    Atividade antibiofilme de substâncias de Croton urucurana em Staphylococcus aureus isolado de mastite bovina

    RESUMO: A mastite bovina é a enfermidade que causa maior impacto na produção leiteira, sendo o microrganismo Staphylococcus aureus o mais prevalente. Este gênero possui a capacidade de produzir biofilmes que é um importante mecanismo de resistência aos antibióticos. Considerando a capacidade terapêutica das plantas, a espécie Croton urucurana, nativa do Cerrado, foi alvo do presente estudo, que teve como objetivo avaliar a atividade antibiofilme in vitro do extrato vegetal e substâncias isoladas desta espécie, frente Staphylococcus aureus, isolados de leite de vacas com mastite, bem como dos antibióticos gentamicina e vancomicina. A atividade antibiofilme foi avaliada por meio do cristal violeta e da contagem de unidades formadoras de colônia. As imagens foram obtidas por microscopia eletrônica de varredura. O extrato bruto e frações de C. urucurana apresentaram atividade antibiofilme superior à gentamicina e semelhante à vancomicina, enquanto a substância isolada α-Costol foi significativamente mais ativa quando comparada aos demais tratamentos avaliados, reduzindo cerca de 6 ciclos logarítmicos da população bacteriana em biofilme. Conclui-se que os fitocomplexos e a substância α-Costol isolados de Croton urucurana são promissores no combate a um dos principais agentes etiológicos da mastite bovina

    The objective of this study is to evaluate the potencial microbial activity in-vitro from the extract of some endemic plants from Cerrado such as Baccharis dracunculifolia, Cochlospermum regium, Croton antisyphiliticus, Eugenia dysenterica and Lippia sidoides, against the agent Staphylococcus aureus isolated from bovine mastitic milk, osteo from cow’s teat, milker equipament, nasal cavitites and milker’s gullet. The extracts were prepared from aerial parts as well as the reticular systems of plants using the solvents methanol, hexane and chloroform at a concentration of 10%. To evaluate the antimicrobial activity, the technique of microdilution in broth was used for determining the Minimal Inibitory Concentration (MIC) followed by the determination of Minimal Bactericidal Concentration (MBC). The extracts from Baccharis dracunculifolia and Croton antisyphiliticus, followed by extracts from Lippia sidoides, reported respectively, presented better inhibitory activity against the multiplication of the bacteria Staphylococcus aureus. Furthermore, the results demonstrate that the isolated strains from the milk and nasal cavities of the milker showed strong resistance against gentamicin, active agent commonly applied to combat mastitis bovine. However, there was sensitivity against extracts from the reported plants, reinforcing the importance of the medicinal plants as a therapeutic resource and its aplicability.O presente estudo teve como objetivo avaliar o potencial de atividade antimicrobiana in vitro dos extratos de algumas plantas endêmicas do Cerrado tais como Baccharis dracunculifolia, Cochlospermum regium, Croton antisyphiliticus, Eugenia dysenterica e Lippia sidoides, frente ao agente Staphylococcus aureus isolado de leite mastítico, ósteo do teto da vaca, equipamento de ordenhadeira, fossas nasais e garganta do ordenhador. Os extratos foram preparados a partir das partes aéreas e sistema radicular das plantas utilizando os solventes metanol, hexano e clorofórmio na concentração de 10%. Para avaliação da atividade antimicrobiana foi aplicada a técnica de microdiluição em caldo para determinação da Concentração Inibitória Mínima (CIM) seguida da determinação da Concentração Bactericida Mínima (MBC). Os extratos de Baccharis dracunculifolia, Croton antisyphiliticus, seguido do extrato de Lippia sidoides, apresentaram, respectivamente, melhor atividade inibitória sobre a multiplicação da bactéria Staphylococcus aureus. Os resultados obtidos demonstraram, ainda, que as estirpes isoladas do leite e das fossas nasais do ordenhador foram resistentes a gentamicina, princípio ativo muito utilizado no combate à mastite bovina, porém sensíveis aos extratos das plantas referidas, reforçando a importância das plantas medicinais como recurso terapêutico e sua aplicabilidade

    Ethanol extract and fractions from aerial parts of Eclipta alba (L.) Hassk (Asteraceae) were screened for the antibacterial and antifungal activities against different species of human pathogenic bacterial ATCC, antibiotic-resistant clinical isolates and strains of the dermatophyte Trichophyton rubrum (wild and mutant for TruMDR2 gene) using a microdilution method. Demethylwedelolactone/wedelolactone (DWL/WL) and only wedelolactone (WL), both in a high homogeneity degree, were efficient to inhibit the ATCC strains of Staphylococus aureus (Minimal Inhibitory Concentration MIC = 75 μg/mL), Staphylococcus epidemidis (MIC = 125 μg/mL) and Escherichia coli (MIC = 125 μg/mL) as well as antibiotic-resistant clinical isolates of Enterococcus spp (MIC = 250 μg/mL) and S. aureus (MIC = 125 μg/mL). Ethanol extract was more effective than the purified fractions against Trichophyton rubrum strains (MIC = 125 μg/mL), suggesting that anti-fungal activity is not only related to demethylwedelolactone and wedelolactone, but also to a synergistic action between these coumestans and other compounds found in that extract. Thus, this work suggests that E. alba possesses a significant antimicrobial activity, including that against multi-drug resistant microorganisms, which could be of relevance for the treatment of infectious diseases.Colegio de Farmacéuticos de la Provincia de Buenos Aire

    Genetic structure and chemical diversity in natural populations of Uncaria guianensis (Aubl.) J.F.Gmel. (Rubiaceae).

    Uncaria guianensis is native to the Amazon and is used traditionally as an anti-inflammatory. Natural populations of the species have declined markedly in recent times because of strong anthropic pressure brought about by deforestation and indiscriminate collection. The aim of the present study was to assess the genetic and chemical diversity among eight natural populations of U. guianensis located in the Brazilian states of Acre, Amapá and Amazonas. A set of four primer combinations was employed in sequence-related amplified polymorphism (SRAP) amplifications of leaf DNA, and the fragments were analyzed in an LI-COR model 4300 DNA Analyzer. Genetic variability within the populations (81%) was substantially greater than that detected between them (19%). The highest percentage of polymorphic loci (90.21%) and the largest genetic variability were observed in the population located in Mazagão, Amapá. Genetic differentiation between populations was high (Fst = 0.188) and the studied populations formed three distinct genetic groups (K = 3). The population located in Assis Brasil, Acre, presented the highest average content of the mitraphylline (0.60 mg/g dry weight,). However, mitraphylline and isomitraphylline not detected in most individuals in the studied populations, and it is questionable whether they should be considered as chemical markers of the species. The genetic data confirm the urgent need for conservation programs for U. guianensis, and for further studies aimed at ascertaining the genetic basis and heritability of alkaloid accumulation
