A mission as complex as the Space Exploration Initiative (SEI) cannot succeed without adhering to sound principles in the planning, development, and execution of logistics support for the exploration crews and their mission equipment. While much attention will focus upon the development of reliable, robust, heavy lift launch vehicles, and scientific, technological breakthroughs for SEI, of equal concern is the supportability and sustainability of systems designed for mission operations and crew life support on the lunar and Martian surfaces
Evaluation of harvest time/temperature and storage temperature on postharvest incidence of red drupelet reversion development and firmness of blackberry (Rubus L. subgenus Rubus Watson)
Since 1964, the University of Arkansas blackberry breeding program has worked to improve fruit quality and shipping capabilities. A major limitation in blackberry fruit is postharvest handling potential for the shipping market. Maintaining fruit firmness in storage is crucial. Red drupelet reversion (or simply reversion) is also an important postharvest disorder in which drupelets change from black to red during storage. It is hypothesized that reversion is increased when fruit is picked at hot temperatures and exposed to a rapid change of temperature. These studies evaluated harvest time/temperature, as well as storage temperature, on berry firmness and the incidence of reversion. In Study One, eight genotypes were evaluated. Fruit was harvested at four harvest times (7:00 AM, 10:00 AM, 1:00 PM and 4:00 PM) and then stored for 7 d at 5 °C before evaluation. Results indicated significant sources of variation were genotype and time of harvest for the variables compression (a measure of firmness) and incidence of reversion. Breeding selection A-2453T maintained high firmness and low incidence of reversion after storage compared to other genotypes. Reversion was also significantly lower at the 7:00 AM harvest time compared to later harvests. Study Two included two genotypes harvested at 7:00 AM and 1:00 PM which were evaluated at different storage temperatures (5 and 1 °C). No significant effects were found; however, trends suggested that A-2453T maintained higher firmness despite storage temperature. These studies confirm differences in firmness and reversion among genotypes as well as reveal harvest time impact on reversion
Chemical Analysis Results for Potable Water from ISS Expeditions 21 to 25
The Johnson Space Center Water and Food Analytical Laboratory (WAFAL) performed detailed ground-based analyses of archival water samples for verification of the chemical quality of the International Space Station (ISS) potable water supplies for Expeditions 21 to 25. Over a 14-month period, the Space Shuttle visited the ISS on five occasions to complete construction and deliver supplies. The onboard supplies of potable water available for consumption by the Expeditions 21 to 25 crews consisted of Russian ground-supplied potable water, Russian potable water regenerated from humidity condensate, and US potable water recovered from urine distillate and condensate. Chemical archival water samples that were collected with U.S. hardware during Expeditions 21 to 25 were returned on Shuttle flights STS-129 (ULF3), STS-130 (20A), STS-131 (19A), STS-132 (ULF4) and STS-133 (ULF5), as well as on Soyuz flights 19-22. This paper reports the analytical results for the returned archival water samples and evaluates their compliance with ISS water quality standards. The WAFAL also received and analyzed aliquots of some Russian potable water samples collected in-flight and pre-flight samples of Rodnik potable water delivered to the Station on the Russian Progress vehicle during Expeditions 21 to 25. These additional analytical results are also reported and discussed in this paper

Immune factors preceding diagnosis of glioma: a Prostate Lung Colorectal Ovarian Cancer Screening Trial nested case-control study.
BackgroundEpidemiological studies of adult glioma have identified genetic and environmental risk factors, but much remains unclear. The aim of the current study was to evaluate anthropometric, disease-related, and prediagnostic immune-related factors for relationship with glioma risk.MethodsWe conducted a nested case-control study among the intervention arm of the Prostate, Lung, Colorectal, and Ovarian Cancer (PLCO) Screening Trial. One hundred and twenty-four glioma cases were identified and each matched to four controls. Baseline characteristics were collected at enrollment and were evaluated for association with glioma status. Serum specimens were collected at yearly intervals and were analyzed for immune-related factors including TGF-β1, TNF-α, total IgE, and allergen-specific IgE. Immune factors were evaluated at baseline in a multivariate conditional logistic regression model, along with one additional model that incorporated the latest available measurement.ResultsA family history of glioma among first-degree relatives was associated with increased glioma risk (OR = 4.41, P = .002). In multivariate modeling of immune factors at baseline, increased respiratory allergen-specific IgE was inversely associated with glioma risk (OR for allergen-specific IgE > 0.35 PAU/L: 0.59, P = .03). A logistic regression model that incorporated the latest available measurements found a similar association for allergen-specific IgE (P = .005) and showed that elevated TGF-β1 was associated with increased glioma risk (P-value for trend <.0001).ConclusionThe results from this prospective prediagnostic study suggest that several immune-related factors are associated with glioma risk. The association observed for TGF-β1 when sampling closer to the time of diagnosis may reflect the nascent brain tumor's feedback on immune function
ISS Potable Water Quality for Expeditions 26 through 30
International Space Station (ISS) Expeditions 26-30 spanned a 16-month period beginning in November of 2010 wherein the final 3 flights of the Space Shuttle program finished ISS construction and delivered supplies to support the post-shuttle era of station operations. Expedition crews relied on several sources of potable water during this period, including water recovered from urine distillate and humidity condensate by the U.S. water processor, water regenerated from humidity condensate by the Russian water recovery system, and Russian ground-supplied potable water. Potable water samples collected during Expeditions 26-30 were returned on Shuttle flights STS-133 (ULF5), STS-134 (ULF6), and STS-135 (ULF7), as well as Soyuz flights 24-27. The chemical quality of the ISS potable water supplies continued to be verified by the Johnson Space Center s Water and Food Analytical Laboratory (WAFAL) via analyses of returned water samples. This paper presents the chemical analysis results for water samples returned from Expeditions 26-30 and discusses their compliance with ISS potable water standards. The presence or absence of dimethylsilanediol (DMSD) is specifically addressed, since DMSD was identified as the primary cause of the temporary rise and fall in total organic carbon of the U.S. product water that occurred in the summer of 2010
Discovery and Identification of Dimethylsilanediol as a Contaminant in ISS Potable Water
In September 2010, analysis of ISS potable water samples was undertaken to determine the contaminant(s) responsible for a rise of total organic carbon (TOC) in the Water Processor Assembly (WPA) product water. As analysis of the routine target list of organic compounds did not reveal the contaminant, efforts to look for unknown compounds were initiated, resulting in discovery of an unknown peak in the gas chromatography/mass spectrometry (GC/MS) analysis for glycols. A mass spectrum of the contaminant was then generated by concentrating one of the samples and analyzing it by GC/MS in full-scan mode. Although a computer match of the compound identity could not be obtained with the instrument database, a search with a more up-to-date mass spectral library yielded a good match with dimethylsilanediol (DMSD). Inductively coupled plasma/mass spectrometry (ICP/MS) analyses showed abnormally high silicon levels in the samples, confirming that the unknown compound(s) contained silicon. DMSD was then synthesized to confirm the identification and provide a standard to develop a calibration curve. Further confirmation was provided by external direct analysis in real time time of flight (DART TOF) mass spectrometry. To routinely test for DMSD in the future, a quantitative method was needed. A preliminary GC/MS method was developed and archived samples from various locations on ISS were analyzed to determine the extent of the contamination and provide data for troubleshooting. This paper describes these events in more detail as well as problems encountered in routine GC/MS analyses and the subsequent development of high performance liquid chromatography and LC/MS/MS methods for measuring DMSD
The diffusion of simple penetrants in tangent site polymer melts
The diffusive behavior of penetrants in simple polymer melts was investigated by molecular dynamics simulation. For the case where the polymer melt consisted of pearl-necklace chains, the diffusive behavior of the loose pearl penetrants was seen to be qualitatively different than would be expected in realistic models of polymer melts. In particular, there was little or no ''non-Fickiano'' region; the variation of the diffusion coefficient with the penetrant diameter was what one would expect for diffusion through small molecular liquids; and, finally, the long time tail of the velocity auto correlation displayed a ''-3/2'' power law form, also as in the small molecular liquid case. When the chains' backbone motion was further constrained by the introduction of a bond angle potential, the qualitative nature of the penetrant diffusion became more ''polymer-like''. A non-Fickian region developed; the diffusion coefficient varied more rapidly with penetrant diameter; and the velocity autocorrelation function developed a ''-5/2'' power law tail as would be expected for the diffusion of particles with a wide distribution of trapping times

In-Flight Water Quality Monitoring on the International Space Station (ISS): Measuring Biocide Concentrations with Colorimetric Solid Phase Extraction (CSPE)
The colorimetric water quality monitoring kit (CWQMK) was delivered to the International Space Station (ISS) on STS-128/17A and was initially deployed in September 2009. The kit was flown as a station development test objective (SDTO) experiment to evaluate the acceptability of colorimetric solid phase extraction (CSPE) technology for routine water quality monitoring on the ISS. During the SDTO experiment, water samples from the U.S. water processor assembly (WPA), the U.S. potable water dispenser (PWD), and the Russian system for dispensing ground-supplied water (SVO-ZV) were collected and analyzed with the CWQMK. Samples from the U.S. segment of the ISS were analyzed for molecular iodine, which is the biocide added to water in the WPA. Samples from the SVOZV system were analyzed for ionic silver, the biocide used on the Russian segment of the ISS. In all, thirteen in-flight analysis sessions were completed as part of the SDTO experiment. This paper provides an overview of the experiment and reports the results obtained with the CWQMK. The forward plan for certifying the CWQMK as operational hardware and expanding the capabilities of the kit are also discussed
Gemcitabine and Irinotecan for Patients with Untreated Extensive Stage Small Cell Lung Cancer: SWOG 0119
IntroductionTo evaluate the activity of a nonplatinum-, nonetoposide-containing regimen for patients with extensive stage small cell lung cancer.MethodsPatients with untreated extensive stage small cell lung cancer were treated with gemcitabine 1000 mg/m2 and irinotecan 100 mg/m2 on days 1 and 8 of a 21-day cycle for a maximum of six cycles. Patients with brain metastases were eligible if asymptomatic or controlled after radiation.ResultsEighty-four eligible patients with untreated extensive stage small cell lung cancer with adequate organ function and a performance status of 0–2 were accrued. The median age was 64 years (range, 42–85) and 45 (54%) were women. Six cycles were completed by 28 (33%) patients. Some degree of diarrhea occurred in 57% (grade 3/4, 18%). Other grade 3/4 toxicities were neutropenia (26%), anemia (10%), thrombocytopenia (8%), febrile neutropenia (5%), fatigue (11%), nausea (10%), and vomiting (8%). The response rate was 32% (95% confidence interval: 22%–43%) among the 81 patients with measurable disease. The median survival was 8.5 months (95% confidence interval: 7.0–9.8) with 1- and 2-year survival rates of 26% and 7%, respectively. Salvage therapy data were captured by prospective collection, and only 50% of patients were treated secondarily.ConclusionThe overall response rate with the combination of gemcitabine and irinotecan was disappointing, and the median survival rate was lower than expected. Further development of this combination in small cell lung cancer is not recommended
