    TiivistelmĂ€. TĂ€mĂ€n tutkielman tarkoitus on selvittÀÀ musiikinopettajien kĂ€sityksiĂ€ musiikillisesta improvisaatiosta ja musiikilliselle improvisaatiolle suotuisasta sosiaalisesta oppimisympĂ€ristöstĂ€ sekĂ€ sen rakentamisesta. Tutkielman tavoitteena on lisĂ€tĂ€ musiikinopettajien tietoisuutta sosiaalisen oppimisympĂ€ristön merkityksestĂ€ musiikillisen improvisaation kannalta. Teoreettisessa viitekehyksessĂ€ avataan sosiaaliseen toimintaan vaikuttavia rakenteellisia tekijöitĂ€: sosiaalisen kehityksen, sosiaalisen oppimisympĂ€ristön ja ryhmĂ€n kehityksen sekĂ€ ryhmĂ€dynamiikan kĂ€sitteitĂ€. LisĂ€ksi esitellÀÀn Engeströmin toiminnan teoria, joka havainnollistaa ihmisen toiminnan kehittymisen prosessia. TĂ€ssĂ€ tutkielmassa toiminnan teoria on sijoitettu musiikillisen improvisaation sosiaaliseen oppimisympĂ€ristöön. Musiikillista improvisaatiota avataan luovuuden ja ryhmĂ€improvisaation nĂ€kökulmista. Luovuuden nĂ€kökulmasta musiikillinen improvisaatio rinnastuu sĂ€veltĂ€miseen. RyhmĂ€improvisaation nĂ€kökulmassa korostuu opettajan rooli improvisaation ohjaajana. TĂ€mĂ€ tutkimus on kvalitatiivinen eli laadullinen kuvaileva tutkimus. Tutkimuksen metodologisena lĂ€htökohtana on fenomenografinen tutkimus, jonka aineisto on kerĂ€tty teemahaastattelulla. Haastateltavina oli neljĂ€ (N=4) musiikinopettajaa, jotka kĂ€yttĂ€vĂ€t työssÀÀn musiikillista improvisaatiota ryhmĂ€kontekstissa, kaksi heistĂ€ oli lukion ja kaksi vapaan sivistystyön musiikinopettajia. Haastateltavia yhdisti musiikillisen improvisaation kĂ€yttö musiikinopetuksen työvĂ€lineenĂ€. Aineistoa on kĂ€sitelty sisĂ€llön analyysin metodein. Tutkimuskysymysten kannalta olennaiset vastaukset pelkistettiin ja edelleen luokiteltiin alaluokiksi, ylĂ€luokiksi ja lopuksi yhdistĂ€viksi luokiksi. YhdistĂ€vĂ€t luokat syntyivĂ€t prosessin aikana tehtyjen analyysien perusteella ja tulkittiin tutkielman tuloksiksi. Tutkielman tuloksina selvisi, ettĂ€ osallistujien kĂ€sitysten mukaan musiikillinen improvisaatio toimintana voi olla konkreettista tekemistĂ€ tai työskentelyn tapa. Ajattelutapana improvisaatio on toimintaa ohjaava myös muualla arjessa. Musiikillisen improvisaation kannalta suotuisa oppimisympĂ€ristö on turvallinen, salliva ja hyvĂ€ksyvĂ€. Opettajan pedagoginen toiminta ja didaktinen osaaminen ohjaavat opettajaa musiikillisen improvisaation kannalta suotuisan oppimisympĂ€ristön rakentamisessa. Opettajan tasapuolisuus, positiivinen palaute ja työrauhasta huolehtiminen ovat olennaisia ilmapiirin sallivuuden ja turvallisuuden nĂ€kökulmasta. LisĂ€ksi selvisi, ettĂ€ oppimisympĂ€ristö rakentuu improvisoivan ryhmĂ€n jĂ€senten vĂ€lisen vuorovaikutuksen ja kommunikaation kautta. TĂ€llĂ€ tutkielmalla toimme nĂ€kyvĂ€ksi musiikillisessa ryhmĂ€improvisaatiossa kommunikaation ja positiivisen vuorovaikutuksen erityisen suuren merkityksen

    TiivistelmĂ€. Perinteisesti robotit ovat olleet tĂ€rkeĂ€ssĂ€ roolissa muun muassa tehdastyön tehokkuuden kasvattamisessa, mutta viime vuosina robotiikan huomio on keskittynyt yhĂ€ enemmĂ€n ihmistĂ€ muistuttaviin robotteihin eli humanoidirobotteihin, joita voitaisiin kĂ€yttÀÀ useissa tehtĂ€vissĂ€ ihmisten korvikkeena. Ihmisen muoto kuitenkin aiheuttaa useita haasteita robotin suunnittelussa, suurimpien joukossa luonnollisilta vaikuttavien liikkeiden toteuttaminen. InMoov-robotin pÀÀlle toteutettiin ohjausjĂ€rjestelmĂ€, joka parantaa aiemmin toteutettua kasvojen seuraamista yhdistĂ€mĂ€llĂ€ siihen pÀÀn liikkeet, ja lisÀÀ puhesynteesin perusteella liikkuvan leuan sekĂ€ erinĂ€isiĂ€ pÀÀn eleitĂ€ ihmisen ja robotin vuorovaikutuksen parantamiseksi. JĂ€rjestelmĂ€n suoriutumista testattiin kĂ€yttötarkoituksen mukaisessa ympĂ€ristössĂ€ kahdella erillisellĂ€ kerralla, joissa ilmi tulleita havaintoja kĂ€ytettiin jĂ€rjestelmĂ€n parantamiseen. Traditionally robots have had an important role in making factories more efficient, among other things, but in recent years the field of robotics has been more and more focused on robots that resemble humans, also known as humanoid robots, which could be used in many tasks as replacements for human workers. However, designing such robots poses many challenges, one of the greatest being the difficulty of creating human-like movements. In this work, a control system for the head of an InMoov robot, which improves on its previously implemented face tracking capabilities by adding head movement and implements speech synthesis-based jaw movements along with various head gestures in order to improve human-robot interaction with the robot, was implemented. The system was tested in its intended environment on two separate occasions in order to observe where the system could be improved further. The final system is able to turn the robot’s eyes and head towards detected faces in a human-like fashion and execute pre-programmed head gestures, in addition to which it has the capability to move its jaw according to speech synthesised in an external program.     Expression of Hyaluronan Synthases (HAS1–3) and Hyaluronidases (HYAL1–2) in Serous Ovarian Carcinomas: Inverse Correlation between HYAL1 and Hyaluronan Content

    <p>Abstract</p> <p>Background</p> <p>Hyaluronan, a tumor promoting extracellular matrix polysaccharide, is elevated in malignant epithelial ovarian tumors, and associates with an unfavorable prognosis. To explore possible contributors to the accumulation of hyaluronan, we examined the expression of hyaluronan synthases (<it>HAS1</it>, <it>HAS2 </it>and <it>HAS3</it>) and hyaluronidases (<it>HYAL1 </it>and <it>HYAL2</it>), correlated with hyaluronidase enzyme activity hyaluronan content and HAS1–3 immunoreactivity.</p> <p>Methods</p> <p>Normal ovaries (n = 5) and 34 serous epithelial ovarian tumors, divided into 4 groups: malignant grades 1+2 (n = 10); malignant grade 3 (n = 10); borderline (n = 4) and benign epithelial tumors (n = 10), were analyzed for mRNA by real-time RT-PCR and compared to hyaluronidase activity, hyaluronan staining, and HAS1–3 immunoreactivity in tissue sections of the same specimens.</p> <p>Results</p> <p>The levels of <it>HAS2 </it>and <it>HAS3 </it>mRNA (<it>HAS1 </it>was low or absent), were not consistently increased in the carcinomas, and were not significantly correlated with HAS protein or hyaluronan accumulation in individual samples. Instead, the median of <it>HYAL1 </it>mRNA level was 69% lower in grade 3 serous ovarian cancers compared to normal ovaries (P = 0.01). The expression of <it>HYAL1</it>, but not <it>HYAL2</it>, significantly correlated with the enzymatic activity of tissue hyaluronidases (r = 0.5; P = 0.006). An inverse correlation was noted between <it>HYAL1 </it>mRNA and the intensity of hyaluronan staining of the corresponding tissue sections (r = -0.4; P = 0.025).</p> <p>Conclusion</p> <p>The results indicate that in serous epithelial ovarian malignancies <it>HAS </it>expression is not consistently elevated but <it>HYAL1 </it>expression is significantly reduced and correlates with the accumulation of hyaluronan.     JNK1 controls adult hippocampal neurogenesis and imposes cell-autonomous control of anxiety behaviour from the neurogenic niche

    Promoting adult hippocampal neurogenesis is expected to induce neuroplastic changes that improve mood and alleviate anxiety. However, the underlying mechanisms remain largely unknown and the hypothesis itself is controversial. Here we show that mice lacking Jnk1, or c-Jun N-terminal kinase (JNK) inhibitor-treated mice, display increased neurogenesis in adult hippocampus characterized by enhanced cell proliferation and survival, and increased maturation in the ventral region. Correspondingly, anxiety behaviour is reduced in a battery of tests, except when neurogenesis is prevented by AraC treatment. Using engineered retroviruses, we show that exclusive inhibition of JNK in adult-born granule cells alleviates anxiety and reduces depressive-like behaviour. These data validate the neurogenesis hypothesis of anxiety. Moreover, they establish a causal role for JNK in the hippocampal neurogenic niche and anxiety behaviour, and advocate targeting of JNK as an avenue for novel therapies against affective disorders.Peer reviewe

    Socioeconomic position and intergenerational associations of ideal health behaviors

    Background: Promoting ideal cardiovascular health behaviors is an objective of the American Heart Association 2020 goals. We hypothesized that ideal health behaviors of parents are associated with health behaviors of their adult offspring, and that higher socioeconomic position in either generation enhances intergenerational associations of ideal health behaviors.Design: Prospective cohort study.Methods: We included 1856 Young Finns Study participants who had repeated measurements of socioeconomic position (education, income, occupation), smoking status, body mass index, physical activity and diet from 2001, 2007 and 2011, and data on parental socioeconomic position and health behaviors from 1980. We calculated the total number of ideal behaviors in both generations using American Heart Association definitions. Intergenerational associations were examined using ordinal and linear multilevel regression with random intercepts, in which each participant contributed one, two or three measurements of adult health behaviors (2001, 2007, 2011). All analyses were adjusted for offspring sex, birth year, age, parental education and single parenthood.Results: Overall, parental ideal health behaviors were associated with ideal behaviors among offspring (odds ratio (OR) 1.28, 95% confidence interval 1.17, 1.39). Furthermore, ORs for these intergenerational associations were greater among offspring whose parents or who themselves had higher educational attainment (OR 1.56 for high vs. OR 1.19 for low parental education; P = 0.01 for interaction, OR 1.32 for high vs. OR 1.04 for low offspring education; P = 0.02 for interaction). Similar trends were seen with parental income and offspring occupation. Results from linear regression analyses were similar.Conclusions: These prospective data suggest higher socioeconomic position in parents or in their adult offspring strengthens the intergenerational continuum of ideal cardiovascular health behaviors.</p

    Non-telecentric two-photon microscopy for 3D random access mesoscale imaging

    Diffraction-limited two-photon microscopy permits minimally invasive optical monitoring of neuronal activity. However, most conventional two-photon microscopes impose significant constraints on the size of the imaging field-of-view and the specific shape of the effective excitation volume, thus limiting the scope of biological questions that can be addressed and the information obtainable. Here, employing a non-telecentric optical design, we present a low-cost, easily implemented and flexible solution to address these limitations, offering a several-fold expanded three-dimensional field of view. Moreover, rapid laser-focus control via an electrically tunable lens allows near-simultaneous imaging of remote regions separated in three dimensions and permits the bending of imaging planes to follow natural curvatures in biological structures. Crucially, our core design is readily implemented (and reversed) within a matter of hours, making it highly suitable as a base platform for further development. We demonstrate the application of our system for imaging neuronal activity in a variety of examples in zebrafish, mice and fruit flies

    Enterprise Education Competitions: A Theoretically Flawed Intervention?

    The demand for including enterprise in the education system, at all levels and for all pupils is now a global phenomenon. Within this context, the use of competitions and competitive learning activities is presented as a popular and effective vehicle for learning. The purpose of this chapter is to illustrate how a realist method of enquiry – which utilises theory as the unit of analysis – can shed new light on the assumed and unintended outcomes of enterprise education competitions. The case developed here is that there are inherent flaws in assuming that competitions will ‘work’ in the ways set out in policy and guidance. Some of the most prevalent stated outcomes – that competitions will motivate and reward young people, that they will enable the development of entrepreneurial skills, and that learners will be inspired by their peers – are challenged by theory from psychology and education. The issue at stake is that the expansion of enterprise education policy into primary and secondary education increases the likelihood that more learners will be sheep dipped in competitions, and competitive activities, without a clear recognition of the potential unintended effects. In this chapter, we employ a realist-informed approach to critically evaluate the theoretical basis that underpins the use of competitions and competitive learning activities in school-based enterprise education. We believe that our findings and subsequent recommendations will provide those who promote and practice the use of competitions with a richer, more sophisticated picture of the potential flaws within such activities.Peer reviewedFinal Published versio
