    Composites: A viable option

    While it sounded great to be asked to talk about composites, I found it difficult to select subject areas that would be of real interest. My choice is based on saying some things about where the maturity of the composite aircraft structures is today and what that means in terms of future criteria for application. This focus was the basis for my title selection. The other issue that will be addressed was requested by NASA and focuses on composites structures cost. This fits well with the state-of-the-art interpretations I will discuss first, since the cost issue must be viewed from both the current status and future points of view. The difficulty in presenting something in these areas is not in the subjects themselves but in trying to present a real world viewpoint to an audience of composite experts. So, with recognition of the expertise of the audience, I hope you will see something in this presentation about how to view composite aircraft structure

    Durability and damage tolerance of Large Composite Primary Aircraft Structure (LCPAS)

    Analysis and testing addressing the key technology areas of durability and damage tolerance were completed for wing surface panels. The wing of a fuel-efficient, 200-passenger commercial transport airplane for 1990 delivery was sized using graphite-epoxy materials. Coupons of various layups used in the wing sizing were tested in tension, compression, and spectrum fatigue with typical fastener penetrations. The compression strength after barely visible impact damage was determined from coupon and structural element tests. One current material system and one toughened system were evaluated by coupon testing. The results of the coupon and element tests were used to design three distinctly different compression panels meeting the strength, stiffness, and damage-tolerance requirements of the upper wing panels. These three concepts were tested with various amounts of damage ranging from barely visible impact to through-penetration. The results of this program provide the key technology data required to assess the durability and damage-tolerance capability or advanced composites for use in commercial aircraft wing panel structure

    Transition services and their effectiveness

    The purpose of this study was to determine if transition services as they are currently performed in a large suburban regional high school district are effective in helping classified students make the transition to the world beyond high school. A review of current literature on transition shows that services vary in scope and effectiveness from state to state and from district to district. Interviews with district personnel reveal no follow-up once a student has left the district through graduation or aging out. A survey instrument was compiled and mailed to 200 former students who were classified while attending school in the district. The survey sought information on the transition conference, help from local, state and private agencies and their feelings regarding the help they received from district personnel. The results of the survey were mixed with the district doing a good job overall and the state needing to improve its help of these young people

    Structural Qualification of Composite Airframes

    The development of fundamental approaches for predicting failure and elongation characteristics of fibrous composites are summarized in this document. The research described includes a statistical formulation for individual fiber breakage and fragmentation and clustered fiber breakage, termed macrodefects wherein the aligned composite may represent a structural component such as a reinforcing bar element, a rebar. Experimental work conducted in support of the future exploitation of aligned composite rebar elements is also described. This work discusses the experimental challenges associated with rebar tensile test evaluation and describes initial numerical analyses performed in support of the experimental program

    Performance of lactating dairy cattle housed in two-row freestall barns equipped with three different cooling systems

    One hundred fifty-nine Holstein cows (66 primiparous and 93 multiparous) were assigned to each of three different cooling systems installed in two-row freestall barns on a northeast Kansas dairy. One barn was equipped with a row of five 48-inch fans mounted every 40 ft over the freestalls and a row of 10 36-inch fans mounted every 20 ft over the cow feed line. Another barn was equipped with five 48-inch fans mounted over the freestalls. Both of these barns were also equipped with identical sprinkler systems mounted over the feed line. The third barn was equipped with a row of five 48-inch fans mounted over the freestalls. In addition to the sprinklers over the feed line, additional sprinklers were mounted on the back alley of the third barn. Data were collected for an 85- day period to evaluate the three systems under heat stress during the summer of 1999. Cows cooled with these three systems produced similar amounts of milk and consumed nearly equal amounts of feed. Summer heat stress generally reduces milk production 20%, if cooling systems are not installed. Based on this estimated loss, these systems returned over $10,000/pen/year above ownership and operational cost. These results indicated that effective cooling in a two-row freestall barn includes a sprinkler system on the feed line and properly sized and spaced fans over the freestalls

    Glial Cell Adhesion Molecule (GlialCAM) Determines Proliferative versus Invasive Cell States in Glioblastoma

    The malignant brain cancer glioblastoma (GBM) contains groups of highly invasive cells that drive tumor progression as well as recurrence after surgery and chemotherapy. The molecular mechanisms that enable these GBM cells to exit the primary mass and disperse throughout the brain remain largely unknown. Here we report using human tumor specimens and primary spheroids from male and female patients that glial cell adhesion molecule (GlialCAM), which has normal roles in brain astrocytes and is mutated in the developmental brain disorder megalencephalic leukoencephalopathy with subcortical cysts (MLC), is differentially expressed in subpopulations of GBM cells. High levels of GlialCAM promote cell-cell adhesion and a proliferative GBM cell state in the tumor core. In contrast, GBM cells with low levels of GlialCAM display diminished proliferation and enhanced invasion into the surrounding brain parenchyma. RNAi-mediated inhibition of GlialCAM expression leads to activation of proinvasive extracellular matrix adhesion and signaling pathways. Profiling GlialCAM-regulated genes combined with cross-referencing to single-cell transcriptomic datasets validates functional links among GlialCAM, Mlc1, and aquaporin-4 in the invasive cell state. Collectively, these results reveal an important adhesion and signaling axis comprised of GlialCAM and associated proteins including Mlc1 and aquaporin-4 that is critical for control of GBM cell proliferation and invasion status in the brain cancer microenvironment

    Space and transatmospheric propulsion technology

    This report focuses primarily on Japan's programs in liquid rocket propulsion and propulsion for spaceplane and related transatmospheric areas. It refers briefly to Japan's solid rocket programs and to new supersonic air-breathing propulsion efforts. The panel observed that the Japanese had a carefully thought-out plan, a broad-based program, and an ambitious but achievable schedule for propulsion activity. Japan's overall propulsion program is behind that of the United States at the time of this study, but the Japanese are gaining rapidly. The Japanese are at the forefront in such key areas as advanced materials, enjoying a high level of project continuity and funding. Japan's space program has been evolutionary in nature, while the U.S. program has emphasized revolutionary advances. Projects have typically been smaller in Japan than in the United States, focusing on incremental advances in technology, with an excellent record of applying proven technology to new projects. This evolutionary approach, coupled with an ability to take technology off the shelf from other countries, has resulted in relatively low development costs, rapid progress, and enhanced reliability. Clearly Japan is positioned to be a world leader in space and transatmospheric propulsion technology by the year 2000

    Intrathecal long-term gene expression by self-complementary adeno-associated virus type 1 suitable for chronic pain studies in rats

    BACKGROUND: Intrathecal (IT) gene transfer is an attractive approach for targeting spinal mechanisms of nociception but the duration of gene expression achieved by reported methods is short (up to two weeks) impairing their utility in the chronic pain setting. The overall goal of this study was to develop IT gene transfer yielding true long-term transgene expression defined as ≥ 3 mo following a single vector administration. We defined "IT" administration as atraumatic injection into the lumbar cerebrospinal fluid (CSF) modeling a lumbar puncture. Our studies focused on recombinant adeno-associated virus (rAAV), one of the most promising vector types for clinical use. RESULTS: Conventional single stranded rAAV2 vectors performed poorly after IT delivery in rats. Pseudotyping of rAAV with capsids of serotypes 1, 3, and 5 was tested alone or in combination with a modification of the inverted terminal repeat. The former alters vector tropism and the latter allows packaging of self-complementary rAAV (sc-rAAV) vectors. Combining both types of modification led to the identification of sc-rAAV2/l as a vector that performed superiorly in the IT space. IT delivery of 3 × 10e9 sc-rAAV2/l particles per animal led to stable expression of enhanced green fluorescent protein (EGFP) for ≥ 3 mo detectable by Western blotting, quantitative PCR, and in a blinded study by confocal microscopy. Expression was strongest in the cauda equina and the lower sections of the spinal cord and only minimal in the forebrain. Microscopic examination of the SC fixed in situ with intact nerve roots and meninges revealed strong EGFP fluorescence in the nerve roots. CONCLUSION: sc-rAAVl mediates stable IT transgene expression for ≥ 3 mo. Our findings support the underlying hypothesis that IT target cells for gene transfer lack the machinery for efficient conversion of the single-stranded rAAV genome into double-stranded DNA and favor uptake of serotype 1 vectors over 2. Experiments presented here will provide a rational basis for utilizing IT rAAV gene transfer in basic and translational studies on chronic pain
