    Přeměna aktivního cytostatika na jeho méně aktivní metabolit - sekundární alkohol, je jedním z důvodů vzniku anthracyklinové rezistence v nádorové terapii. Tohoto procesu se nejčastěji účastní karbonylredukující enzymy, hlavně z nadrodiny aldo-ketoreduktas a dehydrogenas/reduktas s krátkým řetězcem. Proto je aktuální tematikou inhibice těchto enzymů, a tím zvýšení účinnosti anthracyklinů v protinádorové terapii. V této diplomové práci byl zkoumán vliv inhibitoru tirabrutinibu na vybrané lidské karbonylredukující enzymy in vitro ve spojení s cytostatikem daunorubicinem.The conversion of active cytostatic drug to less active metabolite, secondary alcohol, is one of the reasons of development of anthracycline resistance in oncotherapy. Most often carbonyl reducing enzymes are involved in this process, mainly from aldo-ketoreductase and short-chain dehydrogenase/reductase superfamilies. Therefore, the inhibition of above-mentioned enzymes and thereby increase of anthracyclines efficiency in therapy is a very actual topic. In this work the influence of the inhibitor tirabrutinib on human carbonyl reducing enzymes converting daunorubicin in vitro was evaluated.Fakulta chemicko-technologická1.     Dental care of patients after surgical therapy of inborn oro-facial clefts 3D technologies in diagnostics and therapy of cleft patients

    Patients with orofacial defects, inborn or acquired, isolated or in combination with other handicaps in syndroms, suffer from various problems including psychological problems. Morphological changes within these handicaps burden not only the patients themselves, but also their families. Among the main problems are difficulties with feeding, speech or breathing. Anomalies in number or position of deciduous and permanent teeth, development of jaws and dental arches are often present. Multidisciplinary aproach is necessary when solving all of the tasks and the multidiciplinary team consits of experts from field of plastic surgery, foniatry, speach therapist, dentist and oral surgeons. Surgical therapy involves early primary reconstruction of cleft lip and causes necessity of very detailed therapy planing and evaluation. It is very important to create systematic aproach to dental care including valuace diagnostic methods. In the work presented there will be variol methods of diagnosis and evaluation of the treatment introduced with the aim of evaluating modern methods including 3D printing and digital technologies and a valid alternative to traditional methods (plaster casts). It describes this type of diet as an alternative treatment for many diseases. It focused on the issue of inadequate intake of nutritions, which are important for the body.     Cholinesterasy (ChEs) jsou skupinou enzymů, které katalyzují hydrolýzu acetylcholinu na cholin a kyselinu octovou. Jedná se o základní proces umožňující obnovu cholinergního receptoru. Mezi skupinu esteras se řadí acetylcholinesterasa (AChE) a butyrylcholinesterasa (BChE). Oba enzymy mají z 65 % stejnou sekvenční homologii aminokyselin a jsou kódovány různými geny na lidských chromozomech. Tyto dva enzymy se navzájem liší substrátovou specifitou a místem exprese. Inhibitory cholinesteras (galantamin, donepezil a rivastigmin), které zvyšují cholinergní neurotransmisy, jsou využívány pro snížení příznaků Alzheimerovy choroby inhibitory Brutonovy tyrosinkinasy (BTK) (tirabrutinib, evobrutinib a branebrutinib) snižují aktivaci fosfolipasy γ 2, která byla identifikována jako genetický rizikový faktor Alzheimerovy choroby.Cholinesterases (ChEs) are a group of enzymes that catalyze the hydrolysis of acetylcholine to choline and acetic acid. This is a basic process enabling the recovery of the cholinergic receptor. The group of esters includes acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). Both enzymes share 65% amino acid sequence homology and are encoded by different genes on human chromosomes. These two enzymes differ from each other in substrate specificity and site of expression. Cholinesterase inhibitors (galantamine, donepezil and rivastigmine), which increase cholinergic neurotransmissions, are used to reduce the symptoms of Alzheimer's disease Bruton tyrosine kinase (BTK) inhibitors (tirabrutinib, evobrutinib and branebrutinib) reduce the activation of phospholipase γ 2, which has been identified as a genetic risk factor for Alzheimer's disease

    Intra-Oral 3D Scanning for the Digital Evaluation of Dental Arch Parameters

    Intra-oral scanning technology has brought a completely new approach to examination methods in dentistry. In comparison to traditional plaster casts, it allows more precise digital analysis of dental arch components during the treatment of dental disorders. Data acquired can also be used for the creation of three-dimensional (3D) models using 3D printers. This paper describes a data acquisition system, the mathematical processing of resulting three-dimensional model, and the statistical analysis of selected parameters of the dental arch. The study aims at comparing the results achieved from 20 models of the same individual acquired by different specialists. The proposed methodology includes mathematical rotation of objects into the optimal plane defined by the teeth tops’ location to increase the accuracy of the resulting parameters. The mean evaluated distances between canines (3-3) and premolars (5-5) are 24.29 mm and 37.98 mm, respectively, for the lower dental arch. The proposed object rotation increased these distance by 0.9% and 1.4%, respectively, reducing the rotation error. Moreover, the variability of results decreased and the mean value of the coefficient of variation was lowered by 12%. Image registration was then used to evaluate changes of dental arch parameters. This paper presents the advantages of digital models for visualisation of the dental arch allowing its flexible rotation and evaluation of its parameters
