    Recruitment Facilitation and Spatial Pattern Formation in Soft-Bottom Mussel Beds

    Mussels (Mytilus edulis) build massive, spatially complex, biogenic structures that alter the biotic and abiotic environment and provide a variety of ecosystem services. Unlike rocky shores, where mussels can attach to the primary substrate, soft sediments are unsuitable for mussel attachment. We used a simple lattice model, field sampling, and field and laboratory experiments to examine facilitation of recruitment (i.e., preferential larval, juvenile, and adult attachment to mussel biogenic structure) and its role in the development of power-law spatial patterns observed in Maine, USA, soft-bottom mussel beds. The model demonstrated that recruitment facilitation produces power-law spatial structure similar to that in natural beds. Field results provided strong evidence for facilitation of recruitment to other mussels—they do not simply map onto a hard-substrate template of gravel and shell hash. Mussels were spatially decoupled from non-mussel hard substrates to which they can potentially recruit. Recent larval recruits were positively correlated with adult mussels, but not with other hard substrates. Mussels made byssal thread attachments to other mussels in much higher proportions than to other hard substrates. In a field experiment, mussel recruitment was highest to live mussels, followed by mussel shell hash and gravel, with almost no recruitment to muddy sand. In a laboratory experiment, evenly dispersed mussels rapidly self-organized into power-law clusters similar to those observed in nature. Collectively, the results indicate that facilitation of recruitment to existing mussels plays a major role in soft-bottom spatial pattern development. The interaction between large-scale resource availability (hard substrate) and local-scale recruitment facilitation may be responsible for creating complex power-law spatial structure in soft-bottom mussel beds

    Theory of High-Tc Superconductivity: Accurate Predictions of Tc

    The superconducting transition temperatures of high-Tc compounds based on copper, iron, ruthenium and certain organic molecules are discovered to be dependent on bond lengths, ionic valences, and Coulomb coupling between electronic bands in adjacent, spatially separated layers [1]. Optimal transition temperature, denoted as T_c0, is given by the universal expression kBTc0=e2Λ/ℓζk_BT_c0 = e^2 \Lambda / \ell\zeta; ℓ\ell is the spacing between interacting charges within the layers, \zeta is the distance between interacting layers and \Lambda is a universal constant, equal to about twice the reduced electron Compton wavelength (suggesting that Compton scattering plays a role in pairing). Non-optimum compounds in which sample degradation is evident typically exhibit Tc < T_c0. For the 31+ optimum compounds tested, the theoretical and experimental T_c0 agree statistically to within +/- 1.4 K. The elemental high Tc building block comprises two adjacent and spatially separated charge layers; the factor e^2/\zeta arises from Coulomb forces between them.     Controlled synthesis of the DSF cell–cell signal is required for biofilm formation and virulence in Xanthomonas campestris

    Virulence of the black rot pathogen Xanthomonas campestris pv. campestris (Xcc) is regulated by cell–cell signalling involving the diffusible signal factor DSF. Synthesis and perception of DSF require products of genes within the rpf cluster (for regulation of pathogenicity factors). RpfF directs DSF synthesis whereas RpfC and RpfG are involved in DSF perception. Here we have examined the role of the rpf/DSF system in biofilm formation in minimal medium using confocal laser-scanning microscopy of GFP-labelled bacteria. Wild-type Xcc formed microcolonies that developed into a structured biofilm. In contrast, an rpfF mutant (DSF-minus) and an rpfC mutant (DSF overproducer) formed only unstructured arrangements of bacteria. A gumB mutant, defective in xanthan biosynthesis, was also unable to develop the typical wild-type biofilm. Mixed cultures of gumB and rpfF mutants formed a typical biofilm in vitro. In contrast, in mixed cultures the rpfC mutant prevented the formation of the structured biofilm by the wild-type and did not restore wild-type biofilm phenotypes to gumB or rpfF mutants. These effects on structured biofilm formation were correlated with growth and disease development by Xcc strains in Nicotiana benthamiana leaves. These findings suggest that DSF signalling is finely balanced during both biofilm formation and virulence

    Intelligent Liver Function Testing (iLFT):A trial of automated diagnosis and staging of liver disease in Primary Care

    Background: Liver function tests (LFTs) are frequently requested blood tests which may indicate liver disease. LFTs are commonly abnormal, the causes of which can be complex and frequently under investigated. This can lead to missed opportunities to diagnose and treat liver disease at an early stage. We developed an automated investigation algorithm, which would maximise early diagnosis of liver related diseases. Our aim was to determine whether this new pathway of care, Intelligent Liver Function testing (iLFT) increased diagnosis of liver disease and was cost-effective. Methods: We developed an automated system that further investigated abnormal LFTs on initial testing samples to generate a probable diagnosis and management plan. We integrated an automated investigation algorithm into the laboratory management system, based on minimal diagnostic criteria, liver fibrosis estimation, and reflex testing for causes of liver disease. This algorithm then generated a diagnosis and/or management plan. A stepped-wedged trial design was utilised to compare LFT outcomes in General Practices in the 6 months before and after introduction of the iLFT system. Diagnostic outcomes were collated and compared. Results: Using iLFT, the diagnosis of liver disease was increased by 43%. It was cost-effective with a low initial incremental cost-effectiveness ratio (ICER) of £284 per correct diagnosis, and a saving to the NHS of £3,216 per patient lifetime. Conclusions: iLFT increases liver diagnosis, improves quality of care, and is highly cost-effective. This can be achieved with minor changes to working practices and exploitation of functionality existing within modern laboratory diagnostics systems. Lay Summary: There is a growing epidemic of advanced liver disease, this could be offset by early detection and management. Checking liver blood tests (LFTs) should be an opportunity diagnose liver problems, but abnormal results are often incompletely investigated. In this study we were able to substantially increase the diagnostic yield of the abnormal LFTs using the automated iLFT system. With the addition of referral recommendations and management plans, this strategy provides optimum investigation and management of LFTs and is cost saving to the NHS

    Effects of Cyclin Dependent Kinase 9 inhibition on zebrafish larvae

    CDK9 is a known regulator of cellular transcription, growth and proliferation. Small molecule inhibitors are currently being developed and assessed in clinical trials as anti-cancer drugs. The zebrafish embryo provides an ideal model to explore the effects of CDK9 inhibition in-vivo. This has not been adequately explored previously at the level of a whole organism. We have compared and contrasted the effects of pharmacological and molecular inhibition of CDK9 on somatic growth, apoptosis and cellular proliferation in zebrafish larvae between 0 to 120 hours post fertilisation (hpf) using flavopiridol, a selective CDK9 antagonist, and CDK9-targeting morpholino. We demonstrate that the inhibition of CDK9 diminishes cellular proliferation and increases apoptosis. Subsequently, it affects somatic growth and development of a number of key embryonic structures including the brain, heart, eye and blood vessels. For the first time, we have localized CDK9 at a subcellular level in whole-mounted larvae. This works shows, at a high-throughput level, that CDK9 clearly plays a fundamental role in early cellular growth and proliferation

    The status of GNSS data processing systems to estimate integrated water vapour for use in numerical weather prediction models

    Modern Numerical Weather Prediction (NWP) models make use of the GNSS-derived Zenith Total Delay (ZTD) or Integrated Water Vapour (IWV) estimates to enhance the quality of their forecasts. Usually, the ZTD is assimilated into the NWP models on 3- hourly to 6-hourly intervals but with the advancement of NWP models towards higher update rates e.g. 1-hourly cycling in the Rapid Update Cycle (RUC) NWP, it has become of high interest to estimate ZTD on sub-hourly intervals. In turn, this imposes requirements related to the timeliness and accuracy of the ZTD estimates and has lead to a development of various strategies to process GNSS observations to obtain ZTD with different latencies and accuracies. Using present GNSS products and tools, ZTD can be estimated in real-time (RT), near real-time (NRT) and post-processing (PP) modes. The aim of this study is to provide an overview and accuracy assessment of various RT, NRT, and PP IWV estimation systems and comparing their achieved accuracy with the user requirements for GNSS meteorology. The NRT systems are based on Bernese GPS Software 5.0 and use a double-differencing strategy whereas the PP system is based on the Bernese GNSS Software 5.2 using the precise point positioning (PPP) strategy. The RT systems are based on the BKG Ntrip Client 2.7 and the PPPWizard both using PPP. The PPP-Wizard allows integer ambiguity resolution at a single station and therefore the effect of fixing integer ambiguities on ZTD estimates will also be presented
