Atherosclerosis is the leading cause of death in the Western world. Although mitochondrial DNA (mtDNA) damage has been implicated in atherosclerosis, it is unclear whether the damage is sufficient to impair mitochondrial respiration, and mitochondrial dysfunction has not been demonstrated. Treatment of vascular smooth muscle cells (VSMCs) with an atherogenic lipid, oxidised low-density lipoprotein (OxLDL), dose dependently decreased basal and maximal respiration and fat-feeding of apolipoprotein E deficient (ApoE-/-) mice reduced mitochondrial DNA copy number relative to nuclear DNA in aortas. Mitochondrial respiration of ApoE-/- mouse aortas, assessed through a 24-well Seahorse extracellular flux analyser, was not affected prior to the development of atherosclerotic plaques. Developed human carotid atherosclerotic plaques were dissected into defined regions including healthy media, shoulder region, fibrous cap and core and their respiration was investigated. The respiratory reserve capacity (RRC) of the shoulder region was similar to the media. However, the cap RRC was significantly reduced compared to healthy media. In contrast, the extracellular acidification rates (ECAR) of the media, shoulder, cap and core regions were similar. In addition, mtDNA copy number was significantly reduced in tissues derived from human plaques compared to healthy arteries and expression of complexes I and II of the electron transfer chain (ETC) were significantly reduced in plaque VSMCs. OxLDL induced mitophagy in human VSMCs and plaque VSMCs demonstrated increased levels of mitophagy without compensatory upregulation of proteins involved in mitochondrial biogenesis. Understanding the role of mitochondrial metabolism and signalling is important for our understanding of disease progression and may lead to future therapeutic targets
Stochastic topology optimisation with hierarchical tensor reconstruction
A novel approach for risk-averse structural topology optimization under uncertainties is presented which takes into account random material properties and random forces. For the distribution of material, a phase field approach is employed which allows for arbitrary topological changes during optimization. The state equation is assumed to be a high-dimensional PDE parametrized in a (finite) set of random variables. For the examined case, linearized elasticity with a parametric elasticity tensor is used. Instead of an optimization with respect to the expectation of the involved random fields, for practical purposes it is important to design structures which are also robust in case of events that are not the most frequent. As a common risk-aware measure, the Conditional Value at Risk (CVaR) is used in the cost functional during the minimization procedure. Since the treatment of such high-dimensional problems is a numerically challenging task, a representation in the modern hierarchical tensor train format is proposed. In order to obtain this highly efficient representation of the solution of the random state equation, a tensor completion algorithm is employed which only required the pointwise evaluation of solution realizations. The new method is illustrated with numerical examples and compared with a classical Monte Carlo sampling approach
Development steps of 2-color laser-induced fluorescence with MDR-enhanced energy transfer for instantaneous planar temperature measurement of micro-droplets and sprays
[EN] A new method for instantaneous measurement of temperature, size and velocity of micro-droplets has been developed.
The method is based on the well-known 2-color laser-induced fluorescence (2cLIF) technique, but uses
a pulsed laser for 2-dimensional imaging without motion blur and an adjusted dye mixture for suppression of LIFMDRs
by utilizing the MDR-enhanced energy transfer effect. This work presents the development steps that are

Non-intrusive tensor reconstruction for high dimensional random PDEs
This paper examines a completely non-intrusive, sample-based method for the computation of functional low-rank solutions of high dimensional parametric random PDEs which have become an area of intensive research in Uncertainty Quantification (UQ). In order to obtain a generalized polynomial chaos representation of the approximate stochastic solution, a novel black-box rank-adapted tensor reconstruction procedure is proposed. CommunityHub: Potenzialanalyse für die gemeinschaftliche Nutzung innerstädtischer Logistikflächen
Problemstellung:
Durch kontinuierliche Urbanisierungsprozesse und die voranschreitende Wohnraumverdichtung sowie die
Zunahme der Bevölkerungs- und Verkehrsdichte ist ein sparsamer Umgang mit der Ressource Boden von
Nöten. Die steigende Bedeutung des E-Commerce sowie die sich daraus resultierende Veränderung von
Konsumentinnen- und Konsumentenanforderungen und zunehmenden Anzahl an Sendungen stellt vor allem
die innerstädtische Logistik im Hinblick auf eine effiziente Abwicklung bzw. Organisation der First- und
Last-Mile vor eine groĂźe Herausforderung.
Beim Zusammentreffen unterschiedlicher Akteurinnen und Akteure (Stadt, Endnutzerinnen und Endnutzer,
Wirtschaft) sind – bedingt durch verschiedene Interessen und Zielsetzungen – Entwicklungs- und
Nutzungskonflikte – im Speziellen bei knappem Platzangebot – kaum vermeidbar. Es existieren jedoch auch
im urbanen Raum ungenutzte Logistikkapazitäten, die durch ungleichmäßige Aufteilung von Wohn- und
Lagerraum entstehen. Besonderes Augenmerk wird hierbei auf leerstehende ErdgeschoĂźzonen gelegt.
Zielsetzung:
Das Projekt CommunityHub1 zielt auf eine optimale Allokation der Ressource Raum ab. Unter dem Begriff
CommunityHub werden Mikro-Logistikknotenpunkte verstanden, welche eine Versorgung der Bevölkerung
mit logistischen Dienstleistungen und eine gleichberechtigte Zugänglichkeit für alle Akteurinnen und
Akteure (Stadt, Endnutzerinnen und Endnutzer, Wirtschaft) gewährleisten. Dabei werden in städtischen
ErdgeschoĂźzonen logistische Leistungen (Lagerung, Paketzustellung etc.) mit weiteren koppelbaren
(Dienst-)Leistungen (Entsorgung, Versicherungen, Umkleidekabinen etc.) bereit gestellt.
Die unterschiedlichen Komponenten der Partizipation, Nahversorgung und Mehrfachnutzung existieren fĂĽr
sich allein, wurden bisher jedoch noch nicht im Rahmen eines innovativen Konzeptes vereint. Mittels
Primär- und Sekundärdatenerhebung wird die Grundlage für die Durchführung einer Potenzialanalyse am
Anwendungsbeispiel Wien und die Untersuchung möglicher Risiken von CommunityHubs geschaffen.
Darauf aufbauend werden konkrete Lösungsvorschläge und Implementierungsstrategien für
(inner-)städtische Multi-Use-Konzepte erarbeitet.
Ergebnisse:
Auf diese Art kann das Projekt CommunityHub einen Beitrag für die Lösung der First- bzw. Last-Mile-
Problematik leisten, wodurch die Zustellbarkeit von Paketen erhöht und gleichzeitig CO2-Emissionen (z. B.
durch Bündelung von Ressourcen) reduziert werden können. Des Weiteren werden durch die Umnutzung
von – z. B. leerstehenden Geschäfts- und Bankfilialen zu (inner-)städtischen CommunityHubs –
ErdgeschoĂźzonen aufgewertet und die Nahversorgung im urbanen Raum sichergestellt
