    Myofibroblasts proliferation of idiopathic and collagen vascular disorders associated nonspecific interstitial pneumonia.

    Nonspecific interstitial pneumonia (NSIP) has been recognized as a separate histological classification of interstitial lung disease. Similar features are found not only in idiopathic NSIP, but also in NSIP associated with collagen vascular disorder (CVD-NSIP). We examined the clinical symptoms, laboratory findings, and prognosis of 13 cases of idiopathic NSIP and 11 cases of CVD-NSIP. Immunohistochemical staining was performed using the streptavidin/biotin/peroxidase method with anti-alpha-smooth muscle actin antibody. No differences in the distribution of clinical features, laboratory findings, and prognosis were observed between idiopathic NSIP and CVD-NSIP. In immunohistochemical staining of the fibrosing areas, myofibroblasts were observed in 7 of 13 idiopathic NSIP cases, but in 10 of 11 CVD-NSIP cases. With regards to intra-alveolar organization, myofibroblasts were observed in all 10 CVD-NSIP cases, but they were observed in only 2 of 9 idiopathic NSIP cases. We found a significantly higher myofibroblast proliferation in the intra-alveolar organization of CVD-NSIP compared to idiopathic NSIP. Clinically, idiopathic NSIP and CVD-NSIP are similar, but are pathologically different.</p

While specific therapeutics and vaccines are being developed, a broad‐spectrum therapeutic agent for RNA viruses would be beneficial for targeting newly emerging and mutated RNA viruses. In this study, we conducted a screen of repurposed drugs using Sendai virus (an RNA virus of the family Paramyxoviridae), with human‐induced pluripotent stem cells (iPSCs) to explore existing drugs that may present anti‐RNA viral activity. Selected hit compounds were evaluated for their efficacy against two important human pathogens: Ebola virus (EBOV) using Huh7 cells and severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) using Vero E6 cells. Selective estrogen receptor modulators (SERMs), including raloxifene, exhibited antiviral activities against EBOV and SARS‐CoV‐2. Pioglitazone, a PPARγ agonist, also exhibited antiviral activities against SARS‐CoV‐2, and both raloxifene and pioglitazone presented a synergistic antiviral effect. Finally, we demonstrated that SERMs blocked entry steps of SARS‐CoV‐2 into host cells. These findings suggest that the identified FDA‐approved drugs can modulate host cell susceptibility against RNA viruses

    Type IIA orientifolds and orbifolds on non-factorizable tori

    We investigate Type II orientifolds on non-factorizable torus with and without its oribifolding. We explicitly calculate the Ramond-Ramond tadpole from string one-loop amplitudes, and confirm that the consistent number of orientifold planes is directly derived from the Lefschetz fixed point theorem. We furthermore classify orientifolds on non-factorizable Z_N x Z_M orbifolds, and construct new supersymmetric Type IIA orientifold models on them.Comment: 42 pages, 3 figures, v2: typos corrected, references added, version to appear in Nucl.     Role of protease activated receptor-2 in lymph node metastasis of uterine cervical cancers

    <p>Abstract</p> <p>Background</p> <p>Protease activated receptor-2 (PAR-2) has been implicated in cellular proliferation, invasion and metastasis in various tumors. Lymph node metastasis is an important patient prognostic factor for uterine cervical cancers. This prompted us to study the role of PAR-2 in lymph node metastasis of uterine cervical cancers.</p> <p>Methods</p> <p>Thirty patients underwent surgery for uterine cervical cancers. PAR-2 histoscores and mRNA levels were determined by immunohistochemistry and real-time reverse transcription-polymerase chain reaction, respectively. Patient prognosis was analyzed with a 48-month survival rate.</p> <p>Results</p> <p>PAR-2 histoscores and mRNA levels significantly (<it>P </it>< 0.05) increased in 12 of 30 metastatic lymph node lesions from the corresponding primary tumor. The 48-month survival rate of the 12 patients with increased PAR-2 levels in metastatic lymph nodes was 42%, while the rate of the other 18 patients with no change in PAR-2 levels was 82%, regardless of histopathological type.</p> <p>Conclusion</p> <p>PAR-2 might work on lymph node metastasis of uterine cervical cancers, and is considered to be a novel prognostic indicator for uterine cervical cancers.</p

    Expression of centromere protein F (CENP-F) associated with higher FDG uptake on PET/CT, detected by cDNA microarray, predicts high-risk patients with primary breast cancer

    <p>Abstract</p> <p>Background</p> <p>Higher standardized uptake value (SUV) detected by 18F-fluorodeoxyglucose positron emission tomography/computed tomography (FDG PET/CT) correlates with proliferation of primary breast cancer. The purpose of this study is to identify specific molecules upregulated in primary breast cancers with a high SUV and to examine their clinical significance.</p> <p>Methods</p> <p>We compared mRNA expression profiles between 14 tumors with low SUVs and 24 tumors with high SUVs by cDNA microarray. We identified centromere protein F (CENP-F) and CDC6 were upregulated in tumors with high SUVs. RT-PCR and immunohistochemical analyses were performed to validate these data. Clinical implication of CENP-F and CDC6 was examined for 253 archival breast cancers by the tissue microarray.</p> <p>Results</p> <p>The relative ratios of CENP-F and CDC6 expression levels to β-actin were confirmed to be significantly higher in high SUV tumors than in low SUV tumors (<it>p </it>= 0.027 and 0.025, respectively) by RT-PCR. In immunohistochemical analysis of 47 node-negative tumors, the CENP-F expression was significantly higher in the high SUV tumors (74%) than the low SUV tumors (45%) (<it>p </it>= 0.04), but membranous and cytoplasmic CDC6 expressions did not significantly differ between both groups (<it>p </it>= 0.9 each). By the tissue microarray, CENP-F (HR = 2.94) as well as tumor size (HR = 4.49), nodal positivity (HR = 4.1), and Ki67 (HR = 2.05) showed independent impact on the patients' prognosis.</p> <p>Conclusion</p> <p>High CENP-F expression, correlated with high SUV, was the prognostic indicators of primary breast cancer. Tumoral SUV levels may serve as a pretherapeutic indicator of aggressiveness of breast cancer.</p

    Development of observation system for tsunami and crustal deformation

    The 2011 off the Pacific coast of Tohoku earthquake (M9) occurred at 11 March in last year and huge tsunami brought severe damage around the Tohoku area. The huge earthquake brought large crustal movement. The movements were 24m on the forearc and 50m near the trench (Sato et al., 2011; Fujiwara et al., 2012). The observation, however, was measured after the occurrence and not in real-time. Therefore, we are developing new real-time observation system for tsunami and crustal movement developed by Japan Agency for Marine-Earth Science and Technology (JAMSTEC) and Tohoku University. Our system is composed of seafloor stations and a buoy on sea surface, and these data are sent via satellites in real-time. We selected the m-TRITON buoy developed by JAMSTEC as the platform on sea surface. Because our target is the future Tonankai and Nankai earthquake, the buoy has to adapt slack mooring to stand strong sea current like the Kuroshio with speed of over 5 knots around Japan. We will attach recording systems for tsunami and geodetic movement and the transfer system to send data to satellites in real-time on the buoy. The power supply for them is covered with lithium batteries and the solar power generation. The seafloor station is composed of an equipment to transfer pressure data for tsunami detection and some transponders to detect geodetic movement. Tsunami data recorded by the seafloor pressure sensor is sent acoustically with interval of 15 minutes normally, but the interval changes to 15 seconds in tsunami occurrence. The geodetic movement data is collected with interval of one week. As positioning to monitor geodetic movement with high accuracy less than 10 cm, we adapt the Precise Point Positioning (PPP) technical scheme developed by Japan Aerospace Exploration Agency and have a plan to construct total system to four years.Abstract AOGS/AGU (WPGM) Joint Assembly at Singapore, 13-17 August, 201
