Maintenance of head and neck tumor on-chip: gateway to personalized treatment?
Aim: Head and neck squamous cell carcinomas (HNSCC) are solid tumors with low overall survival (40â60%). In a move toward personalized medicine, maintenance of tumor biopsies in microfluidic tissue culture devices is being developed. Methodology/ results: HNSCC (n = 15) was dissected (5â10 mg) and either analyzed immediately or cultured in a microfluidic device (37°C) for 48 h. No difference was observed in morphology between pre- and postculture specimens. Dissociated samples were analyzed using trypan blue exclusion (viability), propidium iodide flow cytometry (death) and MTS assay (proliferation) with no significant difference observed highlighting tissue maintenance. Computational fluid dynamics showed laminar flow within the system. Conclusion: The microfluidic culture system successfully maintained HNSCC for 48 h, the culture system will allow testing of different treatment modalities with response monitoring. Lay abstract: Head and neck cancers often have a poor treatment outcome. In order to study the response of the tissue, a miniaturized culture system has been developed to keep a small piece of tumor alive. In the current study, we show that small pieces of cancer tissue can be maintained in the system, using tissue structure and viability of single cells as a guide. In future work, patient equivalent treatments can be applied to these microculture systems to investigate individual patient tumor responses, which could help to guide treatment selection
Interface mapping in two-dimensional random lattice models
We consider two disordered lattice models on the square lattice: on the
medial lattice the random field Ising model at T=0 and on the direct lattice
the random bond Potts model in the large-q limit at its transition point. The
interface properties of the two models are known to be related by a mapping
which is valid in the continuum approximation. Here we consider finite random
samples with the same form of disorder for both models and calculate the
respective equilibrium states exactly by combinatorial optimization algorithms.
We study the evolution of the interfaces with the strength of disorder and
analyse and compare the interfaces of the two models in finite lattices.Comment: 7 pages, 6 figure
Circadian pattern and burstiness in mobile phone communication
The temporal communication patterns of human individuals are known to be
inhomogeneous or bursty, which is reflected as the heavy tail behavior in the
inter-event time distribution. As the cause of such bursty behavior two main
mechanisms have been suggested: a) Inhomogeneities due to the circadian and
weekly activity patterns and b) inhomogeneities rooted in human task execution
behavior. Here we investigate the roles of these mechanisms by developing and
then applying systematic de-seasoning methods to remove the circadian and
weekly patterns from the time-series of mobile phone communication events of
individuals. We find that the heavy tails in the inter-event time distributions
remain robustly with respect to this procedure, which clearly indicates that
the human task execution based mechanism is a possible cause for the remaining
burstiness in temporal mobile phone communication patterns.Comment: 17 pages, 12 figure
FADS3 is a Î14Z sphingoid base desaturase that contributes to gender differences in the human plasma sphingolipidome.
Sphingolipids (SLs) are structurally diverse lipids that are defined by the presence of a long-chain base (LCB) backbone. Typically, LCBs contain a single Î4E double bond (DB) (mostly d18:1), whereas the dienic LCB sphingadienine (d18:2) contains a second DB at the Î14Z position. The enzyme introducing the Î14Z DB is unknown. We analyzed the LCB plasma profile in a gender-, age-, and BMI-matched subgroup of the CoLaus cohort (n = 658). Sphingadienine levels showed a significant association with gender, being on average âŒ30% higher in females. A genome-wide association study (GWAS) revealed variants in the fatty acid desaturase 3 (FADS3) gene to be significantly associated with the plasma d18:2/d18:1 ratio (p = -log 7.9). Metabolic labeling assays, FADS3 overexpression and knockdown approaches, and plasma LCB profiling in FADS3-deficient mice confirmed that FADS3 is a bona fide LCB desaturase and required for the introduction of the Î14Z double bond. Moreover, we showed that FADS3 is required for the conversion of the atypical cytotoxic 1-deoxysphinganine (1-deoxySA, m18:0) to 1-deoxysphingosine (1-deoxySO, m18:1). HEK293 cells overexpressing FADS3 were more resistant to m18:0 toxicity than WT cells. In summary, using a combination of metabolic profiling and GWAS, we identified FADS3 to be essential for forming Î14Z DB containing LCBs, such as d18:2 and m18:1. Our results unravel FADS3 as a Î14Z LCB desaturase, thereby disclosing the last missing enzyme of the SL de novo synthesis pathway
Associations between plant density and yield components using different sowing times in wheat (Triticum aestivum L.)
The yield potential of wheat depends not only on genetic Ă environmental interactions, but also on various agronomic factors such as sowing date or the seed rate used for sowing. The main aim of this work was to determine possible correlations between the effects of different sowing dates and plant densities on the yield components of a collection of 48 wheat genotypes. Two-way analysis of variance on the data revealed that both sowing date and plant density, as main components, only had a minor effect on the yield component patterns. Correlation analysis, however, indicated that the sowing date had a greater effect on the yield components, while plant density was in closer correlation with the heading time (r = 0.90). The patterns determined for individual yield components at two different sowing dates and plant densities showed significant differences for spike length, spike fertility, grain number in the main spike, number of productive tillers, grain number on side tillers, mean grain number and grain weight. Genotypes that carry the winter (recessive) alleles of genes regulating vernalisation processes (VRN-A1, VRN-B1, VRN-D1) and the sensitive (recessive) alleles of the two genes responsible for photoperiod sensitivity (PPD-B1, PPD-D1) may have better tillering and consequently higher grain yield, though this may depend greatly on the year
Circadian patterns of Wikipedia editorial activity: A demographic analysis
Wikipedia (WP) as a collaborative, dynamical system of humans is an
appropriate subject of social studies. Each single action of the members of
this society, i.e. editors, is well recorded and accessible. Using the
cumulative data of 34 Wikipedias in different languages, we try to characterize
and find the universalities and differences in temporal activity patterns of
editors. Based on this data, we estimate the geographical distribution of
editors for each WP in the globe. Furthermore we also clarify the differences
among different groups of WPs, which originate in the variance of cultural and
social features of the communities of editors
Message-Passing Methods for Complex Contagions
Message-passing methods provide a powerful approach for calculating the
expected size of cascades either on random networks (e.g., drawn from a
configuration-model ensemble or its generalizations) asymptotically as the
number of nodes becomes infinite or on specific finite-size networks. We
review the message-passing approach and show how to derive it for
configuration-model networks using the methods of (Dhar et al., 1997) and
(Gleeson, 2008). Using this approach, we explain for such networks how to
determine an analytical expression for a "cascade condition", which determines
whether a global cascade will occur. We extend this approach to the
message-passing methods for specific finite-size networks (Shrestha and Moore,
2014; Lokhov et al., 2015), and we derive a generalized cascade condition.
Throughout this chapter, we illustrate these ideas using the Watts threshold

Local variation of hashtag spike trains and popularity in Twitter
We draw a parallel between hashtag time series and neuron spike trains. In
each case, the process presents complex dynamic patterns including temporal
correlations, burstiness, and all other types of nonstationarity. We propose
the adoption of the so-called local variation in order to uncover salient
dynamics, while properly detrending for the time-dependent features of a
signal. The methodology is tested on both real and randomized hashtag spike
trains, and identifies that popular hashtags present regular and so less bursty
behavior, suggesting its potential use for predicting online popularity in

Characteristics of cardiorespiratory output determining factors among 11â19-year-old boys at rest and during maximal load: Its impact on systolic hypertension
As consequence of the expansion of sedentary lifestyle among schoolchildren the prevalence of particular symptoms related to decreased cardiorespiratory fitness increases. The purpose of this study was twofolds, on one hand to compare boys in three developmental groups: second childhood (G1), puberty (G2), young adult (G3) and on the other hand to compare groups classified on resting systolic blood pressure (RSBP) to differentiate cardiorespiratory output determining factors both at rest and at maximal load. Randomly selected apparently healthy boys were assessed, all subjects (n = 282) performed an incremental treadmill test until fatigue. Heart rate (HR), systolic and diastolic blood pressure (SBP and DBP), and oxygen consumption were measured. Resting HR was higher and resting SBP and DBP were lower in the G1 as compared to G2 and G3 (p < 0.05) but not differed at maximal loads. However indicators of cardiovascular load differed between groups. The oxygen pulse and Q were the lowest in the G1 and increased significantly between groups (p < 0.05). In conclusion based on our data we can suggest that there is an observable development of hypertension associated with maturation and cardiac output determining factors
