correlation between negative health outcomes and ACEs, Motherhood and the triple interaction of ACEs, resilience, and motherhood. Women with ACEs had 1.04 points higher health scores than those without ACEs, mothers had 1.38 points higher health scores than nonmothers, mothers with ACEs and high resilience had a negative health score 1.4 points higher than mothers with high resilience and no ACEs. Implications for this study include the use of conclusive statistical data to support programs for family services and women’s outreach programs that could help prevent the cycle of ACEs in families and help mothers understand the role of ACEs and resilience on the impact of becoming a mother

    Computational prediction and experimental validation of novel Hedgehog-responsive enhancers linked to genes of the Hedgehog pathway

    Abstract Background The Hedgehog (Hh) signaling pathway, acting through three homologous transcription factors (GLI1, GLI2, GLI3) in vertebrates, plays multiple roles in embryonic organ development and adult tissue homeostasis. At the level of the genome, GLI factors bind to specific motifs in enhancers, some of which are hundreds of kilobases removed from the gene promoter. These enhancers integrate the Hh signal in a context-specific manner to control the spatiotemporal pattern of target gene expression. Importantly, a number of genes that encode Hh pathway molecules are themselves targets of Hh signaling, allowing pathway regulation by an intricate balance of feed-back activation and inhibition. However, surprisingly few of the critical enhancer elements that control these pathway target genes have been identified despite the fact that such elements are central determinants of Hh signaling activity. Recently, ChIP studies have been carried out in multiple tissue contexts using mouse models carrying FLAG-tagged GLI proteins (GLIFLAG). Using these datasets, we tested whether a meta-analysis of GLI binding sites, coupled with a machine learning approach, could reveal genomic features that could be used to empirically identify Hh-regulated enhancers linked to loci of the Hh signaling pathway. Results A meta-analysis of four existing GLIFLAG datasets revealed a library of GLI binding motifs that was substantially more restricted than the potential sites predicted by previous in vitro binding studies. A machine learning method (kmer-SVM) was then applied to these datasets and enriched k-mers were identified that, when applied to the mouse genome, predicted as many as 37,000 potential Hh enhancers. For functional analysis, we selected nine regions which were annotated to putative Hh pathway molecules and found that seven exhibited GLI-dependent activity, indicating that they are directly regulated by Hh signaling (78 % success rate). Conclusions The results suggest that Hh enhancer regions share common sequence features. The kmer-SVM machine learning approach identifies those features and can successfully predict functional Hh regulatory regions in genomic DNA surrounding Hh pathway molecules and likely, other Hh targets. Additionally, the library of enriched GLI binding motifs that we have identified may allow improved identification of functional GLI binding sites.http://deepblue.lib.umich.edu/bitstream/2027.42/134520/1/12861_2016_Article_106.pd
