    Deligne Pairings and Discriminants of Algebraic Varieties

    Let V be a finite dimensional complex vector space and V^* its dual and let X in P(V) be a smooth projective variety of dimension n and degree d at least two. For a generic n-tuple of hyperplanes H_1,...,H_n in P(V^*)^n, the intersection of X with H_1,...,H_n consists of d distinct points. We define the "discriminant of X", to be the the set D(X) of n-tuples for which the set-theoretic intersection is not equal to d points. Then D(X) is a hyper surface in P(V^*)^n and the set of defining polynomials, which is a one-dimensional vector space, is called the "discriminant line". We show that this line is canonically isomorphic to the Deligne pairing where K is the canonical line bundle of X and L is the restriction of the hyperplane bundle to X. As a corollary, we obtain a generalization of Paul"s formula which relates the Mabuchi K-energy on the space of Bergman metrics to \Delta(X), the "hyperdiscriminant of X"

    Grain boundary mobility in anion doped MgO

    Certain anions OH(-), F(-) and Gl(-) are shown to enhance grain growth in MgO. The magnitude of their effect decreases in the order in which the anions are listed and depends on their location (solid-solution, second phase) in the MgO lattice. As most anions exhibit relatively high vapor pressures at sintering temperatures, they retard densification and invariably promote residual porosity. The role of anions on grain growth rates was studied in relation to their effect on pore mobility and pore removal; the atomic process controlling the actual rates was determined from observed kinetics in conjunction with the microstructural features. With respect to controlling mechanisms, the effects of all anions are not the same. OH(-) and F(-) control behavior through creation of a defect structure and a grain boundary liquid phase while Cl(-) promotes matter transport within pores by evaporation-condensation. Studies on an additional anion, S to the minus 2nd power gave results which were no different from undoped MgO, possibly because of evaporative losses during hot pressing. Hence, the effect of sulphur is negligible or undetermined

    Glycan shifting on hepatitis C virus (HCV) E2 glycoprotein is a mechanism for escape from broadly neutralizing antibodies

    Hepatitis C virus (HCV) infection is a major cause of liver disease and hepatocellular carcinoma. Glycan shielding has been proposed to be a mechanism by which HCV masks broadly neutralizing epitopes on its viral glycoproteins. However, the role of altered glycosylation in HCV resistance to broadly neutralizing antibodies is not fully understood. Here, we have generated potent HCV neutralizing antibodies hu5B3.v3 and MRCT10.v362 that, similar to the previously described AP33 and HCV1, bind to a highly conserved linear epitope on E2. We utilize a combination of in vitro resistance selections using the cell culture infectious HCV and structural analyses to identify mechanisms of HCV resistance to hu5B3.v3 and MRCT10.v362. Ultra deep sequencing from in vitro HCV resistance selection studies identified resistance mutations at asparagine N417 (N417S, N417T and N417G) as early as 5 days post treatment. Comparison of the glycosylation status of soluble versions of the E2 glycoprotein containing the respective resistance mutations revealed a glycosylation shift from N417 to N415 in the N417S and N417T E2 proteins. The N417G E2 variant was glycosylated neither at residue 415 nor at residue 417 and remained sensitive to MRCT10.v362. Structural analyses of the E2 epitope bound to hu5B3.v3 Fab and MRCT10.v362 Fab using X-ray crystallography confirmed that residue N415 is buried within the antibody–peptide interface. Thus, in addition to previously described mutations at N415 that abrogate the β-hairpin structure of this E2 linear epitope, we identify a second escape mechanism, termed glycan shifting, that decreases the efficacy of broadly neutralizing HCV antibodies

    Active yellow pages: a pipelined resource management architecture for wide-area network computing

    This paper describes a novel, pipelined resource management architecture for computational grids. The design is based on two key realizations. One is that resource management involves a sequence of tasks that is best handled by a pipeline. As shown in the paper, this approach results, in a scalable architecture for decentralized scheduling. The other realization is that static aggregation of resources for improved scheduling is inadequate in wide-area computing environments because the needs of users and jobs change with both, location and time. The described architecture addresses this problem by dynamically aggregating resources in a manner that continuously optimizes system response. This is accomplished by way of an active yellow pages directory that allows aggregation constraints to be (re)defined on the fly. An initial prototype of the active yellow pages service has been deployed in the PUNCH network computing environment. Experiences with the production PUNCH system and preliminary results from controlled experiments indicate that the active yellow pages service performs well.Peer Reviewe

    Understanding ethnic inequalities in hearing health in the UK: a cross-sectional study of the link between language proficiency and performance on the Digit Triplet Test

    Introduction Research using the UK Biobank data has shown ethnic inequalities in hearing health; however, the hearing test used may exhibit a disadvantage for non-native language speakers. Objectives To validate the results of the UK Biobank hearing test (Digit Triplet Test, DTT) against self-reported measures of hearing in the dataset and create classifications of hearing health. To observe if language proficiency and migration age have the same effect on hearing health classification as on the DTT in isolation. Our hypothesis is that language proficiency acts differently on the DTT, demonstrating that the DTT is biased for non-native speakers of English. Design Latent classes representing profiles of hearing health were identified from the available hearing measures. Factors associated with class membership were tested using multinomial logistic regression models. Ethnicity was defined as (1) White, native English-speaking, (2) ethnic minority, arrived in the UK aged 12. Participants The UK Biobank participants with valid hearing test results and associated covariates (N=151 268). Outcome measures DTT score, self-reported hearing difficulty, self-reported hearing difficulty in noise and hearing aid use. Results Three classes of hearing health were found: ‘normal’, ‘generally poor’ and ‘only subjectively poor’. In a model adjusting for known confounders of hearing loss, a poor or insufficient hearing test result was less likely for those with better language (OR 0.69, 95% CI 0.65 to 0.74) or numerical ability (OR 0.71, 95% CI 0.67 to 0.75) but more likely for those having migrated aged >12 (OR 3.85, 95% CI 3.64 to 4.07). Conclusions The DTT showed evidence of bias, having greater dependence on language ability and migration age than other hearing indicators. Designers of future surveys and hearing screening applications may wish to consider the limitations of speech-in-noise tests in evaluating hearing acuity for populations that include non-native speakers
