    The rubber hand universe:On the impact of methodological differences in the rubber hand illusion

    The rubber hand illusion (RHI) is a widely applied paradigm to investigate changes in body representations. Extensive scientific interest has produced a great variability in the observed results and many contradictory findings have been reported. Taking into account the numerous variations in the experimental implementation of the RHI, many of these contradictive findings can be reconciled, but to date a thorough analysis of the methodological differences between RHI studies is lacking. Here we summarize and analyse methodological differences between RHI studies. In distinction from other reviews focusing on the integration of findings from various studies, the present paper is devoted to the differences in (i) the experimental setup, (ii) the method used to induce the RHI, (iii) the quantification of its effects, and (iv) aspects of the experimental design and data analysis. This approach will provide a reference frame for the interpretation of previous studies as well as for the design of future studies

    Pyeloplasty in children: is there a difference in patients with or without crossing lower pole vessel?

    Introduction: Most of the children with hydronephrosis do not require any surgical intervention. However, in individual cases, irreversible loss of renal function can develop. Predictive criteria have been proven ineffective so far in determining in which children obstruction will lead to renal damage. The aim of our retrospective study was to determine the role of a crossing lower pole vessel (CV) in children undergoing pyeloplasty. Materials and methods: Between 1996 and 2003, 137 patients (age between 6weeks and 16years) with unilateral ureteropelvic junction obstruction and no associated urological pathologies underwent Anderson-Hynes dismembered pyeloplasty. A total of 112 patients were evaluated with complete data. One of the following criteria was considered to be indication for surgery in children with grade 4 hydronephrosis: differential renal function (DRF) 10% and washout patterns II or III b according to O'Reilly. We looked at the age during surgery and the kind of presentation. DRF was measured using diuretic renography preoperatively and 1year postoperatively. A postoperative change in DRF of group A (children without CV, n=84) was compared to that in group B (children with CV, n=28). Results: Median age at the time of surgery was 5months in group A compared to 23months in group B. Only in 21.4% of the children with CV compared to 60.7% without CV hydronephrosis was diagnosed by ultrasound examination antenatally. We found a preoperative DRF of 42.4%±11.2 SD in group A, and of 38.9%±11.7 SD in group B. The percentage of postoperative improvement was 3.3% in group A and 15.4% in group B. Conclusions: Children with ureteropelvic junction obstruction and CV received a delayed surgical treatment and showed a greater reduction in differential renal function preoperatively, in contrast to patients without CV. Our data show that CV is a risk factor for deterioration of renal function in children with hydronephrosis and we advocate for an early pyeloplasty in these children, especially if they have a high-grade dilatation and equivocal washout patterns in diuretic renographies. Further prospective studies are necessary in order to understand the natural history of CV and to reveal the importance of the crossing lower pole vessel as a structural anomaly lacking maturatio

    Collection and classification of influence parameters for safety effectiveness of ADAS

    Virtual scenario-based testing has become an acceptable method for evaluating safety effectiveness of advanced driver assistance systems (ADAS). Due to the complexity of the ADAS operating environment, the scenarios that an ADAS could face are almost infinite. Therefore, it is crucial to find critical scenarios to improve the efficiency of testing without compromising credibility. One popular method is to explore the parameterized scenario space using various intelligent search methods. Selecting parameters to parameterize the scenario space is particularly important to achieve good coverage and high efficiency. However, an extensive collection of (relevant) influence parameters is missing, which allows a thorough consideration when selecting parameters regarding specific scenarios. In addition, the general importance definition for individual influence parameters is not provided, regarding the potential influence of their variations on the safety effectiveness of ADAS, which can also be used as a reference while selecting parameters. Combining knowledge from different sources (the published literature, standardized test scenarios, accident analysis, autonomous vehicle disengagement, accident reports, and specific online surveys), this paper has summarized, in total, 94 influence parameters, given the general definitions of importance for 77 influence parameters based on cluster analysis algorithms. The list of influence parameters provides researchers and system developers a comprehensive basis for pre-selecting influence parameters for evaluating the safety effectiveness of ADAS by virtual scenario-based testing and helps check whether certain influence parameters can be a meaningful extension for the evaluation

    SWEET11b transports both sugar and cytokinin in developing barley grains

    Even though Sugars Will Eventually be Exported Transporters (SWEETs) have been found in every sequenced plant genome, a comprehensive understanding of their functionality is lacking. In this study, we focused on the SWEET family of barley (Hordeum vulgare). A radiotracer assay revealed that expressing HvSWEET11b in African clawed frog (Xenopus laevis) oocytes facilitated the bidirectional transfer of not only just sucrose and glucose, but also cytokinin. Barley plants harboring a loss-of-function mutation of HvSWEET11b could not set viable grains, while the distribution of sucrose and cytokinin was altered in developing grains of plants in which the gene was knocked down. Sucrose allocation within transgenic grains was disrupted, which is consistent with the changes to the cytokinin gradient across grains, as visualized by magnetic resonance imaging and Fourier transform infrared spectroscopy microimaging. Decreasing HvSWEET11b expression in developing grains reduced overall grain size, sink strength, the number of endopolyploid endosperm cells, and the contents of starch and protein. The control exerted by HvSWEET11b over sugars and cytokinins likely predetermines their synergy, resulting in adjustments to the grain's biochemistry and transcriptome

    A selective and orally bioavailable VHL-recruiting PROTAC achieves SMARCA2 degradation in vivo

    Targeted protein degradation offers an alternative modality to classical inhibition and holds the promise of addressing previously undruggable targets to provide novel therapeutic options for patients. Heterobifunctional molecules co-recruit a target protein and an E3 ligase, resulting in ubiquitylation and proteosome-dependent degradation of the target. In the clinic, the oral route of administration is the option of choice but has only been achieved so far by CRBN- recruiting bifunctional degrader molecules. We aimed to achieve orally bioavailable molecules that selectively degrade the BAF Chromatin Remodelling complex ATPase SMARCA2 over its closely related paralogue SMARCA4, to allow in vivo evaluation of the synthetic lethality concept of SMARCA2 dependency in SMARCA4-deficient cancers. Here we outline structure- and property-guided approaches that led to orally bioavailable VHL-recruiting degraders. Our tool compound, ACBI2, shows selective degradation of SMARCA2 over SMARCA4 in ex vivo human whole blood assays and in vivo efficacy in SMARCA4-deficient cancer models. This study demonstrates the feasibility for broadening the E3 ligase and physicochemical space that can be utilised for achieving oral efficacy with bifunctional molecules
