Macrophage exosomes as natural nanocarriers for protein delivery to inflamed brain

Recent work has stimulated interest in the use of exosomes as nanocarriers for delivery of small drugs, RNAs, and proteins to the central nervous system (CNS). To overcome the blood-brain barrier (BBB), exosomes were modified with brain homing peptides that target brain endothelium but likely to increase immune response. Here for the first time we demonstrate that there is no need for such modification to penetrate the BBB in mammals. The naïve macrophage (Mϕ) exosomes can utilize, 1) on the one hand, the integrin lymphocyte function-associated antigen 1 (LFA-1) and intercellular adhesion molecule 1 (ICAM-1), and, 2) on the other hand, the carbohydrate-binding C-type lectin receptors, to interact with brain microvessel endothelial cells comprising the BBB. Notably, upregulation of ICAM-1, a common process in inflammation, promotes Mϕ exosomes uptake in the BBB cells. We further demonstrate in vivo that naïve Mϕ exosomes, after intravenous (IV) administration, cross the BBB and deliver a cargo protein, the brain derived neurotrophic factor (BDNF), to the brain. This delivery is enhanced in the presence of brain inflammation, a condition often present in CNS diseases. Taken together, the findings are of interest to basic science and possible use of Mϕ-derived exosomes as nanocarriers for brain delivery of therapeutic proteins to treat CNS diseases

Alcohol consumption increases health risks and social consequences. It also lowers productivity resulting in economic losses for drinkers and the rest of society. To investigate alcohol drinking behavior and to estimate economic cost incurred by alcohol users in Khon Kaen province in 2007. A cross-sectional survey targeting the population aged 12-65 years old was conducted in 20 communities. Data were collected using full-structured questionnaires through interviews. Among 1,053 respondents, 53.0% drank alcohol sometime in their lives (95% CI: 46.1, 59.9). The percentage of individuals drinking in the past 12 months was 43.3% (95% CI: 37.1, 49.5). The average number of drinking days in past 12 months was 36.8 days. Most respondents drank for social activities, mainly with friends and relatives. Individual costs of alcohol consumption varied greatly. The weighted average cost in 2007 was 975.5 Baht per drinker. The estimated overall cost of alcohol consumption in Khon Kaen, in 2007, was 691.2 million Baht (95% CI: 280.0, 1,102.3 million), or 502.9 Baht per capita. More than half of the Khon Kaen population drank alcohol sometime in their lives and 43.3% were current drinkers. The average number of drinking days in past 12 months was 36.8 days. The estimated cost of alcohol consumption in Khon Kaen province was enormous.link_to_subscribed_fulltex

Investigation of Home Energy Management with Advanced Direct Load Control and Optimal Scheduling of Controllable Loads

Due to the rapid changes in the energy situation on a global scale, the amount of RES installed using clean renewable energy sources such as Photovoltaic (PV) and Wind-power Generators (WGs) is rapidly increasing. As a result, there has been a great deal of research aimed at promoting the adoption of renewable energy. Research on Demand-side Management (DSM) has also been important in promoting the adoption of RES. However, the massive introduction of PV has changed the shape of the demand curve for electricity, which significantly impacts the operational planning of thermal generators. Therefore, this paper proposes an Advanced Direct Load Control (ADLC) model to temporarily shutdown the electric connection between the power grid and Smart Houses (SHs). The most important feature of the proposed model is that it temporarily shuts down the electric connection with the power grid. The shutdown is performed twice to increase the load demand during daytime hours and reduce the peak load during night-time hours. The proposed model also promotes the self-consumption of the generated power during the shutdown period, which is expected to reduce the operating cost. This paper considers six case studies for SH, and the operational costs and carbon dioxide emissions are compared and discussed. The results show that the SH with ADLC successfully reduces the operating costs and carbon dioxide emissions
