Vitamin K dependent Îł-carboxylation in chronic kidney disease
Vascular calcification is present in atherosclerosis, ageing, chronic kidney diseases and diabetes and is strongly associated with an increased morbidity and mortality. Calcification of arteries occurs at the tunica intima and the tunica media. Thereby, vascular smooth muscles cells (VSMC) transdifferentiate into an osteoblastic phenotype. Modifiable calcification inhibitors were identified of which matrix gla protein (MGP) is regarded as the most potent one being expressed in the vascular wall. MGP gets posttranslationally gamma carboxylated at 5 glutamic acid residues, which achieve calcium binding properties. This carboxylation step requires reduced vitamin K as a cofactor. It is provided and recycled in the so called vitamin K cycle, which consists of the vitamin K epoxide reductase (VKOR), DT-diaphorase and Îł-glutamyl carboxylase (GGCX). The effects of uremia on vitamin K recycling via the vitamin K cycle are unknown. Besides MGP, other vitamin K dependent proteins are known. One is the Gas6 protein, which is also expressed by VSMC, but its role is not yet fully understood. Gas6 offers one n-terminal carboxylation site. Aim of this thesis was to characterize the activities of three enzymes of the vitamin K cycle and the role of vitamin K dependent Gas6 protein under uremic conditions. First, a fluorescence method for the quantification of GGCX activity in vitro in tissue samples was developed. This method employs a fluorescein isothiocyanate (FITC) labelled Glu containing hexapeptide which gets carboxylated by the GGCX. Second, the influences of uremia and pharmacological doses of vitamin K supplementation on the activity of the vitamin K cycle and extraosseous calcification were investigated. Uremia was induced in rats by adenine diet, in part supplemented with vitamin K1 or K2 for 4 or 7 weeks. The GGCX activity was decreased under uremic conditions possibly contributing to calcifications. The GGCX activity can be restored by vitamin K supplementation. Third, the influence of Gas6 protein on vascular calcification was investigated in murine in vitro VSMC culture and different in vivo models using a) Warfarin diet, b) uninephrectomy or c) electrocautery of the kidney as well d) ageing mice. No differences were found in tissue calcium content, echocardiography and biochemical serum and urine parameter between Gas6-/- mice and wildtype controls. This does not support a role of Gas6 in the pathogenesis of vascular calcification.Vitamin K abhĂ€ngige Gamma-Carboxylierung in der chronischen NIereninsuffizienz VaskulĂ€re Kalzifizierung tritt als eine Begleiterscheinung von Atherosklerose, Alter, chronischen Nierenerkrankungen und Diabetes auf und geht mit einer stark erhöhten MorbiditĂ€t und MortalitĂ€t einher. Arterielle Kalzifizierungen erfolgen in der Tunica intima und der Tunica media. Hier transdifferenzieren glatte GefĂ€Ămuskelzellen (vascular smooth muscle cells, VSMC) in einen osteoblastĂ€ren PhĂ€notyp. Regulierbare Verkalkungsinhibitoren sind bekannt, wie das Matrix Gla Protein (MGP), welches insbesondere in der GefĂ€Ăwand von VSMC exprimiert wird. MGP wird an 5 Glutamatresten posttranslational Îł-carboxyliert, wodurch eine Kalziumbindung ermöglicht wird. Zur Îł-Carboxylierung wird reduziertes Vitamin K als Cofaktor benötigt. Dieses wird im sogenannten Vitamin K Zyklus bereitgestellt und recycelt. Die beteiligten Enzyme sind die Vitamin K Epoxid-Reduktase (VKOR), DT-Diaphorase und Îł-glutamyl-Carboxylase (GGCX). Der Einfluss einer UrĂ€mie auf das Vitamin K Recycling im Vitamin K Zyklus ist unbekannt. Neben MGP sind weitere Vitamin K abhĂ€ngige Proteine bekannt. Hierzu zĂ€hlt auch das Gas6 Protein, welches ebenfalls von VSMC exprimiert wird, aber dessen Funktion noch nicht vollstĂ€ndig geklĂ€rt ist. Das Ziel dieser Arbeit war die Charakterisierung der EnzymaktivitĂ€ten im Vitamin K Zyklus und die Rolle des Vitamin K abhĂ€ngigen Gas6 Proteins in der experimentellen UrĂ€mie. Dazu wurde zunĂ€chst eine Fluoreszenz-gestĂŒtzte Methode entwickelt, zur Bestimmung der GGCX AktivitĂ€t in Gewebeproben. Verwendet wurde ein Fluorescein Isothiocanat (FITC) gekoppeltes Glu-haltiges Hexapeptid, welches durch die GGCX carboxyliert wird. Zweitens wurde der Einfluss einer UrĂ€mie sowie die Verabreichung pharmakologischer Dosen Vitamin K auf die Enzyme des Vitamin K Zyklus und extraossĂ€re Kalzifikation untersucht. Durch Gabe von Adenin ĂŒber einen Zeitraum von 4 oder 7 Wochen wurde in Ratten eine UrĂ€mie induziert, teilweise unter Supplementation mit Vitamin K1 oder K2. Die GGCX-AktivitĂ€t war unter urĂ€mischen Bedingungen erniedrigt, wodurch möglicherweise Kalzifizierungen getriggert werden. Die erniedrigte GGCX-AktivitĂ€t kann durch zusĂ€tzliche Vitamin K Gabe aufgehoben werden. Drittens wurde der Einfluss des Gas6 Proteins auf die GefĂ€Ăkalzifikation in murinen in vitro VSMC Kultur und in verschiedenen in vivo Modellen untersucht: a) Warfarin DiĂ€t b) Uninephrektomie c) Elektrokoagulation der Niere sowie d) alternde MĂ€use. Keine Unterschiede wurden festgestellt im Calciumgehalt des Gewebes, echokardiographisch und in den gemessenen Serum- und Urin-Parameteren. Diese Daten unterstĂŒtzen keine Rolle von Gas6 in der Pathogenese der vaskulĂ€ren Kalzifikation
Ancient communities in the fossil record: Implications for sedimentary modeling
In recent years the thinking of paleontologists has become increasingly oriented toward the biologic sciences and less toward sedimentology and stratigraphy. For this reason advances in paleontology in the past three decades have not found their way into the repertoire of some sedimentologists. This is perhaps especially true of those who are most heavily involved with sedimentologic modeling. Organisms respond to changes in the environment, but they also contribute to these changes; thus information about them needs to be incorporated into sedimentary models. The advances in paleontology that are important for modeling sedimentologists fall into three categories: the data of paleontology, interactions between fossils and paleoenvironments, and sources of permanent change through time. Interpreting the data of paleontology has been advanced by improvements in the understanding of taphonomy, time averaging, and the temporal resolution of the stratigraphic record. Interactions between fossils and their paleoenvironments can be expressed in terms of succession and diversity, both of which can be incorporated into sedimentary models. Some biologic communities contain highly interactive groups of species. The structure of such communities has typically been attributed to competition, but competition theory has recently been criticized by many ecologists, leaving community paleoecology somewhat up in the air. Studies of communities, however, remain applicable to paleoenvironmental analysis. Permanent changes through time are typically large-scale or long-term events that are rather easily incorporated into sedimentary models. They include organic evolution, community evolution, and extinction

Die Weltgesellschaft in der Perspektive der Zivilisationstheorie
Die vorliegende Arbeit beschÀftigt sich mit einem heute sehr verbreiteten Thema, der Globalisierung. Die Globalisierungsforschung sowie die neuere Literatur zur Weltgesellschaft werden nÀmlich systematisch im Lichte der Zivilisationstheorie von Norbert Elias gedeutet, um einen Bezugsrahmen zu formulieren, in dem bisherige Ergebnisse der Globalisierungsdebatte und Weltgesellschaftsforschung akkumuliert werden. Diese Arbeit ist dabei keine typische Globalisierungsuntersuchung mit entsprechender Analyse und Forderungen. Sie ist auch keine typische Anwendung der Zivilisationstheorie. Es wird vielmehr versucht, die Zivilisationstheorie theoretisch zu vertiefen und zu erweitern.
Die vorliegende Arbeit unterteilt sich in vier Kapitel: Das erste Kapitel liefert einen Ăberblick der Globalisierungsdebatte und zum Begriff der Weltgesellschaft. Daran schlieĂt sich im zweiten Kapitel die Ausarbeitung der Kernthese mit der Ausweitung des âProzessmodells auf mehreren Ebenenâ und dem Konzept der Machtbalance auf die Analyse der Weltgesellschaft an. Zur Erweiterung der Zivilisationstheorie werden hier die Konzepte der âModellierungâ und âĂberlappungâ eingefĂŒhrt. Im dritten und vierten Kapitel werden die Themen Entwicklung und Gesellschaft der Individuen anhand der Theoriediskussion und der Rekonstruktion des Bezugsrahmens diskutiert. Sie sind fĂŒr die Untersuchung der Globalisierung bzw. Weltgesellschaft und zugleich fĂŒr die Weiterentwicklung der Zivilisationstheorie zentrale Themen. Im abschlieĂenden Fazit schlieĂlich wird ein Ausblick auf die mögliche weitere Anwendung des neuen Instrumentariums gewagt.
Der hier vorgestellte analytische Bezugsrahmen vermag solchen empirisch orientierten Forschungsfragen aber eine Orientierungshilfe darzustellen. Der Bezugsrahmen dient also als erster Schritt der Weiterentwicklung der Globalisierung/Weltgesellschaftsforschung und der Zivilisationstheorie in Richtung einer globalen Soziologie
