    Quantifying Ca 2+ Current and Permeability in ATP-gated P2X7 Receptors

    International audienceATP-gated P2X7 receptors are prominently expressed in inflammatory cells and play a key role in the immune response. A major consequence of receptor activation is the regulated influx of Ca(2+) through the self-contained cation non-selective channel. Although the physiological importance of the resulting rise in intracellular Ca(2+) is universally acknowledged, the biophysics of the Ca(2+) flux responsible for the effects are poorly understood, largely because traditional methods of measuring Ca(2+) permeability are difficult to apply to P2X7 receptors. Here we use an alternative approach, called dye-overload patch-clamp photometry, to quantify the agonist-gated Ca(2+) flux of recombinant P2X7 receptors of dog, guinea pig, human, monkey, mouse, rat, and zebrafish. We find that the magnitude of the Ca(2+) component of the ATP-gated current depends on the species of origin, the splice variant, and the concentration of the purinergic agonist. We also measured a significant contribution of Ca(2+) to the agonist-gated current of the native P2X7Rs of mouse and human immune cells. Our results provide cross-species quantitative measures of the Ca(2+) current of the P2X7 receptor for the first time, and suggest that the cytoplasmic N terminus plays a meaningful role in regulating the flow of Ca(2+) through the channel

    Walking behaviours from the 1965–2003 American Heritage Time Use Study (AHTUS)

    <p>Abstract</p> <p>Background</p> <p>The American Heritage Time Use Study (AHTUS) represents a harmonised historical data file of time use by adults, amalgamating surveys collected in 1965–66, 1975–76, 1985, 1992–94, and 2003. The objectives of time-use studies have ranged from evaluating household and other unpaid production of goods and services, to monitoring of media use, to comparing lifestyles of more and less privileged social groups, or to tracking broad shifts in social behaviour. The purpose of this paper is to describe the process and utility of identifying and compiling data from the AHTUS to describe a range of walking behaviours collected using time-use survey methods over almost 40 years in the USA.</p> <p>Methods</p> <p>This is a secondary data analysis of an existing amalgamated data set. Noting source survey-specific limitations in comparability of design, we determined age-standardized participation (and associated durations) in any walking, walking for exercise, walking for transport, walking the dog, sports/exercise (excluding walking), and all physical activity for those survey years for which sufficient relevant data details were available.</p> <p>Results</p> <p>Data processing revealed inconsistencies in instrument administration, coding various types of walking and in prompting other sport/exercise across surveys. Thus for the entire period, application of inferential statistics to determine trend for a range of walking behaviours could not be done with confidence. Focusing on the two most comparable survey years, 1985 and 2003, it appears that walking for exercise in America has increased in popularity on any given day (from 2.9 to 5.4% of adults) and accumulated duration amongst those who walk for exercise (from 30 to 45 mins/day). Dog walking has decreased in popularity over the same time period (from 9.4 to 2.6%). Associated duration amongst dog walkers was stable at 30 mins/day.</p> <p>Conclusion</p> <p>The noted and sometimes substantial differences in methods between the various survey administrations preclude stringent interpretation of these trends in walking behaviours and the use of conventional application of inferential statistics to evaluate significance of time trends. Although the AHTUS offers the most comprehensive attempt at harmonization yet undertaken with these individual time-use surveys, we found that any noted cross-time changes in walking and physical activity behaviour are not easily interpreted in terms of conventional epidemiological approaches and could be true changes, artefact related to instrument and method changes, or both. Public health utilization of the AHTUS, could be enhanced with greater attention to methodological issues known to influence estimation of physical activity behaviour in population. This could be achieved with cross-disciplinary collaboration between groups of experts in the various stages of these surveys.</p

    What factors influence the rediscovery of lost tetrapod species?

    We created a database of lost and rediscovered tetrapod species, identified patterns in their distribution and factors influencing rediscovery. Tetrapod species are being lost at a faster rate than they are being rediscovered, due to slowing rates of rediscovery for amphibians, birds and mammals, and rapid rates of loss for reptiles. Finding lost species and preventing future losses should therefore be a conservation priority. By comparing the taxonomic and spatial distribution of lost and rediscovered tetrapod species, we have identified regions and taxa with many lost species in comparison to those that have been rediscovered—our results may help to prioritise search effort to find them. By identifying factors that influence rediscovery, we have improved our ability to broadly distinguish the types of species that are likely to be found from those that are not (because they are likely to be extinct). Some lost species, particularly those that are small and perceived to be uncharismatic, may have been neglected in terms of conservation effort, and other lost species may be hard to find due to their intrinsic characteristics and the characteristics of the environments they occupy (e.g. nocturnal species, fossorial species and species occupying habitats that are more difficult to survey such as wetlands). These lost species may genuinely await rediscovery. However, other lost species that possess characteristics associated with rediscovery (e.g. large species) and that are also associated with factors that negatively influence rediscovery (e.g. those occupying small islands) are more likely to be extinct. Our results may foster pragmatic search protocols that prioritise lost species likely to still exist

    Genome-wide copy number alterations in subtypes of invasive breast cancers in young white and African American women.

    Genomic copy number alterations (CNA) are common in breast cancer. Identifying characteristic CNAs associated with specific breast cancer subtypes is a critical step in defining potential mechanisms of disease initiation and progression. We used genome-wide array comparative genomic hybridization to identify distinctive CNAs in breast cancer subtypes from 259 young (diagnosed with breast cancer at 40%) for TN breast tumors at 10q, 11p, 11q, 16q, 20p, and 20q. In addition, we report CNAs that differ in frequency between TN breast tumors of AA and CA women. This is of particular relevance because TN breast cancer is associated with higher mortality and young AA women have higher rates of TN breast tumors compared to CA women. These data support the possibility that higher overall frequency of genomic alteration events as well as specific focal CNAs in TN breast tumors might contribute in part to the poor breast cancer prognosis for young AA women

    SLACC: Simion-based Language Agnostic Code Clones

    Successful cross-language clone detection could enable researchers and developers to create robust language migration tools, facilitate learning additional programming languages once one is mastered, and promote reuse of code snippets over a broader codebase. However, identifying cross-language clones presents special challenges to the clone detection problem. A lack of common underlying representation between arbitrary languages means detecting clones requires one of the following solutions: 1) a static analysis framework replicated across each targeted language with annotations matching language features across all languages, or 2) a dynamic analysis framework that detects clones based on runtime behavior. In this work, we demonstrate the feasibility of the latter solution, a dynamic analysis approach called SLACC for cross-language clone detection. Like prior clone detection techniques, we use input/output behavior to match clones, though we overcome limitations of prior work by amplifying the number of inputs and covering more data types; and as a result, achieve better clusters than prior attempts. Since clusters are generated based on input/output behavior, SLACC supports cross-language clone detection. As an added challenge, we target a static typed language, Java, and a dynamic typed language, Python. Compared to HitoshiIO, a recent clone detection tool for Java, SLACC retrieves 6 times as many clusters and has higher precision (86.7% vs. 30.7%). This is the first work to perform clone detection for dynamic typed languages (precision = 87.3%) and the first to perform clone detection across languages that lack a common underlying representation (precision = 94.1%).     First report of generalized face processing difficulties in möbius sequence.

    Reverse simulation models of facial expression recognition suggest that we recognize the emotions of others by running implicit motor programmes responsible for the production of that expression. Previous work has tested this theory by examining facial expression recognition in participants with Möbius sequence, a condition characterized by congenital bilateral facial paralysis. However, a mixed pattern of findings has emerged, and it has not yet been tested whether these individuals can imagine facial expressions, a process also hypothesized to be underpinned by proprioceptive feedback from the face. We investigated this issue by examining expression recognition and imagery in six participants with Möbius sequence, and also carried out tests assessing facial identity and object recognition, as well as basic visual processing. While five of the six participants presented with expression recognition impairments, only one was impaired at the imagery of facial expressions. Further, five participants presented with other difficulties in the recognition of facial identity or objects, or in lower-level visual processing. We discuss the implications of our findings for the reverse simulation model, and suggest that facial identity recognition impairments may be more severe in the condition than has previously been noted
