    Performance in Sharing Economy: Evidence from Room-Sharing Service

    Under the rapid development of sharing economy, performance is important for the prosperity of this innovative commercial model. However, the performance in sharing economy is less discussed in the academic field. Revenue and occupancy rate are widely applied as two ideal measures of performance in hospitality market. This study tries to fill the research gaps with both two measurements. Based on cue utilization theory, we explored the influence of listing cues and host cues on performance of listings and hosts in a representative Chinese room-sharing platform--XiaoZhu.com. The findings indicate that both listing and host cues have significant effects on performance. It’s expected that this study discusses a number of implications and makes contributions for researchers and practitioners

    Synergistic up-regulation of CXCL10 by virus and IFN Îł in human airway epithelial cells.

    Airway epithelial cells are the first line of defense against viral infections and are instrumental in coordinating the inflammatory response. In this study, we demonstrate the synergistic stimulation of CXCL10 mRNA and protein, a key chemokine responsible for the early immune response to viral infection, following treatment of airway epithelial cells with IFN Îł and influenza virus. The synergism also occurred when the cells were treated with IFN Îł and a viral replication mimicker (dsRNA) both in vitro and in vivo. Despite the requirement of type I interferon (IFNAR) signaling in dsRNA-induced CXCL10, the synergism was independent of the IFNAR pathway since it wasn't affected by the addition of a neutralizing IFNAR antibody or the complete lack of IFNAR expression. Furthermore, the same synergistic effect was also observed when a CXCL10 promoter reporter was examined. Although the responsive promoter region contains both ISRE and NFÎșB sites, western blot analysis indicated that the combined treatment of IFN Îł and dsRNA significantly augmented NFÎșB but not STAT1 activation as compared to the single treatment. Therefore, we conclude that IFN Îł and dsRNA act in concert to potentiate CXCL10 expression in airway epithelial cells via an NFÎșB-dependent but IFNAR-STAT independent pathway and it is at least partly regulated at the transcriptional level

    Utilization of direct smears of thyroid fine‐needle aspirates for ancillary molecular testing: A comparison of two proprietary testing platforms

    Background Ancillary molecular testing has been recommended for thyroid fine‐needle aspirates (FNA) with indeterminate cytologic diagnoses. Rosetta Genomics and Interpace Diagnostics have developed assays that can utilize direct smears as the testing substrate. Methods A retrospective study of indeterminate thyroid FNAs with known histologic follow‐up was performed. One Diff‐Quik‐stained smear and one Papanicolaou‐stained smear with similar cellularity (at least 60‐100 lesional cells) from each case were sent to Rosetta and Interpace, respectively, for analysis. The results were directly compared and correlated with the final histopathology. Neither company was aware of the follow‐up histologic findings in these cases. Results A total of 10 thyroid FNAs were identified from our 2015 files. The cytologic diagnoses included follicular lesion of undetermined significance (FLUS, n = 5), follicular neoplasm/suspicious for follicular neoplasm (FN/SFN, n = 4), and suspicious for malignancy (SM, n = 1). Of the seven cases with benign histology, six smears were classified as benign by the RosettaGX microRNA classifier, and one case was designated as suspicious. Five cases were negative by both ThyGenX oncogene panel and ThyraMIR microRNA classifier. One case was negative by ThyGenX and positive on follow‐up ThyraMIR, and one case was positive for KRAS mutation and positive on ThyraMIR. Both the RosettaGX and ThyGenX/ThyraMIR tests demonstrated positive results for the three histologically malignant cases. Conclusion This study demonstrates that two molecular testing platforms performed equally well using our stained direct smears. Both molecular tests revealed a 100% negative predictive rate. RosettaGX showed a 75% positive predictive value in comparison to 60% for ThyGenX/ThyraMIR

    Efficient Path Planning in Narrow Passages via Closed-Form Minkowski Operations

    Path planning has long been one of the major research areas in robotics, with PRM and RRT being two of the most effective classes of path planners. Though generally very efficient, these sampling-based planners can become computationally expensive in the important case of "narrow passages". This paper develops a path planning paradigm specifically formulated for narrow passage problems. The core is based on planning for rigid-body robots encapsulated by unions of ellipsoids. The environmental features are enclosed geometrically using convex differentiable surfaces (e.g., superquadrics). The main benefit of doing this is that configuration-space obstacles can be parameterized explicitly in closed form, thereby allowing prior knowledge to be used to avoid sampling infeasible configurations. Then, by characterizing a tight volume bound for multiple ellipsoids, robot transitions involving rotations are guaranteed to be collision-free without traditional collision detection. Furthermore, combining the stochastic sampling strategy, the proposed planning framework can be extended to solving higher dimensional problems in which the robot has a moving base and articulated appendages. Benchmark results show that, remarkably, the proposed framework outperforms the popular sampling-based planners in terms of computational time and success rate in finding a path through narrow corridors and in higher dimensional configuration spaces

    Semicontinuous Bioreactor Production of Recombinant Butyrylcholinesterase in Transgenic Rice Cell Suspension Cultures.

    An active and tetrameric form of recombinant butyrylcholinesterase (BChE), a large and complex human enzyme, was produced via semicontinuous operation in a transgenic rice cell suspension culture. After transformation of rice callus and screening of transformants, the cultures were scaled up from culture flask to a lab scale bioreactor. The bioreactor was operated through two phases each of growth and expression. The cells were able to produce BChE during both expression phases, with a maximum yield of 1.6 mg BChE/L of culture during the second expression phase. Cells successfully regrew during a 5-day growth phase. A combination of activity assays and Western blot analysis indicated production of an active and fully assembled tetramer of BChE

    Quantitative analysis of several random lasers

    We prescribe the minimal set of experimental data and parameters that should be reported for random-laser experiments and models. This prescript allows for a quantitative comparison between different experiments, and for a criterion whether a model predicts the outcome of an experiment correctly. In none of more than 150 papers on random lasers that we found these requirements were fulfilled. We have nevertheless been able to analyze a number of published experimental results and recent experiments of our own. Using our method we determined that the most intriguing property of the random laser (spikes) is in fact remarkably similar for different random lasers.Comment: 3 pages, 1 figur

    Differentiation of primate primordial germ cell-like cells following transplantation into the adult gonadal niche.

    A major challenge in stem cell differentiation is the availability of bioassays to prove cell types generated in vitro are equivalent to cells in vivo. In the mouse, differentiation of primordial germ cell-like cells (PGCLCs) from pluripotent cells was validated by transplantation, leading to the generation of spermatogenesis and to the birth of offspring. Here we report the use of xenotransplantation (monkey to mouse) and homologous transplantation (monkey to monkey) to validate our in vitro protocol for differentiating male rhesus (r) macaque PGCLCs (rPGCLCs) from induced pluripotent stem cells (riPSCs). Specifically, transplantation of aggregates containing rPGCLCs into mouse and nonhuman primate testicles overcomes a major bottleneck in rPGCLC differentiation. These findings suggest that immature rPGCLCs once transplanted into an adult gonadal niche commit to differentiate towards late rPGCs that initiate epigenetic reprogramming but do not complete the conversion into ENO2-positive spermatogonia

    Primary Osteoarthritis Early Joint Degeneration Induced by Endoplasmic Reticulum Stress Is Mitigated by Resveratrol

    Increasing numbers of people are living with osteoarthritis (OA) due to aging and obesity, creating an urgent need for effective treatment and preventions. Two top risk factors for OA, age and obesity, are associated with endoplasmic reticulum (ER) stress. The I-ERS mouse, an ER stress-driven model of primary OA, was developed to study the role of ER stress in primary OA susceptibility. The I-ERS mouse has the unique ability to induce ER stress in healthy adult articular chondrocytes and cartilage, driving joint degeneration that mimics early primary OA. In this study, ER stress-induced damage occurred gradually and stimulated joint degeneration with OA characteristics including increased matrix metalloproteinase activity, inflammation, senescence, chondrocyte death, decreased proteoglycans, autophagy block, and gait dysfunction. Consistent with human OA, intense exercise hastened and increased the level of ER stress-induced joint damage. Notably, loss of a critical ER stress response protein (CHOP) largely ameliorated ER stress-stimulated OA outcomes including preserving proteoglycan content, reducing inflammation, and relieving autophagy block. Resveratrol diminished ER stress-induced joint degeneration by decreasing CHOP, TNFα, IL-1ÎČ, MMP-13, pS6, number of TUNEL-positive chondrocytes, and senescence marker p16 INK4a. The finding, that a dietary supplement can prevent ER stressed-induced joint degeneration in mice, provides a preclinical foundation to potentially develop a prevention strategy for those at high risk to develop OA
