Assessment of anti-diabetic activity of a novel hydrazine-thiazole derivative: in vitro and in vivo method
Diabetes mellitus is a chronic disease resulting in oxidative stress that promotes tissue damage. The appearance of this disease is highly related to lifestyle and food of the population, being of great interest to search for a dietary supplement that can also act by reducing oxidative alterations. Based on the broad range of biological activity of thiazole derivatives, this work aimed to evaluate the in vitro antioxidant activity of a novel hydrazine-thiazole derivative and studies in vivo. In in vivo experiments, the liver extracts of healthy and diabetic Wistar rats were used, with analysis to determine the enzymatic activity of SOD, CAT, GPx, and GR, and determination of lipid peroxidation. Finally, in the blood of these animals, biochemical parameters were evaluated. Statistical evidence of changes caused in liver enzymes and liquid peroxidation was not detected; however, these parameters were also not changed between control groups with and without diabetes. On the other hand, concerning biochemical parameters, significant differences were detected in uric acid, alkaline phosphatase, ALT, and urea, indicating a possible antioxidant protective role of such substances in the liver and kidney of diabetic animals that could be acting by means other than that commonly reported in the literature
Tumor necrosis factor α, and agonist and antagonists of cannabinoid receptor type 1 and type 2 alter the immunophenotype of stem cells from human exfoliated deciduous teeth
ABSTRACT Objective To verify the involvement of the endocannabinoid system in the immunomodulatory profile of stem cells from human exfoliated deciduous teeth, in the presence or absence of TNF-α, and agonist and antagonists of CB1 and CB2. Methods Stem cells from human exfoliated deciduous teeth were cultured in the presence or absence of an agonist, anandamide, and two antagonists, AM251 and SR144528, of CB1 and CB2 receptors, with or without TNF-α stimulation. For analysis of immunomodulation, surface molecules linked to immunomodulation, namely human leukocyte antigen-DR isotype (HLA-DR), and programmed death ligands 1 (PD-L1) and 2 (PD-L2) were measured using flow cytometry. Results The inhibition of endocannabinoid receptors together with the proinflammatory effect of TNF-α resulted in increased HLA-DR expression in stem cells from human exfoliated deciduous teeth, as well as, in these cells acquiring an anti-inflammatory profile by enhancing the expression of PD-L1 and PD-L2. Isolamento, caracterização e diferenciação de células-tronco do tecido adiposo de glândula mamária bovina
Animal cloning represents a major technological breakthrough, but.it is still an experimental technique and needs some improvement so it can truly be diffused in the agricultural and social environment. Among these techniques, one that has been highlighted is Cloning by nuclear transfer (NT) because of its use in transgenesis techniques, among others, becoming one viable alternative compared to the conventional methods such as pro-nuclear microinjection. However, the low efficiency of the process is a common problem associated with NT, being small the proportion of viable embryos that are reconstructed using somatic cells as nuclear donors. There are many factors playing roles on this low success rate, one of them being the incomplete reprogramming of donor’s somatic cell nucleus, leading to an abnormal expression of important genes related to embryo development. So, for the cloning to work properly, the donor cell nucleus must be completely reprogrammed after its transfer, ceasing its own gene expression programme. The actual stage of cell differentiation and epigenetic commitment of the donor cell may compromise the process, leading to think of using less committed cells in order to obtain a higher efficiency, such as adult stem cells. In the present work we isolate, characterize and test the differentiation potential of stem cells isolated from the adipose tissue of bovine mammary gland (CTGMB), and their characterization as similar to mesenchymal stem cells make interesting their potential use on cattle cell therapy, or even nuclear transfer cloning, once their application in such technique could help increasing its efficiency because CTGMB are more progenitor cells. Furthermore, since they are not committed to a specific cell type, they may be able to bypass epigenetic issues. The know-how and data presented here are novel and innovative, since in literature there is no record of any research group that has come to reach such a detailed characterization in bovine cells, and such protocols for obtaining CTGMB are patentable.A clonagem de animais representa um grande avanço tecnológico, mas ainda é uma técnica em experimento que necessita de aperfeiçoamento para que possa realmente ser difundida no meio agropecuário e social. Entre essas técnicas, uma que tem merecido destaque é a Clonagem por Transferência Nuclear (NT) pelo seu uso em técnicas de transgênese, entre outros, tornando-se uma alternativa viável frente aos métodos convencionais, como a microinjeção pró-nuclear. Porém, um problema frequentemente associado à NT é a baixa eficiência do processo, sendo pequena a proporção de embriões viáveis que são reconstruídos a partir de células somáticas como doadoras de núcleo. Muitos fatores podem influenciar nessa baixa taxa de sucesso, entre eles a incompleta reprogramação do núcleo da célula somática doadora, podendo levar a uma expressão anormal de importantes genes relacionados ao desenvolvimento do embrião. Isso significa que, para a clonagem funcionar, o núcleo da célula doadora precisa ser totalmente reprogramado após sua transferência, cessando sua programação própria de expressão gênica. O grau de diferenciação celular e comprometimento epigenético da célula doadora pode comprometer esse processo, levando-se a pensar na utilização de células menos comprometidas a fim de obter-se uma maior eficiência, tais como as células-tronco adultas. No presente trabalho isolamos, caracterizamos e testamos o potencial de diferenciação de células-tronco isoladas a partir do tecido adiposo da glândula mamária bovina (CTGMB), e sua caracterização semelhante a células-tronco mesenquimais torna interessante o potencial dessas células para terapia celular em bovinos, ou mesmo para clonagem por transferência nuclear, já que sua aplicação na técnica pode contribuir para aumentar sua eficiência, por se tratarem de células mais progenitoras. Além disso, por não terem comprometimento com um dado tipo celular, talvez possam contornar problemas epigenéticos. O know-how e os dados aqui apresentados são inéditos e inovadores, pois na literatura não há registro de algum grupo de pesquisa que tenha chegado a tal caracterização com riqueza de detalhes em células bovinas, e tais protocolos para obtenção das CTGMB podem ser patenteados.CNPq - Conselho Nacional de Desenvolvimento Científico e Tecnológic
Participação do sistema endocanabinóide na modulação da função imunológica de células-tronco da polpa dentária humana
The endogenous cannabinoid signaling system (eCBS) is composed of
endocannabinoid molecules that interact with G-protein coupled receptors CB1
and CB2, and it is present in many tissues and cell types, including central
nervous system (CNS) cells, the immune system cells and mesenchymal stem
cells (MSC). The presence of such receptors in both immune and stem cells
may be indicative of the cellular signaling pathway through which the two
systems communicate. Lately there has been shown a growth in treatments of
various CNS-related diseases using cannabis derivatives such as cannabidiol,
showing positive results. However, we still do not know the consequences of
treatments involving eCBS in other cell types.Thus, there is a need to
understand such communication, as well as the systemic and related side
effects upon receptors stimulation, since it may influence the activation and
response of human dental pulp stem cells (DPSC). Such knowledge also form
the basis for a better understanding of cannabis medicinal effects, contributing
to clarify its action for example in neurodegenerative diseases and also i the
development of new medicines. Thus, the present study aims at evaluating the
effects of cannabinoid receptor stimulatory and inhibitory agents uppon DPSC
immune and proliferative responses. So, human DPSC were isolated from
dental pulp and characterized to confirm their identity as stem cells. DPSC were
then cultured in the presence or absence of an agonist (anandamide) and
antagonists (AM251 and SR144528) for eCBS receptors, whether or not
stimulated by TNF-α. For immunomodulation analysis, immunosuppressionrelated
surface molecules (HLA-DR, PD-L1, PD-L2) were measured by flow
cytometry. eCBS components mRNA expression was verified by Real time RTPCR
as well as the expression for TGF-β and IL-6 cytokines. The proliferation
rate of DPSCs under study was also evaluated. The characterization of the
isolated cells confirmed their identity as stem cells. Cytometry for HLA-DR, PDL1
and PD-L2 markers indicated that DPSCs express these markers on their
cell surface and that their expression responds differentially to eCBS stimulus
as well as to the presence of TNF-α, which proves the role of such system in
influencing the DPSCs immunomodulatory profile. Gene expression analysis for
eCBS-related enzymes and receptors provided a deep and detailed
understanding of the system's functioning in DPSCs, both in terms of its selfregulation
and its response to the presence of TNF-α. Such results confirmed
that eCBS is a present and functionally active system in DPSCs, directly
involved with immunomodulation. Gene expression analysis for TGF-β and IL-6
cytokines showed that DPSCs express genes for these cytokines even when
cultured without specific stimulation. It also showed that this expression is
profoundly altered as a response to eCBS stimuli and TNF-α stimulation, again
indicating the role of this system in coordinating the cellular response of DPSCs,
especially concerning immunomodulation. Analysis of DPSC proliferation rate
under eCBS and TNF-α stimulation showed that eCBS is not able to
significantly affect DPSC proliferation, but TNF-α does. The combined analysis
of these data indicates that eCBS present in DPSCs not only responds to
immune stimulation, but also alters key immune response molecules, providing
the necessary stimuli for DPSCs to act in the coordination of such environment.O sistema de sinalização de canabinoides endógeno (eCBS) é composto por
moléculas endocanabinóides que interagem com os receptores CB1 e CB2
acoplados à proteína G, estando presente em diversos tecidos e tipos celulares,
entre eles em células do sistema nervoso central (SNC), sistema imune e
células-tronco mesenquimais (MSC). A presença desses receptores, em
específico, tanto em células do sistema imune como em células-tronco pode
ser indício do caminho de sinalização celular pelo qual os dois sistemas se
comunicam. Ultimamente houve crescimento de tratamentos de diversas
doenças relacionadas ao SNC utilizando derivados de canábis, como o
canabidiol, demonstrando resultados positivos. No entanto, não sabemos os
reflexos dos tratamentos que envolvem o eCBS em outros tipos celulares.
Existe assim a necessidade do entendimento dessa comunicação, bem como
dos efeitos sistêmicos e prováveis efeitos colaterais relacionados à estimulação
desses receptores, uma vez que o estímulo dos mesmos pode influenciar a
ativação e resposta de células-tronco de polpa dentária humana (DPSC). Tais
conhecimentos formam ainda a base para uma melhor compreensão do efeito
medicinal da canábis, contribuindo no esclarecimento de sua ação por exemplo
em doenças neurodegenerativas e no desenvolvimento de novos
medicamentos. O presente trabalho objetivou então avaliar os efeitos de
agentes estimulatórios e inibitórios dos receptores canabinóides sobre a
resposta imunológica e proliferativa das DPSC. Para tal, DPSC humanas foram
isoladas da polpa dentária e caracterizadas para confirmação de sua
identidade como células-tronco (CT). As DPSC foram então cultivadas na
presença ou ausência de um agonista (anandamida) e de antagonistas (AM251
e SR144528) dos receptores CB1 e CB2 do eCBS, sob estímulo ou não de
TNF-α. Para análise da imunomodulação, moléculas de superfície ligadas à
imunossupressão (HLA-DR, PD-L1, PD-L2) foram mensuradas por citometria
de fluxo. Paralelamente, a expressão de mRNA relacionado às moléculas do
eCBS foi verificada por Real time RT-PCR, bem como a expressão para as
citocinas TGF-β e IL-6. Ainda, a taxa de proliferação das DPSCs em estudo
também foi avaliada. A caracterização das células isoladas confirmou sua
identidade como CT. A citometria para os marcadores HLA-DR, PD-L1 e PD-L2
indicou que as DPSCs expressam esses marcadores em sua superfície celular
e que sua expressão responde diferencialmente ao estímulo do eCBS, bem
como à presença de TNF-α, o que comprova o papel desse sistema na
influência do perfil imunomodulador atribuído às DPSCs. A análise da
expressão gênica para os componentes do eCBS forneceu uma compreensão
detalhada do funcionamento do sistema em DPSCs em termos de sua
autorregulação, e em termos de sua resposta à presença de TNF-α. Isso
permitiu confirmar que o eCBS é um sistema presente e funcionalmente ativo
em DPSCs, envolvido diretamente com a imunomodulação. A análise da
expressão gênica para as citocinas TGF-β e IL-6 mostrou que as DPSCs
expressam os genes para estas citocinas mesmo quando cultivadas sem
estímulo específico. Mostrou também que essa expressão é profundamente
alterada como resposta aos estímulos do eCBS e ao estímulo por TNF-α,
indicando mais uma vez o papel desse sistema na coordenação da resposta
celular de DPSCs, em especial relativa à imunomodulação. A análise da taxa
de proliferação das DPSCs frente aos estímulos do eCBS e ao estímulo por
TNF-α mostrou que o eCBS não é capaz de afetar significativamente a
proliferação das DPSCs, mas o TNF-α sim. A análise conjunta desses dados
indica que o eCBS presente nas DPSCs não só responde ao estímulo
imunológico, como também altera moléculas chave da resposta imune,
fornecendo os estímulos necessários para que as DPSCs atuem na
coordenação do ambiente inflamatório
O ENSINO DA BIOTECNOLOGIA NA EJA: RELATO DE EXPERIÊNCIA
A biotecnologia é um conjunto de técnicas que envolve diversas áreas e conceitos, permitindo analisar múltiplas questões, sobre alimentação, meio ambiente, agricultura entre outras. Nesse contexto, o presente trabalho foi desenvolvido com alunos da Educação de Jovens e Adultos (EJA), especificamente EJA Médio, com o objetivo de aprimorar o ensino de biotecnologia, além de desenvolver práticas que facilitem a aprendizagem dos conteúdos relacionados à mesma. Para tal, inicialmente analisou-se o livro didático utilizado em sala, que é um dos principais materiais utilizados como fonte de pesquisa pelo professor. Os itens observados no livro foram: ano de publicação; presença do tema biotecnologia enquanto item isolado; número de páginas/capítulos dedicados ao tema; aproximação à realidade do aluno; linguagem acessível e contemporânea. Assim, pode-se se verificar que os autores do livro adotado referenciam a biotecnologia em um capítulo com 14 páginas, sendo inseridos conceitos sobre DNA, transgênicos, bem como uma prática com extração do DNA do morango, preocupando-se em aproximar o conteúdo à realidade do aluno. Posteriormente à análise do livro, foi realizada uma discussão com os alunos a fim de analisar o conhecimento prévio dos mesmos sobre o tema biotecnologia. Estes demonstraram em suas respostas a grande influência das mídias na divulgação sobre biotecnologia. Em seguida foram realizadas três práticas com os alunos: extração do DNA do tomate; ação de enzimas proteases atuando no colágeno existente na gelatina; e indicador ácido-base com extrato de repolho roxo. Durante estas práticas foram discutidos os conceitos e conteúdos que envolvem as mesmas, além de ser demonstrado aos alunos que a biotecnologia está inserida na produção de produtos comuns, como pães e iogurtes, entre outros. Como resultado, percebe-se um grande interesse dos alunos nas práticas realizadas, que funcionam como um ótimo método de ensino e aprendizagem. Foi verificado ainda que a biotecnologia necessita de uma abordagem maior e mais ligada à realidade do aluno durante as aulas para que esta não se torne complexa e distante dos conhecimentos dos mesmos.
 
Isolamento de células-tronco de dentes decíduos exfoliados humanos e sua capacidade para diferenciação osteogênica
As células-tronco de dentes decíduos exfoliados, isoladas como uma subpopulação das células-tronco da polpa dentária, demonstram possuir a capacidade de se diferenciarem in vitro em neurônios, adipócitos, odontoblastos e osteoblastos. Devido a sua facilidade de obtenção, que não envolve um procedimento invasivo, esta linhagem celular é considerada uma grande promessa para a medicina regenerativa. Os objetivos deste trabalho foram estabelecer e caracterizar duas linhagens de células-tronco de dentes decíduos humanos. As linhagens foram isoladas a partir de amostras de crianças saudáveis de 6 anos de idade. Os resultados mostraram, por citometria de fluxo, que as células foram positivas para marcadores de células-tronco mesenquimais (CD105 (SH2), SH3 e SH4) e negativa para a marcação hematopoiética (CD45) e que as linhagens estabelecidaseram constituídas de uma população homogênea de células-tronco. As células obtidas foram avaliadas quanto a sua proliferação e capacidade de diferenciação osteogênica. As linhagens tiveram estatisticamente a mesma taxa de proliferação e foram capazes de diferenciação osteogênica
Toward Personalized Cell Therapies by Using Stem Cells: Seven Relevant Topics for Safety and Success in Stem Cell Therapy
Stem cells, both embryonic and adult, due to the potential for application in tissue regeneration have been the target of interest to the world scientific community. In fact, stem cells can be considered revolutionary in the field of medicine, especially in the treatment of a wide range of human diseases. However, caution is needed in the clinical application of such cells and this is an issue that demands more studies. This paper will discuss some controversial issues of importance for achieving cell therapy safety and success. Particularly, the following aspects of stem cell biology will be presented: methods for stem cells culture, teratogenic or tumorigenic potential, cellular dose, proliferation, senescence, karyotyping, and immunosuppressive activity
Intestinal Microbiota as Modulators of the Immune System and Neuroimmune System: Impact on the Host Health and Homeostasis
Many immune-based intestinal disorders, such as ulcerative colitis and Crohn’s disease, as well as other illnesses, may have the intestines as an initial cause or aggravator in the development of diseases, even apparently not correlating directly to the intestine. Diabetes, obesity, multiple sclerosis, depression, and anxiety are examples of other illnesses discussed in the literature. In parallel, importance of the gut microbiota in intestinal homeostasis and immunologic conflict between tolerance towards commensal microorganisms and combat of pathogens is well known. Recent researches show that the immune system, when altered by the gut microbiota, influences the state in which these diseases are presented in the patient directly and indirectly. At the present moment, a considerable number of investigations about this subject have been performed and published. However, due to difficulties on correlating information, several speculations and hypotheses are generated. Thus, the present review aims at bringing together how these interactions work—gut microbiota, immune system, and their influence in the neuroimmune system
Satellite Cells: Regenerative Mechanisms and Applicability in Muscular Dystrophy
The satellite cells are long regarded as heterogeneous cell population, which is intimately linked to the processes of muscular recovery. The heterogeneous cell population may be classified by specific markers. In spite of the significant amount of variation amongst the satellite cell populations, it seems that their activity is tightly bound to the paired box 7 transcription factor expression, which is, therefore, used as a canonical marker for these cells. Muscular dystrophic diseases, such as Duchenne muscular dystrophy, elicit severe tissue injuries leading those patients to display a very specific pattern of muscular recovery abnormalities. There have been works on the application of precursors cells as a therapeutic alternative for Duchenne muscular dystrophy and initial attempts have proven the cells inefficient; however later endeavours have proposed solutions for the experiments improving significantly the results. The presence of a range of satellite cells populations indicates the existence of specific cells with enhanced capability of muscular recovery in afflicted muscles
