    Antioxidants and myocardial infarction : the EURAMIC study

    This thesis reports the background, design and results of a multi-centre study on the relationship between diet-derived antioxidants and the risk of acute myocardial infarction (MI) in men. Levels of α-tocopherol and ÎČ-carotene in adipose tissue and of selenium in toenails were compared between almost 700 patients with first MI and a similar number of control subjects, recruited in 8 European countries and Israel. The concentration of ÎČ-carotene in adipose tissue, expressed in quintiles of the distribution in controls, was inversely associated with the risk of MI (p for trend 0.001), independently of other risk factors. This association was strongest in current cigarette smokers and in subjects with a high proportion of polyunsaturated fatty acids in the adipose tissue. The risk of MI was not related to α-tocopherol in adipose tissue. In persons with low vitamin E levels, low toenail selenium was associated with a 2.5-fold increased risk of MI, compared to high selenium levels. An additional study among 85 healthy, non-smoking volunteers, aged 50-70, showed only a modest correlation of adipose tissue concentrations with dietary intake of α-tocopherol and ÎČ-carotene assessed by a food frequency questionnaire.Randomized controlled trials with varying doses and combinations of antioxidant nutrients should clarify whether the observed associations are causal. Until then, supplement use is not recommended, but a generous consumption of fruits and vegetables may be encouraged

    We reveal the enigmatic origin of one of the earliest surviving botanical collections. The 16th-century Italian En Tibi herbarium is a large, luxurious book with c. 500 dried plants, made in the Renaissance scholarly circles that developed botany as a distinct discipline. Its Latin inscription, translated as “Here for you a smiling garden of everlasting flowers”, suggests that this herbarium was a gift for a patron of the emerging botanical science. We follow an integrative approach that includes a botanical similarity estimation of the En Tibi with contemporary herbaria (Aldrovandi, Cesalpino, “Cibo”, Merini, Estense) and analysis of the book’s watermark, paper, binding, handwriting, Latin inscription and the morphology and DNA of hairs mounted under specimens. Rejecting the previous origin hypothesis (Ferrara, 1542–1544), we show that the En Tibi was made in Bologna around 1558. We attribute the En Tibi herbarium to Francesco Petrollini, a neglected 16th-century botanist, to whom also belongs, as clarified herein, the controversial “Erbario Cibo” kept in Rome. The En Tibi was probably a work on commission for Petrollini, who provided the plant material for the book. Other people were apparently involved in the compilation and offering of this precious gift to a yet unknown person, possibly the Habsburg Emperor Ferdinand I. The En Tibi herbarium is a Renaissance masterpiece of art and science, representing the quest for truth in herbal medicine and botany. Our multidisciplinary approach can serve as a guideline for deciphering other anonymous herbaria, kept safely “hidden” in treasure rooms of universities, libraries and museums

    Risicobeoordeling blauwalgen in zwemwater : Nieuwe technieken voor de bepaling van de aanwezigheid van blauwalgtoxines

    Waterbeheerders voeren op grond van de Europese Zwemwaterrichtlijn analyses uit om in te schatten hoe groot het kans is op het voorkomen van blauwalgen(toxines) in zwemwater. Ze krijgen daarmee een beeld van het gezondheidsrisico voor zwemmers, op basis waarvan er zwemadviezen worden gegeven. De technieken die daarvoor nu worden gebruikt, meten alleen de hoeveelheden blauwalgen, maar niet de toxines die blauwalgen kunnen produceren, terwijl dat nu juist de ziekteverwekkende stoffen zijn. In dit onderzoek zijn de huidige technieken vergeleken met nieuwe technieken. Het gaat om technieken die direct(er) toxines bepalen. Hiervoor zijn in 2019 in totaal 103 watermonsters (afkomstig van 11 verschillende locaties) geanalyseerd met meerdere methoden. De uitkomsten zijn met elkaar vergeleken, waarbij de analyse van toxines met LC-MS/MS als 'gouden standaard’ is genomen. Het uiteindelijke doel is om een (kosten)effectieve analysetechniek te vinden die het meest geschikt is om het gezondheidsrisico voor zwemmers in kaart te brengen

    Adipose Tissue Omega-3 and Omega-6 Fatty Acid Content and Breast Cancer in the EURAMIC Study

    The fatty acid content of adipose tissue in postmenopausal breast cancer cases and controls from five European countries in the European Community Multicenter Study on Antioxidants, Myocardial Infarction, and Cancer (EURAMIC) breast cancer study (1991 -1992) was used to explore the hypothesis that fatty acids of the omega-3 family inhibit breast cancer and that the degree of inhibition depends on background levels of omega-6 polyunsaturates. Considered in isolation, the level of omega-3 or omega-6 fat in adipose tissue displayed little consistent association with breast cancer across study centers. The ratio of long-chain omega-3 fatty acids to total omega-6 fat showed an inverse association with breast cancer in four of five centers. In Malaga, Spain, the odds ratio for the highest tertile relative to the lowest reached 0.32 (95% confidence interval 0.13-0.82). In this center, total omega-6 fatty acid was strongly associated with breast cancer. With all centers pooled, the odds ratio for long-chain omega-3 to total omega-6 reached 0.80 for the second tertile and 0.65 for the third tertile, a downward trend bordering on statistical significance (p for trend = 0.055). While not definitive, these results provide evidence for the hypothesis that the balance between omega-3 and omega-6 fat may play a rolein breast cancer. Am J Epidemiol 1998; 147: 342-5

    Case study on the efficacy of a lanthanum-enriched clay (PhoslockÂź) in controlling eutrophication in Lake Het Groene Eiland (The Netherlands)

    Lake Het Groene Eiland was created in the beginning of 2008 by construction of dikes for isolating it from the surrounding 220-ha water body. This so-called claustrum of 5 ha was treated using lanthanum-modified clay (PhoslockÂź) to control eutrophication and mitigate cyanobacterial nuisance. Cyanobacteria chlorophyll-a were significantly lower in the claustrum than those in the reference water body, where a massive bloom developed in summer, 2008. However, PO4-P and TP did not statistically differ in these two waters. TN and NO3-N were significantly lower in the claustrum, where dense submerged macrophytes beds developed. Lanthanum concentrations were elevated after the applications of the modified clay in the claustrum, but filterable lanthanum dropped rapidly below the Dutch standard of 10.1 ÎŒg l−1. During winter, dozens of Canada geese resided at the claustrum. Geese droppings contained an average of 2 mg PO4-P g−1 dry weight and 12 mg NH3-N g−1 dry weight and might present a growing source of nutrients to the water. Constructing the claustrum enabled unrestricted bathing in subsequent three summers, as no swimming bans had to be issued due to cyanobacteria blooms. However, the role of the modified clay in this positive outcome remains unclear, and longevity of the measures questionable.

    Lycopene and Myocardial Infarction Risk in the EURAMIC Study

    A multicenter case-control study was conducted to evaluate the relations between antioxidant status assessed by biomarkers and acute myocardial infarction. Incidence cases and frequency matched controls were recruited from 10 European countries to maximize the variance in exposure within the study. Adipose tissue needle aspiration biopsies were taken shortly after the infarction and analyzed for levels of carotenoids and tocopherols. An examination of colinearity including all covariates and the three carotenoids, α-carotene, ÎČ-carotene, and lycopene, showed that the variables were sufficiently independent to model simultaneously. When examined singularly, each of the carotenoids appeared to be protective. Upon simultaneous analyses of the carotenoids, however, using conditional logistic regression models that controlled for age, body mass index, socioeconomic status, smoking, hypertension, and maternal and paternal history of disease, lycopene remained independently protective, with an odds ratio of 0.52 for the contrast of the 10th and 90th percentiles (95% confidence interval 0.33-0.82, p= 0.005). The associations for α- and ÎČ-carotene were largely eliminated.
