Diffusion and activation of n-type dopants in germanium
The diffusion and activation of -type impurities (P and As) implanted into
-type Ge(100) substrates were examined under various dose and annealing
conditions. The secondary ion mass spectrometry profiles of chemical
concentrations indicated the existence of a sufficiently high number of
impurities with increasing implanted doses. However, spreading resistance probe
profiles of electrical concentrations showed electrical concentration
saturation in spite of increasing doses and indicated poor activation of As
relative to P in Ge. The relationships between the chemical and electrical
concentrations of P in Ge and Si were calculated, taking into account the
effect of incomplete ionization. The results indicated that the activation of P
was almost the same in Ge and Si. The activation ratios obtained experimentally
were similar to the calculated values, implying insufficient degeneration of
Ge. The profiles of P in Ge substrates with and without damage generated by Ge
ion implantation were compared, and it was clarified that the damage that may
compensate the activated -type dopants has no relationship with the
activation of P in Ge.Comment: 6 pages, 4 figure
A Novel Role for Niemann-Pick Disease Type 2C Protein in Papillae Formation
BACKGROUND: Despite the presence of papillary structures and papillary tumors in humans, the mechanism of papillae formation is unknown. We describe herein a novel role for Niemann-Pick disease type 2C (NPC2) protein, a cholesterol binding protein in the lysosome, in papillae formation. METHODOLOGY/PRINCIPAL FINDING: We examined NPC2 protein expression in surgical samples of papillary tissues by immunohistochemical stain, and all papillary tissues expressed NPC2 protein in the epithelium. To examine our hypothesis of NPC2 protein-mediated papillae formation, we carried out xenograft experiments using wild H460 cells (large cell lung carcinoma cell line) that constitutively expressed abundant NPC2 protein and NPC2 protein-depleted H460 cells by NPC2 shRNA. The xenografts of wild H460 cells and empty shRNA vector cells showed distinct papillae formation, whereas NPC2 protein-depleted H460 cells displayed markedly reduced or no papillae. Since all papillary tissues have open spaces we examined whether NPC2 protein might also contribute to the creation of open spaces. The TUNEL assay in the xenografts of wild and empty shRNA vector H460 cells showed massive cell death, and NPC2 protein-depleted cells displayed minimal cell death. Measurement of caspase 3/7 activities in cultured H460 cells supported NPC2 protein-mediated apoptotic cell death. The presence of excess NPC2 protein, however, did not always produce papillae as seen in the xenografts of CHO cells that were stably transfected with NPC2. CONCLUSIONS/SIGNIFICANCE: The NPC2 protein of certain cells forms papillae coupled with apoptosis that creates open space. This protein may have future applications to modulate papillae formation and papillary growth in tumor tissues
Chandra and ASCA Observations of the X-ray-brightest T-Tauri Stars in the Rho Ophiuchi Cloud
We present the Chandra ACIS and ASCA GIS results for a series of four
long-term observations on DoAr 21, ROXs 21 and ROXs 31; the X-ray brightest
T-Tauri stars (TTSs) in the Rho Ophiuchi cloud. In the four observations with a
net exposure of ~600 ksec, we found six, three and two flares from DoAr 21,
ROXs 21 and ROXs 31, respectively; hence the flare rate is fairly high. The
spectra of DoAr 21 are well fitted with a single-temperature plasma model,
while those of ROXs 21 and ROXs 31 need an additional soft plasma component.
Since DoAr 21 is younger (~10^5 yr) than ROXs 21 and ROXs 31 (~10^6 yr), these
results may indicate that the soft component gradually increases as T-Tauri
stars age. The abundances are generally sub-solar and vary from element to
element. Both high-FIP (first ionization potential) and low-FIP elements show
enhancement over the mean abundances. An unusual giant flare is detected from
ROXs 31. The peak luminosity and temperature are ~10^33 ergs s^-1 and ~10 keV,
respectively. The temperature reaches its peak value before the flux maximum,
and is nearly constant (4--5 keV) during the decay phase, indicating successive
energy release during the flare. The abundances and absorption show dramatic
variability from the quiescent to flare phase.Comment: 13 pages, 18 figures, accepted for publication in Ap
First Data Release of the Hyper Suprime-Cam Subaru Strategic Program
The Hyper Suprime-Cam Subaru Strategic Program (HSC-SSP) is a three-layered
imaging survey aimed at addressing some of the most outstanding questions in
astronomy today, including the nature of dark matter and dark energy. The
survey has been awarded 300 nights of observing time at the Subaru Telescope
and it started in March 2014. This paper presents the first public data release
of HSC-SSP. This release includes data taken in the first 1.7 years of
observations (61.5 nights) and each of the Wide, Deep, and UltraDeep layers
covers about 108, 26, and 4 square degrees down to depths of i~26.4, ~26.5, and
~27.0 mag, respectively (5sigma for point sources). All the layers are observed
in five broad bands (grizy), and the Deep and UltraDeep layers are observed in
narrow bands as well. We achieve an impressive image quality of 0.6 arcsec in
the i-band in the Wide layer. We show that we achieve 1-2 per cent PSF
photometry (rms) both internally and externally (against Pan-STARRS1), and ~10
mas and 40 mas internal and external astrometric accuracy, respectively. Both
the calibrated images and catalogs are made available to the community through
dedicated user interfaces and database servers. In addition to the pipeline
products, we also provide value-added products such as photometric redshifts
and a collection of public spectroscopic redshifts. Detailed descriptions of
all the data can be found online. The data release website is

Identification and structural analysis of C-terminally truncated collapsin response mediator protein-2 in a murine model of prion diseases
<p>Abstract</p> <p>Background</p> <p>Prion diseases are fatal neurodegenerative disorders that accompany an accumulation of the disease-associated form(s) of prion protein (PrPSc) in the central nervous system. The neuropathological changes in the brain begin with focal deposits of PrPSc, followed by pathomorphological abnormalities of axon terminal degeneration, synaptic loss, atrophy of dendritic trees, and eventual neuronal cell death in the lesions. However, the underlying molecular basis for these neuropathogenic abnormalities is not fully understood.</p> <p>Results</p> <p>In a proteomic analysis of soluble proteins in the brains of mice challenged intracerebrally with scrapie prion (Obihiro I strain), we found that the amount of the full-length form of collapsin response mediator protein-2 (CRMP-2; 61 kDa) decreased in the late stages of the disease, while the amount of its truncated form (56 kDa) increased to comparable levels observed for the full-length form. Detailed analysis by liquid chromatography-electrospray ionization-tandem mass spectrometry showed that the 56-kDa form (named CRMP-2-ΔC) lacked the sequence from serine518 to the C-terminus, including the C-terminal phosphorylation sites important for the regulation of axonal growth and axon-dendrite specification in developing neurons. The invariable size of the mRNA transcript in Northern blot analysis suggested that the truncation was due to post-translational proteolysis. By overexpression of CRMP-2-ΔC in primary cultured neurons, we observed the augmentation of the development of neurite branch tips to the same levels as for CRMP-2T514A/T555A, a non-phosphorylated mimic of the full-length protein. This suggests that the increased level of CRMP-2-ΔC in the brain modulates the integrity of neurons, and may be involved in the pathogenesis of the neuronal abnormalities observed in the late stages of the disease.</p> <p>Conclusions</p> <p>We identified the presence of CRMP-2-ΔC in the brain of a murine model of prion disease. Of note, C-terminal truncations of CRMP-2 have been recently observed in models for neurodegenerative disorders such as ischemia, traumatic brain injury, and Wallerian degeneration. While the structural identity of CRMP-2-ΔC in those models remains unknown, the present study should provide clues to the molecular pathology of degenerating neurons in prion diseases in connection with other neurodegenerative disorders.</p
The whole blood transcriptional regulation landscape in 465 COVID-19 infected samples from Japan COVID-19 Task Force
「コロナ制圧タスクフォース」COVID-19患者由来の血液細胞における遺伝子発現の網羅的解析 --重症度に応じた遺伝子発現の変化には、ヒトゲノム配列の個人差が影響する--. 京都大学プレスリリース. 2022-08-23.Coronavirus disease 2019 (COVID-19) is a recently-emerged infectious disease that has caused millions of deaths, where comprehensive understanding of disease mechanisms is still unestablished. In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection
DOCK2 is involved in the host genetics and biology of severe COVID-19
「コロナ制圧タスクフォース」COVID-19疾患感受性遺伝子DOCK2の重症化機序を解明 --アジア最大のバイオレポジトリーでCOVID-19の治療標的を発見--. 京都大学プレスリリース. 2022-08-10.Identifying the host genetic factors underlying severe COVID-19 is an emerging challenge. Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target
地震発生帯における深部掘削孔を用いた長期計測
Large earthquakes occur frequently in subduction zones. Most earthquakes are generated in the seismogenic zone, a fairly limited area confined to the shallower regions of the subduction plate boundary. To understand the processes of earthquake generation, it is essential to monitor the physical and mechanical properties of the seismogenic zone over long periods. At present, there are no deep borehole observations of the seismogenic zone more than 3km below seafloor, because it has, until now, been impossible to penetrate to such depths below the sea floor. The Integrated Ocean Drilling Program (IODP), scheduled to begin in 2003, plans to drill boreholes beneath the ocean floor using a multiple-drilling platform operation. The IODP riser-quipped drilling ship (Chikyu) enables the emplacement of boreholes up to 0km beneath the ocean floor, and will provide opportunities to conduct long-term deep borehole observations in the seismogenic zone. Long-term borehole observations in the seismogenic zone are expected to require the development of advanced sampling, monitoring, and recording technology. Here, we discuss the scientific objectives, engineering and technical challenges, and experimental design for a deep borehole, long-term deepborehole monitoring system aimed at understanding the processes of earthquake generation in the seismogenic zone of subduction plate boundaries. We focus specifically on the relationships between environmental conditions in the deep subsurface, details of monitoring and recording, and design and implementation of scientific tools and programs
The Hyper Suprime-Cam SSP survey: Overview and survey design
Hyper Suprime-Cam (HSC) is a wide-field imaging camera on the prime focus of the 8.2-m Subaru telescope on the summit of Mauna Kea in Hawaii. A team of scientists from Japan, Taiwan, and Princeton University is using HSC to carry out a 300-night multi-band imaging survey of the high-latitude sky. The survey includes three layers: the Wide layer will cover 1400 deg2 in five broad bands (grizy), with a 5 σ point-source depth of r ≈ 26. The Deep layer covers a total of 26 deg2 in four fields, going roughly a magnitude fainter, while the UltraDeep layer goes almost a magnitude fainter still in two pointings of HSC (a total of 3.5 deg2). Here we describe the instrument, the science goals of the survey, and the survey strategy and data processing. This paper serves as an introduction to a special issue of the Publications of the Astronomical Society of Japan, which includes a large number of technical and scientific papers describing results from the early phases of this survey
