    Public sector procurement and ethical trade: Governance and social responsibility in some hidden global supply chains

    This paper places a critical spotlight on the ways in which governance and social responsibility concerning labour standards work in the context of public sector procurement. Supply chains provisioning the public sector, incorporating a vast array of materials used in public services such as education, health, social housing and transportation, have been under-researched and under-theorized in the geographical and wider social science literature on Global Production Networks (GPNs), Global Value Chains (GVCs) and consumption ethics. Against this backdrop, the paper evaluates the emergence of ethical trading programmes involving labour standards in UK public sector sourcing, drawing on examples from the health, higher education, construction and transportation sectors and a methodology combining interviews with participant observation. Findings show that ethical public procurement is shaped, and often constrained, by: the relatively low profile of many materials used in public services, which challenge effective campaigning and advocacy work; the consequent limits to reputational risk for state departments and first-tier suppliers if labour issues are encountered in their supply chains; and the significant role of EU and UK procurement law, which until 2014 rendered social responsibility in public sector sourcing a legal risk to specify. Through a practice-orientated approach and adopting the notion of responsibilization inspired by governmentality perspectives, the paper grasps how these challenges for ethical public procurement are encountered and negotiated by procurement managers and those influencing them in a neoliberal environment. It is shown that ethical sourcing is significantly less advanced in the UK public sector than it is in consumer goods sectors, with implications for social justice in a whole realm of under-researched global supply chains

    Non-essential role for TLR2 and its signaling adaptor Mal/TIRAP in preserving normal lung architecture in mice

    Myeloid differentiation factor 88 (MyD88) and MyD88-adaptor like (Mal)/Toll-interleukin 1 receptor domain containing adaptor protein (TIRAP) play a critical role in transducing signals downstream of the Toll-like receptor (TLR) family. While genetic ablation of the TLR4/MyD88 signaling axis in mice leads to pulmonary cell death and oxidative stress culminating in emphysema, the involvement of Mal, as well as TLR2 which like TLR4 also signals via MyD88 and Mal, in the pathogenesis of emphysema has not been studied. By employing an in vivo genetic approach, we reveal here that unlike the spontaneous pulmonary emphysema which developed in Tlr42/2 mice by 6 months of age, the lungs of Tlr22/2 mice showed no physiological or morphological signs of emphysema. A more detailed comparative analysis of the lungs from these mice confirmed that elevated oxidative protein carbonylation levels and increased numbers of alveolar cell apoptosis were only detected in Tlr42/2 mice, along with up-regulation of NADPH oxidase 3 (Nox3) mRNA expression. With respect to Mal, the architecture of the lungs of Mal2/2 mice was normal. However, despite normal oxidative protein carbonylation levels in the lungs of emphysema-free Mal2/2 mice, these mice displayed increased levels of apoptosis comparable to those observed in emphysematous Tlr42/2 mice. In conclusion, our data provide in vivo evidence for the non-essential role for TLR2, unlike the related TLR4, in maintaining the normal architecture of the lung. In addition, we reveal that Mal differentially facilitates the anti-apoptotic, but not oxidant suppressive, activities of TLR4 in the lung, both of which appear to be essential for TLR4 to prevent the onset of emphysema

    The protein phosphatase 1 regulator NIPP1 is essential for mammalian spermatogenesis

    NIPP1 is one of the major nuclear interactors of protein phosphatase PP1. The deletion of NIPP1 in mice is early embryonic lethal, which has precluded functional studies in adult tissues. Hence, we have generated an inducible NIPP1 knockout model using a tamoxifen-inducible Cre recombinase transgene. The inactivation of the NIPP1 encoding alleles (Ppp1r8) in adult mice occurred very efficiently in testis and resulted in a gradual loss of germ cells, culminating in a Sertoli-cell only phenotype. Before the overt development of this phenotype Ppp1r8 -/- testis showed a decreased proliferation and survival capacity of cells of the spermatogenic lineage. A reduced proliferation was also detected after the tamoxifen-induced removal of NIPP1 from cultured testis slices and isolated germ cells enriched for undifferentiated spermatogonia, hinting at a testis-intrinsic defect. Consistent with the observed phenotype, RNA sequencing identified changes in the transcript levels of cell-cycle and apoptosis regulating genes in NIPP1-depleted testis. We conclude that NIPP1 is essential for mammalian spermatogenesis because it is indispensable for the proliferation and survival of progenitor germ cells, including (un)differentiated spermatogonia.publishe
