    Preparation of a Nanoemulsion with Carapa guianensis

    Andiroba (Carapa guianensis) seeds are the source of an oil with a wide range of biological activities and ethnopharmacological uses. However, few studies have devoted attention to innovative formulations, including nanoemulsions. The present study aimed to obtain a colloidal system with the andiroba oil using a low-energy and organic-solvent-free method. Moreover, the preliminary residual larvicidal activity of the nanoemulsion against Aedes aegypti was evaluated. Oleic and palmitic acids were the major fatty acids, in addition to the phytosterol β-sitosterol and limonoids (tetranortriterpenoids). The required hydrophile-lipophile was around 11.0 and the optimal nanoemulsion was obtained using polysorbate 85. The particle size distribution suggested the presence of small droplets (mean diameter around 150 nm) and low polydispersity index (around 0.150). The effect of temperature on particle size distribution revealed that no major droplet size increase occurred. The preliminary residual larvicidal assay suggested that the mortality increased as a function of time. The present study allowed achievement of a potential bioactive oil in water nanoemulsion that may be a promising controlled release system. Moreover, the ecofriendly approach involved in the preparation associated with the great bioactive potential of C. guianensis makes this nanoemulsion very promising for valorization of this Amazon raw material

    Update on the approach to smoking in patients with respiratory diseases.

    Smoking is the leading cause of respiratory disease (RD). The harmful effects of smoking on the respiratory system begin in utero and influence immune responses throughout childhood and adult life. In comparison with ?healthy? smokers, smokers with RD have peculiarities that can impede smoking cessation, such as a higher level of nicotine dependence; nicotine withdrawal; higher levels of exhaled carbon monoxide; low motivation and low self-efficacy; greater concern about weight gain; and a high prevalence of anxiety and depression. In addition, they require more intensive, prolonged treatment. It is always necessary to educate such individuals about the fact that quitting smoking is the only measure that will reduce the progression of RD and improve their quality of life, regardless of the duration and severity of the disease. Physicians should always offer smoking cessation treatment. Outpatient or inpatient smoking cessation treatment should be multidisciplinary, based on behavioral interventions and pharmacotherapy. It will thus be more effective and cost-effective, doubling the chances of success

    Mimiviruses and the Human Interferon System: Viral Evasion of Classical Antiviral Activities, But Inhibition By a Novel Interferon-beta Regulated Immunomodulatory Pathway

    International audienceIn this review we discuss the role of mimiviruses as potential human pathogens focusing on clinical and evolutionary evidence. We also propose a novel antiviral immunomodulatory pathway controlled by interferon-beta (IFN-beta) and mediated by immune-responsive gene 1 (IRG1) and itaconic acid, its product. Acanthamoeba polyphaga Mimivirus (APMV) was isolated from amoebae in a hospital while investigating a pneumonia outbreak. Mimivirus ubiquity and role as protist pathogens are well understood, and its putative status as a human pathogen has been gaining strength as more evidence is being found. The study of APMV and human cells interaction revealed that the virus is able to evade the IFN system by inhibiting the regulation of interferon-stimulated genes, suggesting that the virus and humans have had host-pathogen interactions. It also has shown that the virus is capable of growing on IFN-alpha 2, but not on IFN-beta-treated cells, hinting at an exclusive IFN-beta antiviral pathway. Our hypothesis based on preliminary data and published articles is that IFN-beta preferentially upregulates IRG1 in human macrophagic cells, which in turn produces itaconic acid. This metabolite links metabolism to antiviral activity by inactivating the virus, in a novel immunomodulatory pathway relevant for APMV infections and probably to other infectious diseases as well

    Soroprevalência de Toxoplasma gondii em equídeos do Nordeste do Brasil

    RESUMO: Este estudo teve como objetivo determinar a soroprevalência da toxoplasmose em equídeos mantidos em diferentes formas de manejo no estado de Pernambuco. Para tanto, um total de 400 amostras de soro sanguíneo de equídeos clinicamente saudáveis foram analisados através do teste de aglutinação modificado (MAT) considerando-se cut-off de 1:25. Dados referentes às características dos animais e dos rebanhos, sistema de criação, presença de outros animais, idade, sexo, raça, aptidão, condição física foram coletados por meio de questionários investigativos. Anticorpos IgG anti-Toxoplasma gondii foram detectados em 12,5% (50/400) dos animais analisados. Dos 12 municípios estudados, houve positividade em 91,67% (11/12) com variação entre 4,4% e 33,3%. Quando avaliados os fatores de risco, apenas o fator mesorregião (p=0,029) apresentou associação com a infecção, particularmente Zona da Mata (OR=3), seguida de Região Metropolitana do Recife (OR=2,2), Agreste (OR=1,7) e Sertão (OR=1). Os resultados revelam a presença do parasito na área estudada, o que pode representar um elo na cadeia de transmissão da toxoplasmose a qual tem repercussão em saúde pública tendo em vista que o Brasil é o oitavo maior exportador de carne equina do mundo

    Ammonia emissions from dairy cows grazing an annual temperate pasture with or without energetic supplementation

    Ammonia (NH3) emissions from animal excretions has been a concern in grazing dairy systems, and is dependent of the proportion of total urinary and fecal nitrogen (N) lost as NH3. The aim of this study was to assess NH3 emissions from excreta of dairy cows grazing an annual temperate pasture (A. strigosa Schum + L. multiflorum Lam.) exclusively or receiving energetic supplementation. Treatments were grazing without supplementation, grazing + 4.2 kg DM corn silage and grazing + 3.2 kg DM ground corn. Supplements were calculated in order to provide the same amount of metabolisable energy. Feces and urine were sampled from four cows of each treatment during one day, at 7:30 and 16:30 h. One hundred and thirty g of feces and 80 mL of urine from each treatment plus a control without excreta were assigned into the pasture area, considering a randomized complete block design with three replications. The NH3 was captured by a semi-open chamber (10 cm diameter) containing a foam strip soaked with a solution of H2SO4 1M + 2% glycerin. The foam strips were replaced 2, 4, 7, 10, 13, 17, 21 and 25 days after distribution of excreta into the pasture, and ammonium concentration was performed by steam distillation. The amount of N recovered was multiplied by a factor of 1.74 calculated from the N-NH3 recovery (1/0.63) of this technic. The fraction of excreta N volatilized as NH3 was calculated by the ratio between the N volatilized from excreta corrected to the volatilization from the control. The fraction of urinary N lost as NH3 was similar between treatments, averaging 19.2%, but the fraction of fecal N lost as NH3 decreased from 4.6% in no supplemented cows to 2.4 and 0.6% in animals supplemented with corn silage and corn ground, respectively. Total excreta (urine + feces) NH3-N emissions factors were similar between treatments, averaging 22%. Supplementing dairy cows grazing an annual temperate pasture with corn silage or corn ground may mitigate NH3 emissions factors from feces but not from urine or total excreta. The amount of N excreted by these animals will be considered to obtain the real amounts of NH3-N emissions in each treatment

    Padronização de multiplex PCR para detecção de dermatófitos em pelos e crostas de cães e gatos

    RESUMO: Objetivou-se padronizar uma reação do tipo multiplex PCR (mPCR) para detectar Microsporum canis, Microsporum gypseum e o complexo Trichophyton mentagrophytes em amostras de pelos e/ou crostas de cães e gatos. 250 amostras de pelos e/ou crostas de cães e gatos foram analisadas por meio de exame direto e cultura, o DNA das mesmas foi extraído para mPCR. Primers foram desenhados e como controle positivo da reação utilizou-se o DNA extraído de colônias de M. canis (URM 6273), M. gypseum (URM 6921) e T. mentagrophytes (URM 6211), provenientes da Coleção de Culturas (Micoteca URM), Departamento de Micologia, Centro de Ciências Biológicas da Universidade Federal de Pernambuco (CCB/UFPE). Como controles negativos de reação, utilizou-se água destilada esterilizada e DNA extraído de Alternaria sp. para verificar a especificidade dos primers. Do total de amostras analisadas, 15 (6%) foram identificadas, em cultura, como dermatófitos, e destas, 10 foram M. canis, três M. gypseum e dois T. mentagrophytes (complexo). Destas 15 amostras positivas, 11 (73,3%) foram detectadas por meio da mPCR. Além destas, seis outras, negativas em cultura, foram identificadas como M. gypseum. Verificou-se uma boa concordância entre os resultados da cultura e mPCR (Kappa: 0,66). O protocolo padronizado neste estudo pode ser utilizado como um método de triagem, por apresentar uma sensibilidade maior que a da cultura, usado paralelamente aos exames de rotina, permitindo um diagnóstico em menor tempo

    Serological response to Neospora caninum infection in goats and agreement between three diagnostic techniques to detect caprine neosporosis

    ABSTRACT: The present study aimed to measure the serological response of goats infected with Neospora caninum by assessing the diagnostic performance and agreement between three techniques (indirect immunofluorescent antibody test, IFAT; Neospora agglutitation test, NAT; enzyme-linked immunosorbent assay, ELISA). The panel of sera were comprised of 500 samples of goats, and 60 reference serum samples. These reference and field serum samples were tested by ELISA, NAT, and IFAT. In the field serum samples tested, the seroprevalences of anti-N. caninum antibodies were 3.2%, 4.6%, and 6.4% in the NAT, IFAT and ELISA, respectively. Using the IFAT as the gold standard, the NAT and the ELISA agreement was considered weak (k=0.28) and strong (k=0.75), respectively. When the IFAT performance was used for comparison purposes, the ELISA showed 91.3% sensitivity and 97.7%, specificity with a PPV of 65.2% and a NPV of 99.6%; The NAT presented sensitivity of 26.1% and specificity of 97.9% with a PPV of 37.5% and a NPV of 96.5%. Accordingly, the IFAT should remain the assay of choice for studies about N. caninum infection in goats in individual serum samples. A combination of serological assays with high sensitivity and specificity is recommended in serosurveys of caprine neosporosis

    Effects of training loads on stress tolerance and mucosal immunity in high-intensity functional fitness athletes

    Purpose: This study aimed to analyze the effects of training load on stress tolerance (ST) and secretory immunoglobulin A (SIgA) in male and female high-intensity functional fitness (HIFF) athletes during two different 10 and consecutive weekly training volume loads [higher (week 1) and lower volume (week 2)]. Methods: 14 athletes [7 males: 29.3 (+/- 5.8) years; 86.3 (+/- 8.2) kg and 176.8 (+/- 3.8) cm and 7 females: 32.7 (+/- 4.4) years; 60.0 (+/- 6.7) kg and 162.5 (+/- 5.9) cm] participated. The ST, assessed by Daily Analysis of Life Demand in Athletes questionnaire (DALDA) and Saliva sampling were performed in four time-points (pre (T1) and post (T2) week 1; pre (T3) and post (T4) week 2). Results: Female athletes showed a decrease in ST (symptoms of stress) from 15 T1 to T3 [F-(3,F-36) = 7.184, p .05). There is a significant difference of SIgA concentration [F-(3.36) = 3.551; p = .024; eta p(2) = 0.228], with a significant decrease in female athletes group in T2 compared to T1 (p = .013) and T4 (p = .023). In addition, the different training volume loads did not impact mucosal immunity in male athletes (p > .05). Conclusion: The current findings suggest that higher HIFF volume results in decreased ST and SIgA concentration in female 20 athletes and a subsequent decrease in training volume loads contributed to restoring these variables

    The Effect of Particulate Matter Exposure on the Inflammatory Airway Response of Street Runners and Sedentary People

    Physical exercise promotes many health benefits. However, its effects are not well known in a polluted environment. Thus, this study aimed to compare upper airway inflammatory responses between street runners and sedentary individuals. Twenty-eight volunteers were recruited: runners (n = 14) and sedentary individuals (n = 14), who lived and worked in the same metropolitan area of São Paulo, Brazil. Particulate matter (PM) levels were monitored ten weeks before winter (low PM levels) and ten weeks after the beginning of winter (high PM levels) [PM10 (p < 0.0001) and PM2.5 (p < 0.0001)]. The cytokines (TNF-α, IL-6, IL-10, and IL-17A) levels in the nasal lavage and fractional exhaled nitric oxide (FeNO) were taken at the beginning of the winter (baseline) and ten weeks afterwards (after ten weeks of high PM exposure). IL-6 concentration increased in both runners (p = 0.037) and sedentary individuals (p = 0.027) after high PM exposure compared to the baseline. IL-10 concentration increased in sedentary individuals (p = 0.037) while IL-17A levels were increased in runners (p = 0.001) after high PM exposure compared to the baseline. FeNO levels decreased in runners (p = 0.025) after high PM exposure compared to the baseline. Outdoor endurance training acts as an inducer of a differentiated immune response in the upper airways of runners compared to individuals with a sedentary lifestyle from the same community after elevated PM exposure.status: publishe
