    Smoking influences the yield of dendritic cells for cancer immunotherapy

    Background: Dendritic cell (DC)-based vaccination is considered to be a potentially effective therapeutic strategy against advanced cancer. The aim of this study was to address the smoking history that might affect the preparation of DC vaccines in validated instructional manufacture. Materials and Methods: Data on mature DCs generated from 102 sessions of leukapheresis performed on 92 patients with advanced cancer or sarcoma were retrospectively evaluated and compared in relation to the data between their smoking history and the generation of DCs from these patients. 61 patients with adenocarcinoma, including 7 with lung, 10 with breast, 8 with stomach, 12 with colorectal, and 23 with pancreatic adenocarcinoma were enrolled. Results: The average yield of autologous DCs (15.5 ± 8.3x107) was thought to be dependent on the number of monocytes (124.2 ± 74.1x107) collected by leukapheresis. The average ratio of DCs/apheresed monocytes (DC/aM ratio) was lower in the smoker group (11.1 ± 7.2%) than that in the non-smoker group (17.2 ± 9.3%, p=0.001). The number of DCs and the DC/aM ratio were lower in the patients with gastric and pancreatic cancer than in those with adenocarcinoma of other sites.     Deformation transfer is a type of retargeting method that operates directly on the mesh and, by doing so, enables reuse of animation without setting up character rigs and a mapping between the source and target geometries. Deformation transfer can potentially reduce the costs of animation and give studios a competitive edge when keeping up with the latest computer animation technology. Unfortunately, deformation transfer has limitations and is yet to become standard practice in the industry. This survey starts by introducing Sumner and Popović’s [18] seminal work and highlights key issues for industry settings. We then review related work in sections, organized by these key issues. After surveying related work, we discuss how their advances open the door to several practical applications of deformation transfer. To conclude, we highlight areas of future work

    Abstract When examining the standard of the air-gap in open tanks with overflow pipes, it is necessary to determine the height of the highest water level and maximum capacity of overflow pipe in tanks. However, it is not cleared not only to determine the height and the capacity, but also to decide the size of overflow pipe. Therefore, the experimental study was carried out for the purpose to obtain them. In this paper, it was reported the experiment result concerning the FRP water tank with overflow pipe of inside diameters 50 mm and 100 mm. As a result, empirical formulae of the height by incoming flow rates and inside diameters, and the maximum flow rate of overflow pipes are shown. Moreover, methods to decide the size of overflow pipes are proposed

    Central Glucagon-like Peptide-1 Receptor Signaling via Brainstem Catecholamine Neurons Counteracts Hypertension in Spontaneously Hypertensive Rats

    Glucagon-like peptide-1 receptor (GLP-1R) agonists, widely used to treat type 2 diabetes, reduce blood pressure (BP) in hypertensive patients. Whether this action involves central mechanisms is unknown. We here report that repeated lateral ventricular (LV) injection of GLP-1R agonist, liraglutide, once daily for 15 days counteracted the development of hypertension in spontaneously hypertensive rats (SHR). In parallel, it suppressed urinary norepinephrine excretion, and induced c-Fos expressions in the area postrema (AP) and nucleus tractus solitarius (NTS) of brainstem including the NTS neurons immunoreactive to dopamine beta-hydroxylase (DBH). Acute administration of liraglutide into fourth ventricle, the area with easy access to the AP and NTS, transiently decreased BP in SHR and this effect was attenuated after lesion of NTS DBH neurons with anti-DBH conjugated to saporin (anti-DBH-SAP). In anti-DBH-SAP injected SHR, the antihypertensive effect of repeated LV injection of liraglutide for 14 days was also attenuated. These findings demonstrate that the central GLP-1R signaling via NTS DBH neurons counteracts the development of hypertension in SHR, accompanied by attenuated sympathetic nerve activity

    The Off State of GX 339-4

    We report BeppoSAX and optical observations of the black hole candidate GX 339-4 during its X-ray `off' state in 1999. The broad-band (0.8-50 keV) X-ray emission can be fitted by a single power law with spectral index, \alpha ~1.6. The observed luminosity is 6.6e33 erg s^{-1} in the 0.5-10 keV band, which is at the higher end of the flux distribution of black hole soft X-ray transients in quiescence, comparable to that seen in GS 2023+338 and 4U 1630-47. An optical observation just before the BeppoSAX observation shows the source to be very faint at these wavelengths as well (B=20.1, V=19.2). By comparing with previously reported `off' and low states (LS), we conclude that the `off' state is actually an extension of the LS, i.e. a LS at lower intensities. We propose that accretion models such as the advection-dominated accretion flows are able to explain the observed properties in such a state.Comment: Accepted for publication in MNRA

    Inversions of Levy Measures and the Relation Between Long and Short Time Behavior of Levy Processes

    The inversion of a Levy measure was first introduced (under a different name) in Sato 2007. We generalize the definition and give some properties. We then use inversions to derive a relationship between weak convergence of a Levy process to an infinite variance stable distribution when time approaches zero and weak convergence of a different Levy process as time approaches infinity. This allows us to get self contained conditions for a Levy process to converge to an infinite variance stable distribution as time approaches zero. We formulate our results both for general Levy processes and for the important class of tempered stable Levy processes. For this latter class, we give detailed results in terms of their Rosinski measures

    Increased S-nitrosylation and proteasomal degradation of caspase-3 during infection contribute to the persistence of adherent invasive escherichia coli (AIEC) in immune cells

    Adherent invasive Escherichia coli (AIEC) have been implicated as a causative agent of Crohn's disease (CD) due to their isolation from the intestines of CD sufferers and their ability to persist in macrophages inducing granulomas. The rapid intracellular multiplication of AIEC sets it apart from other enteric pathogens such as Salmonella Typhimurium which after limited replication induce programmed cell death (PCD). Understanding the response of infected cells to the increased AIEC bacterial load and associated metabolic stress may offer insights into AIEC pathogenesis and its association with CD. Here we show that AIEC persistence within macrophages and dendritic cells is facilitated by increased proteasomal degradation of caspase-3. In addition S-nitrosylation of pro- and active forms of caspase-3, which can inhibit the enzymes activity, is increased in AIEC infected macrophages. This S-nitrosylated caspase-3 was seen to accumulate upon inhibition of the proteasome indicating an additional role for S-nitrosylation in inducing caspase-3 degradation in a manner independent of ubiquitination. In addition to the autophagic genetic defects that are linked to CD, this delay in apoptosis mediated in AIEC infected cells through increased degradation of caspase-3, may be an essential factor in its prolonged persistence in CD patients

    Accretion flow dynamics during the evolution of timing and spectral properties of GX 339-4 during its 2010-11 outburst

    The Galactic black-hole candidate GX 339-4 exhibited several outbursts at regular intervals of \sim 2-3 years in the Rossi X-ray Timing Explorer (RXTE) era. After remaining in an almost quiescent state for 3 years, it again became X-ray active in 2010 January, continuing to be so over the next \sim 14 months. We study the timing and spectral properties of the black hole candidate (BHC) during its recent outburst using RXTE PCA data, starting from 2010 January 12 to 2011 March 6. Our study provides a comprehensive understanding of the mass accretion processes and properties of the accretion disk of the black hole candidate. The PCA spectra of 2.5-25 keV are mainly fitted with a combination of two components, namely, a disk black body and a power-law. The entire outburst as observed by RXTE, is divided into 4 spectral states, namely, hard, hard-intermediate, soft-intermediate, and soft. Quasi-periodic oscillations (QPOs) were found in 3 out of the 4 states, namely hard, hard-intermediate, and soft-intermediate. The QPO frequencies increase monotonically from 0.102 Hz to 5.692 Hz in the rising phase of the outburst, while during the declining phase QPO frequencies decrease monotonically from 6.420 to 1.149 Hz. The recent outburst of GX 339-4 gives us an opportunity to understand the evolution of the two-component accretion rates starting from the onset to the end of the outburst phase. We found that the QPO frequency variation could be explained by the propagating oscillatory shock model (POS) and the hardness versus intensity variation can be reproduced if we assume that higher viscosity causes the conversion of a low angular momentum disk component into a Keplerian component during the outburst phase.
