    Development and application of a self-referencing glucose microsensor for the measurement of glucose consumption by pancreatic ?-cells

    Glucose gradients generated by an artificial source and ?-cells were measured using an enzyme-based glucose microsensor, 8-?m tip diameter, as a self-referencing electrode. The technique is based on a difference measurement between two locations in a gradient and thus allows us to obtain real-time flux values with minimal impact of sensor drift or noise. Flux values were derived by incorporation of the measured differential current into Fick's first equation. In an artificial glucose gradient, a flux detection limit of 8.2 ± 0.4 pmol·cm-2·s-1 (mean ± SEM, n = 7) with a sensor sensitivity of 7.0 ± 0.4 pA/mM (mean ± SEM, n = 16) was demonstrated. Under biological conditions, the glucose sensor showed no oxygen dependence with 5 mM glucose in the bulk medium. The addition of catalase to the bulk medium was shown to ameliorate surface-dependent flux distortion close to specimens, suggesting an underlying local accumulation of hydrogen peroxide. Glucose flux from ?-cell clusters, measured in the presence of 5 mM glucose, was 61.7 ± 9.5 fmol·nL-1·s-1 (mean ± SEM, n = 9) and could be pharmacologically modulated. Glucose consumption in response to FCCP (1 ?M) transiently increased, subsequently decreasing to below basal by 93 ± 16 and 56 ± 6%, respectively (mean ± SEM, n = 5). Consumption was decreased after the application of 10 ?M rotenone by 74 ± 5% (mean ± SEM, n = 4). These results demonstrate that an enzyme-based amperometric microsensor can be applied in the self-referencing mode. Further, in obtaining glucose flux measurements from small clusters of cells, these are the first recordings of the real-time dynamic of glucose movements in a biological microenvironment. <br/

    Diversity-induced resonance

    We present conclusive evidence showing that different sources of diversity, such as those represented by quenched disorder or noise, can induce a resonant collective behavior in an ensemble of coupled bistable or excitable systems. Our analytical and numerical results show that when such systems are subjected to an external subthreshold signal, their response is optimized for an intermediate value of the diversity. These findings show that intrinsic diversity might have a constructive role and suggest that natural systems might profit from their diversity in order to optimize the response to an external stimulus.Comment: 4 pages, 3 figure

    Joint-specific changes in locomotor complexity in the absence of muscle atrophy following incomplete spinal cord injury

    ackground Following incomplete spinal cord injury (iSCI), descending drive is impaired, possibly leading to a decrease in the complexity of gait. To test the hypothesis that iSCI impairs gait coordination and decreases locomotor complexity, we collected 3D joint angle kinematics and muscle parameters of rats with a sham or an incomplete spinal cord injury. Methods 12 adult, female, Long-Evans rats, 6 sham and 6 mild-moderate T8 iSCI, were tested 4 weeks following injury. The Basso Beattie Bresnahan locomotor score was used to verify injury severity. Animals had reflective markers placed on the bony prominences of their limb joints and were filmed in 3D while walking on a treadmill. Joint angles and segment motion were analyzed quantitatively, and complexity of joint angle trajectory and overall gait were calculated using permutation entropy and principal component analysis, respectively. Following treadmill testing, the animals were euthanized and hindlimb muscles removed. Excised muscles were tested for mass, density, fiber length, pennation angle, and relaxed sarcomere length. Results Muscle parameters were similar between groups with no evidence of muscle atrophy. The animals showed overextension of the ankle, which was compensated for by a decreased range of motion at the knee. Left-right coordination was altered, leading to left and right knee movements that are entirely out of phase, with one joint moving while the other is stationary. Movement patterns remained symmetric. Permutation entropy measures indicated changes in complexity on a joint specific basis, with the largest changes at the ankle. No significant difference was seen using principal component analysis. Rats were able to achieve stable weight bearing locomotion at reasonable speeds on the treadmill despite these deficiencies. Conclusions Decrease in supraspinal control following iSCI causes a loss of complexity of ankle kinematics. This loss can be entirely due to loss of supraspinal control in the absence of muscle atrophy and may be quantified using permutation entropy. Joint-specific differences in kinematic complexity may be attributed to different sources of motor control. This work indicates the importance of the ankle for rehabilitation interventions following spinal cord injury

    The Effects of Pet Ownership on Anxiety and Depression Among Trauma-Exposed College Students

    Rates of anxiety and depression are prevalent in college students and can be attributed in part to stress and trauma-related events. However, studies suggest that pet ownership has the possibility of alleviating symptoms of anxiety, depression, negative emotions, and suicide. The purpose of this study was to determine the relationship between pet ownership and levels of anxiety and depression among those who have experienced a traumatic event. The sample was comprised of five hundred and forty-seven VCU students who completed an online survey from Spit for Science during their junior year. Linear regressions were performed to determine the nature and strength of the relationship between our two variables. After controlling for gender, race/ethnicity, personality, social support, and resiliency, we found statistically significant lower levels of anxiety and depression among pet owners compared to non-pet owners (p=0.004). This study reinforces how pets can impact our mental health, and lends further research to support programs such as VCU’s Center for Human-Animal Interaction (CHAI) and their Dogs on Call program.https://scholarscompass.vcu.edu/uresposters/1238/thumbnail.jp

    A 100-element HBT grid amplifier

    A 100-element 10-GHz grid amplifier has been developed. The active devices in the grid are chips with heterojunction-bipolar-transistor (HBT) differential pairs. The metal grid pattern was empirically designed to provide effective coupling between the HBTs and free space. Two independent measurements, one with focusing lenses and the other without, were used to characterize the grid. In each case, the peak gain was 10 dB at 10 GHz with a 3-dB bandwidth of 1 GHz. The input and output return losses were better than 15 dB at 10 GHz. The maximum output power was 450 mW, and the minimum noise figure was 7 dB. By varying the bias, a signal could be amplitude modulated with a modulation index as large as 0.65. Tests show that the grid was quite tolerant of failures-the output power dropped by only 1 dB when 10% of the inputs were detuned. The grid amplifier is a multimode device that amplifies beams of different shapes and angles. Beams with incidence angles up to 30° were amplified with less than a 3-dB drop in gain

    Workstation environment for wastewater treatment design using AI and mathematical models

    This research explores the use of computer-based environments to facilitate environmental engineering decision making. A prototype system is developed for wastewater treatment plant design as an exploration tool to demonstrate the techniques and principles proposed. Several mathematical techniques, interactive graphic displays, and friendly user interfaces are used. The mathematical techniques are: (1) mass and water balances for an analysis program for wastewater treatment plant design, (2) a rule-based system for sludge bulking judgment, and (3) a standard processor for checking a design against existing design standards.
