    Families and Communities Against Child Sexual Exploitation (FCASE) : final evaluation report

    This is the final evaluation report for the Barnardo’s Families and Communities Against Sexual Exploitation project (FCASE), produced by the International Centre, researching Child Sexual Exploitation, Violence and Trafficking at the University of Bedfordshire. The programme was launched in April 2013, funded by the Department for Education (DfE) and concluded in March 2015. The evaluation was undertaken during the same period. The FCASE model has been piloted in three sites, which for the purposes of this report have been anonymised and will be referred to using pseudonyms. It consists of the following elements: a structured programme of six to eight weeks direct work with young people and families where a risk of child sexual exploitation (CSE) has been identified; delivery of CSE training with professionals; and undertaking community awareness raising. The evaluation has been informed by a range of qualitative data. The report identifies the elements that work well and some of the challenges in its implementation. This had been done in order to determine good practice in supporting families and communities and embed more effective practice on protecting children and young people, including those in foster care, from sexual exploitation, harnessing the protective factors within a child’s family and/or foster home. The learning from the project is intended to help other agencies to implement the FCASE model. An on-line learning resource is to be produced in order to facilitate this process

    A rapid culture independent methodology to quantitatively detect and identify common human bacterial pathogens associated with contaminated high purity water

    Background: Water and High Purity Water (HPW) distribution systems can be contaminated with human pathogenic microorganisms. This biocontamination may pose a risk to human health as HPW is commonly used in the industrial, pharmaceutical and clinical sectors. Currently, routine microbiological testing of HPW is performed using slow and labour intensive traditional microbiological based techniques. There is a need to develop a rapid culture independent methodology to quantitatively detect and identify biocontamination associated with HPW. Results: A novel internally controlled 5-plex real-time PCR Nucleic Acid Diagnostics assay (NAD), was designed and optimised in accordance with Minimum Information for Publication of Quantitative Real-Time PCR Experiments guidelines, to rapidly detect, identify and quantify the human pathogenic bacteriaStenotrophomonas maltophilia, Burkholderia species, Pseudomonas aeruginosa and Serratia marcescenswhich are commonly associated with the biocontamination of water and water distribution systems. The specificity of the 5-plex assay was tested against genomic DNA isolated from a panel of 95 microorganisms with no cross reactivity observed. The analytical sensitivities of the S. maltophilia, B. cepacia, P. aeruginosa and the S. marcescens assays are 8.5, 5.7, 3.2 and 7.4 genome equivalents respectively. Subsequently, an analysis of HPW supplied by a Millipore Elix 35 water purification unit performed using standard microbiological methods revealed high levels of naturally occurring microbiological contamination. Five litre water samples from this HPW delivery system were also filtered and genomic DNA was purified directly from these filters. These DNA samples were then tested using the developed multiplex real-time PCR NAD assay and despite the high background microbiological contamination observed, both S. maltophilia andBurkholderia species were quantitatively detected and identified. At both sampling points the levels of both S. maltophilia and Burkholderia species present was above the threshold of 10 cfu/100 ml recommended by both EU and US guidelines. Conclusions: The novel culture independent methodology described in this study allows for rapid (<5 h), quantitative detection and identification of these four human pathogens from biocontaminated water and HPW distribution systems. We propose that the described NAD assay and associated methodology could be applied to routine testing of water and HPW distribution systems to assure microbiological safety and high water quality standards

    Examination of access and equity by gender, race and ethnicity in a non-traditional leadership development program in the United States

    In developing the next generation of school leadership, school districts across the United States and internationally must consider who is being promoted, the training they are able to access beyond traditional university degree work, the schools in which these emerging leaders enter their first principalships, and how prepared these new leaders are to succeed and remain in the role. This study explores international literature regarding school leader, particularly new leader, development and placement. The study discusses what is happening internationally in terms of the gender distribution of school leaders and the literature of non-traditional leadership development. To explore gender, race, and ethnicity in hiring and promotion practices, the study employs the methodology of case study analysis. This case study analysis formed from a need to understand the changing dynamics of race, gender, and ethnicity in school leadership in four American school districts participating in a non-traditional regional leadership development program. These districts, in the Richmond, Virginia (U.S.A.) metro area, are experiencing changing student demographics by race, ethnicity, and socioeconomic status. Specifically, the study addresses the demographic profile of those seeking leadership, the changing demographics of these communities, and the professional assignment of participating early career leaders

    Valuing Breastfeeding: Health Care Professionals’ Experiences of Delivering a Conditional Cash Transfer Scheme for Breastfeeding in Areas With Low Breastfeeding Rates

    Alongside a randomized controlled trial testing the effectiveness of offering a cash transfer scheme (shopping vouchers) to mothers in areas with low breastfeeding rates, qualitative interviews were conducted with health care professionals delivering the scheme to explore their experiences. Health care professionals (n = 34; mainly midwives and health visitors) were interviewed in depth. Transcripts from recorded interviews were analyzed using a Framework Analysis approach. There was widespread acceptance of the scheme by health care professionals, with prior concerns regarding bribery and coercion being quickly allayed. Health care professionals reported that the scheme fitted in well with their routine ways of promoting and endorsing breastfeeding. They described their experiences of women’s positive reaction toward the scheme and how the scheme encouraged breastfeeding and gave breastfeeding higher value. Health care professionals reported that the incentives helped them engage women and promote and support breastfeeding in areas with low breastfeeding rates

    The Adolescent Transition Program (ATP) is a multi-component preventive intervention designed to reduce escalation in problem behaviors among high-risk young adolescents. A previous evaluation of this program showed that aggregating high-risk youths for intervention purposes led to an escalation in delinquency and smoking. The results of this study also suggested that an intervention targeting parenting practices was more beneficial. These findings led to the development of a new intervention program aimed at the modification of parenting practices. This school-based program proposes a multiple gating approach to parent intervention with each level of intervention building on the previous one to reduce the overall prevalence of risk. A pilot study designed to evaluate the implementation of this intervention program suggested that schools seem to be an appropriate setting for reaching parents of high-risk adolescents and delivering intervention services

    Plausible responses to the threat of rapid sea-level rise for the Thames Estuary

    This paper considers the perceptions and responses of selected stakeholders to a scenarion of rapid rise in sea-level due to the collapse of the West Antarctic ice sheet, which could produce a global rise in sea-level of 5 to 6 metres. Through a process of dialogue involving one-to one interviews and a one-day policy exercise, we addressed influences on decision-making when information is uncertain and our ability to plan, prepare for and implement effective ways of coping with this extreme scenario. Through these interactions we hoped to uncover plausible responses to the scenario and identify potential weaknesses in our current flood management approaches to dealing with such an occurrence. By undertaking this exploratory exercise we hoped to find out whether this was a feasible way to deal with such a low probability but high consequence scenario. It was the process of finding a solution that interested us rather than the technical merits of one solution over another. We were not intending to produce definitive set of recommendations on how to respond but to gain insights into the process of making a decision, specifically what influences it and what assumptions are made.Sea level rise, London

    Developments in contact lens measurement:a comparative study of industry standard geometric inspection and optical coherence tomography

    Purpose: The aim of this study was to compare a developmental optical coherence tomography (OCT) based contact lens inspection instrument to a widely used geometric inspection instrument (Optimec JCF), to establish the capability of a market focused OCT system. Methods: Measurements of 27 soft spherical contact lenses were made using the Optimec JCF and a new OCT based instrument, the Optimec is830. Twelve of the lenses analysed were specially commissioned from a traditional hydrogel (Contamac GM Advance 49%) and 12 from a silicone hydrogel (Contamac Definitive 65), each set with a range of back optic zone radius (BOZR) and centre thickness (CT) values. Three commercial lenses were also measured; CooperVision MyDay (Stenfilcon A) in −10D, −3D and +6D powers. Two measurements of BOZR, CT and total diameter were made for each lens in temperature controlled saline on both instruments. Results: The results showed that the is830 and JCF measurements were comparable, but that the is830 had a better repeatability coefficient for BOZR (0.065 mm compared to 0.151 mm) and CT (0.008 mm compared to 0.027 mm). Both instruments had similar results for total diameter (0.041 mm compared to 0.044 mm). Conclusions: The OCT based instrument assessed in this study is able to match and improve on the JCF instrument for the measurement of total diameter, back optic zone radius and centre thickness for soft contact lenses in temperature controlled saline

    Poly(ADP-ribose) polymerase 1 (PARP-1) is a multidomain multifunctional nuclear enzyme involved in the regulation of the chromatin structure and transcription. PARP-1 consists of three functional domains: the N-terminal DNA-binding domain (DBD) containing three zinc fingers, the automodification domain (A), and the C-terminal domain, which includes the protein interacting WGR domain (W) and the catalytic (Cat) subdomain responsible for the poly(ADP ribosyl)ating reaction. The mechanisms coordinating the functions of these domains and determining the positioning of PARP-1 in chromatin remain unknown. Using multiple deletional isoforms of PARP-1, lacking one or another of its three domains, as well as consisting of only one of those domains, we demonstrate that different functions of PARP-1 are coordinated by interactions among these domains and their targets. Interaction between the DBD and damaged DNA leads to a short-term binding and activation of PARP-1. This hit and run activation of PARP-1 initiates the DNA repair pathway at a specific point. The long-term chromatin loosening required to sustain transcription takes place when the C-terminal domain of PARP-1 binds to chromatin by interacting with histone H4 in the nucleosome. This long-term activation of PARP-1 results in a continuous accumulation of pADPr, which maintains chromatin in the loosened state around a certain locus so that the transcription machinery has continuous access to DNA. Cooperation between the DBD and C-terminal domain occurs in response to heat shock (HS), allowing PARP-1 to scan chromatin for specific binding sites

    What young people want from health-related online resources: a focus group study

    The growth of the Internet as an information source about health, particularly amongst young people, is well established. The aim of this study was to explore young people's perceptions and experiences of engaging with health-related online content, particularly through social media websites. Between February and July 2011 nine focus groups were facilitated across Scotland with young people aged between 14 and 18 years. Health-related user-generated content seems to be appreciated by young people as a useful, if not always trustworthy, source of accounts of other people's experiences. The reliability and quality of both user-generated content and official factual content about health appear to be concerns for young people, and they employ specialised strategies for negotiating both areas of the online environment. Young people's engagement with health online is a dynamic area for research. Their perceptions and experiences of health-related content seem based on their wider familiarity with the online environment and, as the online environment develops, so too do young people's strategies and conventions for accessing it
