    An Approach to Relate Viewpoints and Modeling Languages

    The architectural design of distributed enterprise applications from the viewpoints of different stakeholders has been proposed for some time, for example, as part of RM-ODP and IEEE 1471, and seems now-a-days to gain acceptance in practice. However, much work remains to be done on the relationships between different viewpoints. Failing to relate viewpoints may lead to a collection of viewpoint models that is inconsistent, and may therefore lead to an incorrect implementation. This paper defines an approach that helps designers to relate different viewpoints to each other. Thereby, it helps to enforce the consistency of the overall design. The results of this paper are expected to be particularly interesting for Model Driven Architecture (MDA) projects, since the proposed models can be used for the explicit definition of the models and relationships between models in an MDA trajectory

    Carbon and nitrogen cycling in the Scheldt estuary: the major players, long-term changes and an integrated view

    The Scheldt estuary is a highly heterotrophic, nutrient-rich, turbid, tidal estuary in a densely populated area (Belgium/The Netherlands). Here we present the results (1) on the long-term changes in nutrient loadings and transformations within the estuary and (2) on nitrogen cycling rate measurements obtained with isotopic tracers. Moreover, we have developed and applied novel techniques that allow direct linking of process rates to the identity and biomass of the organisms involved. Monitoring data and process studies have been used in numerical models to integrate the various biogeochemical cycles and to advance our understanding of the evolving estuarine filter function of the Scheldt estuary

    De SumWing is een innovatief vistuig dat ontwikkeld wordt om brandstof te besparen in de boomkorvisserij op platvis en bij te dragen aan het verminderen van ongewenste ecosysteem effecten om zodoende een toekomstperspectief voor deze tak van visserij te bewerkstelligen. De ontwikkeling startte in 2006 in het kader van de Taskforce Duurzame Visserijontwikkeling en wordt momenteel als project onder begeleiding van het Visserij Innovatie Platvorm (VIP) voortgezet. Een indicatieve berekening van de penetratiediepte van een SumWing-tuig geeft een vermindering van ca. 10% t.o.v. een boomkortuig met sloffen onder de aanname dat de wekkers gelijk zijn. Combinatie van de SumWing en de pulskor zou dit percentage kunnen verhogen tot 50% en is hierdoor een interessante optie om bodemimpact verder te verminderen

    G protein-coupled receptors (GPCRs) are the largest class of membrane receptors, playing a key role in the regulation of processes as varied as neurotransmission and immune response. Evidence for GPCR oligomerisation has been accumulating that challenges the idea that GPCRs function solely as monomeric receptors; however, GPCR oligomerisation remains controversial primarily due to the difficulties in comparing evidence from very different types of structural and dynamic data. Using a combination of single-molecule and ensemble FRET, double electron–electron resonance spectroscopy, and simulations, we show that dimerisation of the GPCR neurotensin receptor 1 is regulated by receptor density and is dynamically tuneable over the physiological range. We propose a “rolling dimer” interface model in which multiple dimer conformations co-exist and interconvert. These findings unite previous seemingly conflicting observations, provide a compelling mechanism for regulating receptor signalling, and act as a guide for future physiological studies

    The additional value of first pass myocardial perfusion imaging during peak dose of dobutamine stress cardiac MRI for the detection of myocardial ischemia

    Purpose of this study was to assess the additional value of first pass myocardial perfusion imaging during peak dose of dobutamine stress Cardiac-MR (CMR). Dobutamine Stress CMR was performed in 115 patients with an inconclusive diagnosis of myocardial ischemia on a 1.5 T system (Magnetom Avanto, Siemens Medical Systems). Three short-axis cine and grid series were acquired during rest and at increasing doses of dobutamine (maximum 40 μg/kg/min). On peak dose dobutamine followed immediately by a first pass myocardial perfusion imaging sequence. Images were graded according to the sixteen-segment model, on a four point scale. Ninety-seven patients showed no New (Induced) Wall Motion Abnormalities (NWMA). Perfusion imaging showed absence of perfusion deficits in 67 of these patients (69%). Perfusion deficits attributable to known previous myocardial infarction were found in 30 patients (31%). Eighteen patients had NWMA, indicative for myocardial ischemia, of which 14 (78%) could be confirmed by a corresponding perfusion deficit. Four patients (22%) with NWMA did not have perfusion deficits. In these four patients NWMA were caused by a Left Bundle Branch Block (LBBB). They were free from cardiac events during the follow-up period (median 13.5 months; range 6–20). Addition of first-pass myocardial perfusion imaging during peak-dose dobutamine stress CMR can help to decide whether a NWMA is caused by myocardial ischemia or is due to an (inducible) LBBB, hereby preventing a false positive wall motion interpretation

    Pulmonary MTBVAC vaccination induces immune signatures previously correlated with prevention of tuberculosis infection

    To fight tuberculosis, better vaccination strategies are needed. Live attenuated Mycobacterium tuberculosis-derived vaccine, MTBVAC, is a promising candidate in the pipeline, proven to be safe and immunogenic in humans so far. Independent studies have shown that pulmonary mucosal delivery of Bacillus Calmette-Guérin (BCG), the only tuberculosis (TB) vaccine available today, confers superior protection over standard intradermal immunization. Here we demonstrate that mucosal MTBVAC is well tolerated, eliciting polyfunctional T helper type 17 cells, interleukin-10, and immunoglobulins in the airway and yielding a broader antigenic profile than BCG in rhesus macaques. Beyond our previous work, we show that local immunoglobulins, induced by MTBVAC and BCG, bind to M. tuberculosis and enhance pathogen uptake. Furthermore, after pulmonary vaccination, but not M. tuberculosis infection, local T cells expressed high levels of mucosal homing and tissue residency markers. Our data show that pulmonary MTBVAC administration has the potential to enhance its efficacy and justifies further exploration of mucosal vaccination strategies in preclinical efficacy studies
