    Development and optimization of system for measuring of watering

    Cílem této práce je seznámení se senzory a problematikou měření zálivky rostlin. Výstupem této práce je tvorba inteligentního prototypu modulu, který má měřit teplotu, vodivost a pH zálivky. Automatické měření těchto vlastností ulehčí a zrychlí vyhodnocování parametrů. Tento měřicí modul je rozšířením stávajícího zařízení o možnost měření vlastností zálivky rostlin.The main goal of this thesis is to become acquainted with questions of sensors and the problems of measuring the plant watering. The result is creating an intelligent prototype of a module that should measure temperature, conductivity and pH of the watering. Automatic measuring of these properties makes the parameter evaluation easier and quicker. This measuring module extends contemporary device with the possibility to measure properties of the plant watering.

    Tvorba a analýza mutantních myších modelů pro studium patofyziologické role KLK5 a KLK7 v pokožce

    Peptidázy příbuzné kallikreinu (KLK) představují rodinu vzájemně spřízněných serinových proteáz, které jsou kódovány geny umístěnými v jediném chromozomálním lokusu. Jejich exprese byla popsána v mnoha tkáních a řada experimentů in vitro naznačuje významnou roli KLK v mnoha fyziologických i patologických procesech. Přesto jsou skutečné úlohy KLK v živém organismu stále ne zcela objasněny, což je způsobeno zejména nedostatkem vhodných zvířecích modelů pro jejich studium. Přestože genově-deficientní myší modely pro některé KLK již byly vytvořeny, jejich význam pro objasnění rolí těchto proteáz v komplexním prostředí živého organismu byl jen omezený. Jednou z hlavních překážek je částečný funkční překryv mezi některými KLK. Inaktivace jednoho genu tudíž často nevede ke změně fenotypu v důsledku funkční kompenzace mezi jednotlivými proteázami. Zdá se tedy, že klasické myší modely deficientní pro jeden gen jsou ne zcela vhodným nástrojem pro studium KLK in vivo. V této práci jsme použili TAL efektor nukleázy (TALENy) pro přípravu několika nových mutantních myších linií, které umožňují analýzu komplexních proteolytických sítí tvořených KLK a studium jejich role ve zdravém organismu i během chorobných procesů. Although gene deficient mouse models were generated for several KLK genes, they had limited use for understanding the roles of individual proteases in the complex environment in vivo. One of the main obstacles which hampers in vivo analysis is partial functional overlap between some KLKs. This makes traditional single-gene deficient animal models an inadequate tool to address the biological impact of the gene deficiency as compensatory mechanisms often result in a lack of phenotype. In this work, we used the transcription activator-like effector nuclease (TALEN) technology to generate several novel mutant mouse models to study the complex KLK proteolytic pathways and their roles in healthy organism and in disease.     Processing of Emotion in Functional Neurological Disorder

    Emotions have traditionally been considered crucial in the development of functional neurological disorder, but the evidence underpinning this association is not clear. We aimed to summarize evidence for association between functional neurological disorder and emotions as formulated by Breuer and Freud in their conception of hysterical conversion. Based on a systematic literature search, we identified 34 controlled studies and categorized them into four groups: (i) autonomic arousal, (ii) emotion-motion interactions, (iii) social modulation of symptoms, and (iv) bodily awareness in FND. We found evidence for autonomic dysregulation in FND; convergent neuroimaging findings implicate abnormal limbic-motor interactions in response to emotional stimuli in FND. Our results do not provide enough empirical evidence for social modulation of the symptoms, but there is a clinical support for the role of suggestion and placebo in FND. Our results provide evidence for abnormal bodily awareness in FND. Based on these findings, we propose that functional neurological symptoms are forms of emotional reactions shaped into symptoms by previous experience with illness and possibly reinforced by actual social contexts. Additional research should investigate the effect of social context on the intensity of functional neurological symptoms and associated brain regions

    Generation of transgenic mouse model to study biological role of KLK5 in epidermis

    A number of studies provide strong evidence that KLK5 is one of the most important serine proteases in the epidermis and is involved in processes such as desquamation, processing of antimicrobial peptides or induction of inflammatory reaction. The role of KLK5 has been deduced from in vitro experiments and thus its functions should be verified in vivo. This work aimed to develop a specific tissue targeting strategy to study the role of murine kallikreins in the epidermal compartment in vivo and to generate a transgenic model overexpressing mKlk5 in the mouse epidermis. Using the modified promoter of human involucrin, transgenic mice expressing the fluorescent marker, tdTomato, were generated in the first step. This transgenic reporter mouse showed specific targeting pattern of the reporter in the upper epidermal layers and, thus, the modified involucrin promoter could be employed for targeting further gene of interest in the differentiated epidermal compartment. In the second step transgenic mouse lines expressing murine kallikrein 5 were successfully generated. Among them two lines exhibited approximately 6-9 fold overexpression at the mRNA and protein levels, however it appeared that the immature protease was not activated under normal healthy conditions.     My thesis is dealing with the social system of an organisation and its quality. It is divided in the theoretical and practical part. In the theoretical part I describe general aspects of an organisation concerning it a system of social relationships. I look at the development of an organisation in the course of time and I make an overview of the indicators and instruments used for measuring the system's quality. I applied my theoretical knowledge in the practical part. I measured the quality of social relationships with the indicators as social values and standards, corporate ethics and social climate. I consider them the most important in my thesis

    Příprava transgenní myši jako modelu pro studium role KLK5 v epidermis

    Klk5 patří mezi nejvýznamnější proteolytické enzymy v pokožce a podílí se na procesech jako je deskvamace, štěpení antimikrobiálních peptidů nebo indukce zánětlivé reakce. Role Klk5 v epidermis byla dosud popisována na základě experimentů in vitro, je proto nezbytné potvrdit jeho funkci in vivo. Tato práce si kladla za cíl vytvořit metodické postupy pro zacílení genové exprese myších kalikreinů do svrchní vrstvy pokožky a připravit transgenní myší model exprimující mKlk5 v pokožce. Prvním krokem byla příprava transgenních myší exprimujících fluorescenční marker tdTomato pod kontrolou upraveného involukrinového promotoru. Vytvořené transgenní myši vykazovaly expresi proteinu tdTomato výhradně ve svrchních vrstvách pokožky, čímž byla potvrzena vhodnost upraveného involukrinového promotoru pro cílení genové exprese do diferencovaných keratinocytů. Dále byly úspěšně připraveny transgenní myši exprimující mKlk5. Dvě transgenní linie vykazovaly 6-9 násobnou overexpresi mKlk5 na mRNA i proteinové úrovni v porovnání s wt zvířaty, transgenní mKlk5 byl ovšem za fyziologických podmínek přítomen pouze ve formě neaktivního zymogenu. Transgenní myši byly proto podrobeny dvěma modelům způsobujícím defekty v epidermální bariéře a potenciálně vedoucím aktivaci proteáz.
