    Caracterización de H3K4me3 y H3K27me3 en una deficiencia primaria de CoQ10

    Motivación: El coenzima Q10 (CoQ10) es una molécula esencial para la célula, destacando su papel en la cadena respiratoria. Su deficiencia da lugar a un síndrome heterogéneo, cuya herencia es autosómica recesiva, que provoca la muerte prematura del individuo afectado. El grupo de Fernández-Ayala mantiene que las células deficientes podrían sufrir una adaptación en etapas tempranas del desarrollo fetal, consistente en alteraciones de las modificaciones epigenéticas que justificarían el transcriptoma de supervivencia encontrado (Fernández-Ayala et al.). Además, han estudiado los efectos que la suplementación con CoQ10, que es la terapia actual, puede tener sobre ambos. Nuestro objetivo es analizar si las modificaciones H3K4me3 y H3K27me3 pueden estar regulando la expresión y si la suplementación con CoQ10 tiene algún efecto sobre estas. Métodos: Se ha realizado un ChIP-Seq de fibroblastos de un paciente con una deficiencia primaria tratados durante una semana y sin tratar, y de un individuo sano de similar edad. Mediante el uso de diversas herramientas informáticas (Nielsen et al. y Zhang et al.) se ha obtenido allí donde la presencia de H3K4me3 y H3K27me3 destaca. Se ha analizado su distribución genómica y el GO de los genes posiblemente afectados. Finalmente nos hemos centrado en la zona de los promotores, donde se halla el promotor en sí y algunos elementos reguladores, para ver si se justifica el transcriptoma de supervivencia. Resultados: La adaptación a la deficiencia de CoQ10 parece alterar mucho más los patrones de H3K4me3 en las células, que los de H3K27me3. Estas afectarían sobre todo a genes implicados en procesos metabólicos, celulares, regulación biológica o de un proceso biológico. En los promotores estas marcas tienden a desaparecer en la deficiencia, aunque también pueden aparecer nuevos promotores marcados. Resultados preliminares demuestran que estos patrones justifican los datos globales del transcriptoma, así como la restauración parcial tras la suplementación. Además, CoQ10 parece tener otros efectos: puede incrementar las diferencias entre control y paciente y promover la eliminación de modificaciones en promotores de genes no relacionados con la enfermedad o la adición de estas en promotores anteriormente no afectados . Conclusiones: La deficiencia parece alterar el patrón de H3K4me3 en la célula, eliminándolo principalmente, sobre todo en los promotores. La suplementación sólo consigue una restauración parcial

    Dynamic changes in the epigenomic landscape regulate human organogenesis and link to developmental disorders

    How the genome activates or silences transcriptional programmes governs organ formation. Little is known in human embryos undermining our ability to benchmark the fidelity of stem cell differentiation or cell programming, or interpret the pathogenicity of noncoding variation. Here, we study histone modifications across thirteen tissues during human organogenesis. We integrate the data with transcription to build an overview of how the human genome differentially regulates alternative organ fates including by repression. Promoters from nearly 20,000 genes partition into discrete states. Key developmental gene sets are actively repressed outside of the appropriate organ without obvious bivalency. Candidate enhancers, functional in zebrafish, allow imputation of tissue-specific and shared patterns of transcription factor binding. Overlaying more than 700 noncoding mutations from patients with developmental disorders allows correlation to unanticipated target genes. Taken together, the data provide a comprehensive genomic framework for investigating normal and abnormal human development

    Molecular Structure, Biosynthesis, and Distribution of Coenzyme Q

    Coenzyme Q is a very old molecule in evolutionary terms that has accumulated numerous functions in the cellular metabolism beyond its primordial function, the electron transport. In all organisms, coenzyme Q maintains a highly conserved structure allowing a localization inside cell membranes in a hydrophobic environment thanks to having an isoprenoid tail, and at the same time allows the polar ring benzene to interact with acceptors and electron donors. Coenzyme Q deficiency constitutes a group of mitochondrial diseases. Affected patients suffer mainly a decrease in energy production that induces dysfunctions in most organs and body systems. Current therapeutic alternatives are based on increasing coenzyme Q levels either through induction of endogenous mechanisms or exogenous supplementation. This chapter includes both aspects, the mechanisms associated with the coenzyme Q supplementation and the regulatory mechanisms of coenzyme Q biosynthesis. In terms of synthesis, the structure of coenzyme Q is complicated since it requires the participation of two well-differentiated pathways that must be carefully regulated. The synthesis is carried out through the participation of a multienzyme complex located in the inner mitochondrial membrane and controlled by different levels of regulation that at this time are not well-known
