    Prevalence of clinical autistic traits within a homeless population: barriers to accessing homeless services

    Recent research suggests a high prevalence rate of Autism Spectrum Conditions (ASC) amongst the homeless population. Although, it is well-documented that autistic people experienced many barriers to accessing health services, little is known about their challenges in accessing homeless services. Thus, the present study aimed to measure prevalence of high levels of autistic traits, and to identify barriers that prevent autistic people accessing homeless services. Participants recruited from homeless services (n = 65) completed the Autism Quotient-10 (AQ-10) alongside a questionnaire regarding perceived accessibility of homeless services. Results revealed that 18.5% of participants scored Above the Clinical Threshold of the AQ-10 (ACT-AQ). Moreover, the ACT-AQ group reported that encountering big groups in shared accommodation represent a significant barrier to engaging with homeless services. Further research is needed to identify the full degree of ASC representation and the factors that might prevent autistic homeless people accessing homeless services, and thus overcoming homelessness

    Animal-Assisted Interventions: Relationship Between Standards and Qualifications

    Ethical practice of animal- assisted interventions (AAI) requires appropriate qualifications and experience for all parties involved. Recently introduced and updated Standards of Practice emphasize the importance of AAI- specific training and qualification for different types of AAI, which should only be delivered within the scope of one’s professional competence. The purpose of this study was (1) to explore how a self- selected group of practitioners delivering AAI describe their work in relation to recent attempts to develop a terminological consensus (IAHAIO, 2014, 2018), and (2) to describe how AAI best practice recommendations (AAII, 2018) are implemented into professional practice among these AAI professionals working with dogs. The study employed an online survey that identified the participants’ qualification level, AAI specific training background, level of AAI experience, and their compliance with recommended guidelines on the documentation and measurement of clients’ progress. Data was collected from an international sample of 239 AAI professionals. A high proportion of respondents who were practitioners in a given field reported holding the necessary basic academic qualification for delivering animal- assisted education (AAE) (82.1%) or animal- assisted therapy (AAT) (91.4%). A substantial proportion delivered specific types of AAI that were beyond their scope of professional practice. A large proportion of respondent practitioners reported that they do not document (27.5%) or measure (28.5%) their clients’ progress as recommended by the professional standards. Experience in AAI was not a significant predictor of compliance with the recommended guidelines on documentation and measurement of clients’ progress; however, professional experience was related to their AAI experience. It is suggested that limitations in the provision of AAI- specific training and awareness of recently updated best practice recommendations influence the quality of AAI across practitioners. Overcoming these issues is essential to further professionalize AAI practice and enhance the quality of intervention programs

    Mechanism of completion of peptidyltransferase centre assembly in eukaryotes.

    During their final maturation in the cytoplasm, pre-60S ribosomal particles are converted to translation-competent large ribosomal subunits. Here, we present the mechanism of peptidyltransferase centre (PTC) completion that explains how integration of the last ribosomal proteins is coupled to release of the nuclear export adaptor Nmd3. Single-particle cryo-EM reveals that eL40 recruitment stabilises helix 89 to form the uL16 binding site. The loading of uL16 unhooks helix 38 from Nmd3 to adopt its mature conformation. In turn, partial retraction of the L1 stalk is coupled to a conformational switch in Nmd3 that allows the uL16 P-site loop to fully accommodate into the PTC where it competes with Nmd3 for an overlapping binding site (base A2971). Our data reveal how the central functional site of the ribosome is sculpted and suggest how the formation of translation-competent 60S subunits is disrupted in leukaemia-associated ribosomopathies.Bloodwise, MRC, Wellcome Trus

    Acetate supplementation modulates brain histone acetylation and decreases interleukin-1β expression in a rat model of neuroinflammation

    <p>Abstract</p> <p>Background</p> <p>Long-term acetate supplementation reduces neuroglial activation and cholinergic cell loss in a rat model of lipopolysaccharide-induced neuroinflammation. Additionally, a single dose of glyceryl triacetate, used to induce acetate supplementation, increases histone H3 and H4 acetylation and inhibits histone deacetylase activity and histone deacetylase-2 expression in normal rat brain. Here, we propose that the therapeutic effect of acetate in reducing neuroglial activation is due to a reversal of lipopolysaccharide-induced changes in histone acetylation and pro-inflammatory cytokine expression.</p> <p>Methods</p> <p>In this study, we examined the effect of a 28-day-dosing regimen of glyceryl triacetate, to induce acetate supplementation, on brain histone acetylation and interleukin-1β expression in a rat model of lipopolysaccharide-induced neuroinflammation. The effect was analyzed using Western blot analysis, quantitative real-time polymerase chain reaction and enzymic histone deacetylase and histone acetyltransferase assays. Statistical analysis was performed using one-way analysis of variance, parametric or nonparametric when appropriate, followed by Tukey's or Dunn's post-hoc test, respectively.</p> <p>Results</p> <p>We found that long-term acetate supplementation increased the proportion of brain histone H3 acetylated at lysine 9 (H3K9), histone H4 acetylated at lysine 8 and histone H4 acetylated at lysine 16. However, unlike a single dose of glyceryl triacetate, long-term treatment increased histone acetyltransferase activity and had no effect on histone deacetylase activity, with variable effects on brain histone deacetylase class I and II expression. In agreement with this hypothesis, neuroinflammation reduced the proportion of brain H3K9 acetylation by 50%, which was effectively reversed with acetate supplementation. Further, in rats subjected to lipopolysaccharide-induced neuroinflammation, the pro-inflammatory cytokine interleukin-1β protein and mRNA levels were increased by 1.3- and 10-fold, respectively, and acetate supplementation reduced this expression to control levels.</p> <p>Conclusion</p> <p>Based on these results, we conclude that dietary acetate supplementation attenuates neuroglial activation by effectively reducing pro-inflammatory cytokine expression by a mechanism that may involve a distinct site-specific pattern of histone acetylation and histone deacetylase expression in the brain.</p

    Child-display interaction: Exploring avatar-based touchless gestural interfaces

    During the last decade, touchless gestural interfaces have been widely studied as one of the most promising interaction paradigms in the context of pervasive displays. In particular, avatars and silhouettes have been proved to be effective in communicating the touchless gestural interactivity supported by displays. In the paper, we take a child-display interaction perspective by exploring avatar-based touchless gestural interfaces. We believe that large displays offer an opportunity to stimulate child experience and engagement, for instance when learning about art, as well as bringing a number of challenges. The purpose of this study is twofold: 1) identifying the relevant aspects of children's interactions with a large display based on a touchless avatar-based interface, and 2) understanding the impact on recalling the content that arises from the interaction. We engaged 107 children over a period of five days during a public event at the university premises.     Dog owner mental health is associated with dog behavioural problems, dog care and dog-facilitated social interaction: a prospective cohort study

    Despite numerous qualitative and cross-sectional studies investigating how dog-related factors may impact owners’ well-being, empirical studies to test these causal effects are lacking. This prospective cohort study examined the correlation and potential causal effect of 17 dog-related factors with six well-being outcomes (depression, anxiety, loneliness, suicidal ideation, hedonic well-being and eudaimonic well-being) in dog owners. Over a four-week period, 709 adult dog owners reported their weekly well-being and occurrence of each dog-related factor (e.g. how many times they ran with their dogs). A random intercept cross-lagged panel model (RI-CLPM) with significance threshold set at 0.001 was used. Six factors correlated with poorer owner well-being (i.e. aggressive dog behaviour, fearful dog behaviour, poor dog health, failure to provide for the dog, lack of control over the dog, and dog presence). Only ‘friendly conversation with others due to the dog’ correlated with better well-being. Purposeful reductions in the frequency of dog behavioural and health-related issues are likely to improve owner well-being, as well as greater consistency in dog care (i.e. provide for the dog) and more engagement in friendly dog-facilitated social interactions. No potential causal effects were significant. Further studies investigating causal relationships are essential to improve people’s well-being through dog ownership

    Potential Psychosocial Explanations for the Impact of Pet Ownership on Human Well-Being: Evaluating and Expanding Current Hypotheses

    There is a set of hypotheses commonly used in the literature to explain how pets affect human well-being (e.g., pets as social catalysts). Many studies are reported as giving results consistent with one or more of these, but they may not appear to be sufficient to explain the impacts of several pet-related activities on owner well-being. Confirmation bias may also overshadow the consideration of alternative mechanisms. This report aims to review and evaluate a range of psychosocial hypotheses that might help to explain how pets affect the well-being of their owners. This included a theoretical testing of the hypotheses against the recurring themes which emerged from four previously published qualitative frameworks relating to pet-related activities and their well-being outcomes. Twelve psychosocial hypotheses were generated and evaluated using this process: (1) social catalyst-repellent, (2) emotional contagion and empathy, (3) social support, (4) biophilia, (5) attributed fault, (6) social norms, (7) annoyance by noises, (8) routine, (9) caring, (10) exercise, (11) learning, and (12) affective touch. Only three presented potentially contradictory evidence (i.e., social catalyst-repellent, routine, and caring hypotheses), but closer examination revealed that these could not be rejected. These twelve hypotheses are a source of reference for a broader consideration of how pets might affect human well-being. Researchers are encouraged to use, test and/or challenge these hypotheses using established methods of scientific falsification in order to identify which are of the most important in relation to specific owner well-being outcomes.</p
