    Sexuality Generates Diversity in the Aflatoxin Gene Cluster: Evidence on a Global Scale

    Aflatoxins are produced by Aspergillus flavus and A. parasiticus in oil-rich seed and grain crops and are a serious problem in agriculture, with aflatoxin B₁ being the most carcinogenic natural compound known. Sexual reproduction in these species occurs between individuals belonging to different vegetative compatibility groups (VCGs). We examined natural genetic variation in 758 isolates of A. flavus, A. parasiticus and A. minisclerotigenes sampled from single peanut fields in the United States (Georgia), Africa (Benin), Argentina (CĂłrdoba), Australia (Queensland) and India (Karnataka). Analysis of DNA sequence variation across multiple intergenic regions in the aflatoxin gene clusters of A. flavus, A. parasiticus and A. minisclerotigenes revealed significant linkage disequilibrium (LD) organized into distinct blocks that are conserved across different localities, suggesting that genetic recombination is nonrandom and a global occurrence. To assess the contributions of asexual and sexual reproduction to fixation and maintenance of toxin chemotype diversity in populations from each locality/species, we tested the null hypothesis of an equal number of MAT1-1 and MAT1-2 mating-type individuals, which is indicative of a sexually recombining population. All samples were clone-corrected using multi-locus sequence typing which associates closely with VCG. For both A. flavus and A. parasiticus, when the proportions of MAT1-1 and MAT1-2 were significantly different, there was more extensive LD in the aflatoxin cluster and populations were fixed for specific toxin chemotype classes, either the non-aflatoxigenic class in A. flavus or the B₁-dominant and G₁-dominant classes in A. parasiticus. A mating type ratio close to 1∶1 in A. flavus, A. parasiticus and A. minisclerotigenes was associated with higher recombination rates in the aflatoxin cluster and less pronounced chemotype differences in populations. This work shows that the reproductive nature of the population (more sexual versus more asexual) is predictive of aflatoxin chemotype diversity in these agriculturally important fungi

    Red-Cockaded Woodpecker Status and Management: West Gulf Coastal Plain and Interior Highlands

    Red-cockaded woodpecker populations declined precipitously following European settlement and expansion and cutting of the original pine forests across the southeastern United States. By 1990 most residual populations lacked demographic viability, existed in degraded habitat, and were isolated from other populations. The primary causes of this situation were harvest of the original pine forests of the southeastern United States, conversion of forested lands to other uses, short-rotation silvicultural practices, and alteration of the fire regime in the regenerated forests. As social and legal mandates changed, management of red-cockaded woodpeckers became a higher priority. Intensive management for red-cockaded woodpeckers is currently practiced on most public and a few private lands that still support populations. Recent population trends and the current status of red-cockaded woodpeckers in Oklahoma, Arkansas, Texas, and Louisiana reflect historical factors and the efficacy of recent management

    Reflections on Sculptural Thinking in Fashion Design

    This paper explores three-dimensional thinking in fashion design; it does this by engaging with theories, concepts and philosophies related to thought and the experience of creating three-dimensional artifacts, which are common to both sculpture and fashion. Central to this relationship is the employment of the senses with respect to perception and cognition. Of particular interest is the sense of touch,and how sensory experience encounters notions of empathy and mimicry in a phenomenological encounter with others: whether animate or inanimate. The research emerged through conversations between a fashion designer, Kevin Almond and a contemporary artist, Stephen Swindells. The sensibility of the paper, and much ofthe analysis and debates, thus adopt a creative practitioners perspective. A conceptual current running through the conversation, and subsequently the paper, touched upon whether following a line of thought becomes analogous to visually and mentally tracing a human form in a psychological space – and what is the significance for fashion of the interrelationships between sculptural thinking and phenomenological encounters with others within urban environments

    Evaluation of Services Provided to Pregnant African Refugees by Catholic Social Services Refugee Resettlement Agency in Dayton Ohio

    In 2016 Public Health - Dayton and Montgomery County declared birth outcomes the number one priority in their Community Health Improvement Plan. To improve the infant mortality rate (IMR), racial disparities must be address and considerations should be given to Dayton’s newest and most vulnerable neighbors. Dayton receives between 200 and 250 refugees each year, 70% of whom are from the Democratic Republic of the Congo. African refugees have higher infant mortally and poorer birth outcomes than native populations in the U.S. Early and regular prenatal care is protective against infant mortality, and African refugees consistently report delayed and infrequent prenatal care. Five categories affect utilization of prenatal services by African refugees; migration factors, cultural factors, treatment and communication factors, socioeconomic and accessibility factors, and social and psychological factors. This paper describes the services that Catholic Social Services (CSS) Refugee Resettlement Agency provides to newly-arrived pregnant refugees and compares these services to published guidelines and best practice models. When compared, CSS provide adequate services to address socioeconomic and accessibility factors through funding provided by the Department of State. While CSS provided some services towards the other four categories, none of them are comprehensive. The acculturation process provides an opportunity for CSS to address migration and cultural factors. But it is through strong community partnerships with local stakeholders that CSS can provide comprehensive services to address all the factors that affect prenatal care utilization in African refugees

    Full Genome Characterization of the Culicoides-Borne Marsupial Orbiviruses: Wallal Virus, Mudjinbarry Virus and Warrego Viruses

    Viruses belonging to the species Wallal virus and Warrego virus of the genus Orbivirus were identified as causative agents of blindness in marsupials in Australia during 1994/5. Recent comparisons of nucleotide (nt) and amino acid (aa) sequences have provided a basis for the grouping and classification of orbivirus isolates. However, full-genome sequence data are not available for representatives of all Orbivirus species. We report full-genome sequence data for three additional orbiviruses: Wallal virus (WALV); Mudjinabarry virus (MUDV) and Warrego virus (WARV). Comparisons of conserved polymerase (Pol), sub-core-shell 'T2' and core-surface 'T13' proteins show that these viruses group with other Culicoides borne orbiviruses, clustering with Eubenangee virus (EUBV), another orbivirus infecting marsupials. WARV shares <70% aa identity in all three conserved proteins (Pol, T2 and T13) with other orbiviruses, consistent with its classification within a distinct Orbivirus species. Although WALV and MUDV share <72.86%/67.93% aa/nt identity with other orbiviruses in Pol, T2 and T13, they share >99%/90% aa/nt identities with each other (consistent with membership of the same virus species - Wallal virus). However, WALV and MUDV share <68% aa identity in their larger outer capsid protein VP2(OC1), consistent with membership of different serotypes within the species - WALV-1 and WALV-2 respectively

    Simultaneous assessment of acidogenesis-mitigation and specific bacterial growth-inhibition by dentifrices

    Dentifrices can augment oral hygiene by inactivating bacteria and at sub-lethal concentrations may affect bacterial metabolism, potentially inhibiting acidogenesis, the main cause of caries. Reported herein is the development of a rapid method to simultaneously measure group-specific bactericidal and acidogenesis-mitigation effects of dentifrices on oral bacteria. Saliva was incubated aerobically and anaerobically in Tryptone Soya Broth, Wilkins-Chalgren Broth with mucin, or artificial saliva and was exposed to dentifrices containing triclosan/copolymer (TD); sodium fluoride (FD); stannous fluoride and zinc lactate (SFD1); or stannous fluoride, zinc lactate and stannous chloride (SFD2). Minimum inhibitory concentrations (MIC) were determined turbidometrically whilst group-specific minimum bactericidal concentrations (MBC) were assessed using growth media and conditions selective for total aerobes, total anaerobes, streptococci and Gram-negative anaerobes. Minimum acid neutralization concentration (MNC) was defined as the lowest concentration of dentifrice at which acidification was inhibited. Differences between MIC and MNC were calculated and normalized with respect to MIC to derive the combined inhibitory and neutralizing capacity (CINC), a cumulative measure of acidogenesis-mitigation and growth inhibition. The overall rank order for growth inhibition potency (MIC) under aerobic and anaerobic conditions was: TD> SFD2> SFD1> FD. Acidogenesis-mitigation (MNC) was ordered; TD> FD> SFD2> SFD1. CINC was ordered TD> FD> SFD2> SFD1 aerobically and TD> FD> SFD1> SFD2 anaerobically. With respect to group-specific bactericidal activity, TD generally exhibited the greatest potency, particularly against total aerobes, total anaerobes and streptococci. This approach enables the rapid simultaneous evaluation of acidity mitigation, growth inhibition and specific antimicrobial activity by dentifrices
